Abstract
멜라닌색소는 동물이나 식물에 흔히 존재하며 이들의 생합성은 타이로시나제 라는 효소의 작용으로 아미노산인 타이로신이 도파로 만들어지고 도파는 도파퀴논으로 변하면서 이들이 서로 중합하여 만들어진다. 그러나 엘도파라는 물질은 여러가지 pH 농도에서 공기중의 산소와 반응하여 스스로 산화되어 도파퀴논으로 되면서 중합하여 생체 내에서 만들어지는 멜라닌 색소와 유사한 중합화합물이 되고 이들은 액상의 콜로이드 형태로 존재하게 된다. 이러한 콜로이드의 농도는 pH를 조정하여 다양하게 멜라닌량을 조절할수 있고 또한 고농도로 만들어진 것은 적당량 희석하여 색상을 조절할 수 있다. 이에 본 발명자는 이들 도파 멜라닌 콜로이드액을 이용하여 피부에 착색을 유도 할 수 있다고 생각되어 백반증 및 저색소반에서의 도파멜라닌을 피부에 주입하여 치료에 이용하는 것에 관하여 특허출원 심사를 받고자한다. Melanin pigments are commonly present in animals and plants, and their biosynthesis is caused by the action of an enzyme called tyrosinase to make amino acids tyrosine into dopas and dopas into dopaquinones, which are polymerized together. However, eldopara is reacted with oxygen in the air at various pH concentrations to oxidize itself into dopaquinone and polymerize to form melanin pigments that are produced in vivo, and they exist in the form of liquid colloids. The concentration of these colloids can be adjusted in various ways to adjust the melanin amount, and also made of high concentration can be adjusted by diluting the appropriate amount. Accordingly, the present inventors believe that the dopa melanin colloidal fluid can be used to induce pigmentation on the skin. Thus, the present inventors intend to undergo a patent application examination regarding the use of dopa melanin in vitiligo and low pigmented plaques for skin treatment.

Description
본 도파 멜라닌색소는 의료용 및 화장품류의 기술 분야로 분류 할수 있고 이와 가장 유사한 종래의 기술은 문신과 같다. 현재 문신에 사용되는 잉크는 탄소, 타이다니움 옥사이드, 아이언 옥사드등이 사용되고 있으나 이들은 피부에 있는 멜라닌 색소가 아닌 관계로 주위 피부색과 조화를 이루기에 매우 어려운 점이 있으며 이들 색소가 주위 피부로 번지는 경우가 흔하여 백반증이나 저색소증에서 사용하기에는 문제가 있어 왔다. 이에 본 발명자는 피부색과 가장 유사하며 피부에 이식후에도 정상 피부와 가장 유사한 도파멜라닌을 발명하였다.The dopa melanin pigment can be classified into the technical field of medical and cosmetics, and the most similar conventional technology is like a tattoo. 피부색은 주로 피부중 표피에 있는 멜라닌 색소에 따라 크게 흑인, 황인, 백인으로 나눌수 있다. 멜라닌 색소의 생합성은 타이로신으로부터 타이로시나제의 효소에 의해 만들어지나 엘도파(L-DOPA, L-dioxyphenylalanine)로부터 자연적인 산화 로 이루어 지기도한다. 이에 본 발명자는 엘도파를 여러가지 알카리 용액에 노출 시키면 다양한 농도의 도파멜라닌 색소를 만들수 있다는 사실과 이들 색소를 피부에 주입하면 자연적인 피부 색조를 얻을 수 있음을 발견하였다. 이들 색소를 백반증 및 저색소성 반에 주입하면 주위의 정상적인 피부색과 유사한 색조의 회복을 가져올 수 있음을 확인하였다. 도파멜라닌 색소를 만드는 방법은 엘도파를 수산화나트륨 0.001 M 에서 0.04M 의 여러가지 농도의 용액에 녹이고 이들을 밀폐용기에 넣은 후 산소와 약 5일간 노출시키면, 이들은 수산화 나트륨의 농도에 따라 다양한 농도의 포화 도파멜라닌 콜로이드가 용액중에 생성되고 이들 액의 알카리 pH는 산화과정을 거치면서 마지막으로는 중성 pH를 나타낸다는 것을 발견하였다. 그리고 불포화 엘도파는 2000 rpm에서 원심 분리하면 침전물로서 분리된다. 이중 상층액을 취하여 백반증의 환부 혹은 저색소반의 피부중 상부 진피에 주입하면 정상과 유사한 피부색을 회복 시킬 수 있다.
Skin color can be largely divided into black, yellow, and white depending on the melanin pigment in the epidermis of the skin. The biosynthesis of melanin pigments is made by tyrosine from tyrosinase enzymes, but also by natural oxidation from L-DOPA (L-dioxyphenylalanine). Accordingly, the present inventors have found that when the EL dopa is exposed to various alkaline solutions, dopamelanin pigments of various concentrations can be made, and natural pigments can be obtained by injecting these pigments into the skin. Injecting these pigments into vitiligo and hypopigmented plaques has been shown to lead to recovery of the tone similar to the normal skin color of the surroundings. The method of making dopamelanin pigment is to dissolve eldopa in a solution of various concentrations from 0.001 M to 0.04 M sodium hydroxide, put them in an airtight container and expose them to oxygen for about 5 days, and they are saturated in various concentrations depending on the concentration of sodium hydroxide. It was found that melanin colloids were formed in the solution and the alkali pH of these solutions finally showed neutral pH as they were oxidized. And unsaturated eldopa is separated as a precipitate by centrifugation at 2000 rpm. Taking the supernatant and injecting it into the upper dermis of the affected or visceral plaque of the vitiligo can restore normal skin color.
