Claims (10)
- 두충잎 중의 퀘르세틴-3-O-β-D-자일로피라노실(1→2)-β-D-글루코피라노시드의 함량을 측정하는 것을 특징으로 하는, 두충(Eucommia ulmoidesOlv.) 잎의 품질 검정 방법.
- 제1항에 있어서,(1) 건조된 두충잎을 유기용매로 추출한 후, 여과 및 건조하여 건조 추출물을 얻는 단계;(2) 1 단계에서 얻은 두충잎의 건조 추출물을 물에 현탁시키고, 이 현탁액에 비극성용매를 가하여 분획한 후 비극성용매 가용부를 제거하고 남은 물 분획물을 수득하는 단계;(3) 상기 2 단계에서 얻어진 물 분획물을 0 내지 100 % 농도 구배의 저급알콜을 함유하는 물을 전개용매로 하여 다공성 비이온성 흡착 수지 컬럼 크로마토그래피법으로 정제한 후, 겔 여과 크로마토그래피하여 퀘르세틴-3-O-β-D-자일로피라노실(1→2)-β-D-글루코피라노시드를 분리하는 단계; 및(4) 상기 3 단계에서 분리된 퀘르세틴-3-O-β-D-자일로피라노실(1→2)-β-D-글루코피라노시드의 함량을 측정하는 단계를 포함하는 것을 특징으로 하는 방법.
- 제2항에 있어서,1 단계의 추출방법이 초음파 추출법 또는 환류 냉각 추출법인 방법.
- 제2항에 있어서,1 단계의 유기용매가 메탄올 또는 에탄올인 방법.
- 제2항에 있어서,1 단계의 추출온도는 20 내지 70℃인 방법.
- 제2항에 있어서.1 단계의 추출시간은 1시간 내지 24시간인 방법.
- 제2항에 있어서,2 단계에서 분획에 사용하는 비극성 용매가 에테르, 에틸아세테이트, 시클로헥산, 헥산, 벤젠, 클로로포름 및 아세톤으로 이루어진 군으로부터 선택되는 방법.
- 제2항에 있어서,3 단계에서 다공성 비이온성 흡착 수지는 앰버라이트(Amberlite) XAD-2, 앰버라이트 XAD-4, 다이아이온(Diaion) HP-20 및 이들의 조합으로 이루어진 그룹으로부터 선택되는 방법.
- 제2항에 있어서,3 단계에서 겔 여과 크로마토그래피의 수지는 세파덱스형 수지인 방법.
