Claims (5)
- 글리세르알데히드-3-포스페이트 데히드로게나제(GAPDH) 프로모터(GAP), 사카로미세스 세레비시아(Saccharomyces cerevisiae)의 α-인자 리더 서열, 인간 인터루킨-2 유전자 및 번역종결코드를 포함하는 재조합 벡터 pGAPZαA/rhIL-2.
- pGAPZαA/rhIL-2로 형질전환된 피키아 파스토리스.
- 제 2항에 있어서, 상기 피키아 파스토리스는 피키아 파스토리스/rhIL-2(KCCM10322)인 것인 피키아 파스토리스.
- 피키아 파스토리스/rhIL-2(KCCM10322)에서 발현된 재조합 인터루킨-2 단백질.
- (1) pGAPZαA 벡터에 인터루킨-2 유전자를 삽입하여 pGAPZαA/rhIL-1를 제조하고;(2) pGAPZαA/rhIL-1를 피키아 파스토리스 X33에 형질전환하고;(3) 형질전환된 피키아 파스토리스 X33은 제오신을 포함하는 배지에 배양하여 pGAPZαA/rhIL-1가 다수카피로 삽입된 피키아 파스토리스/rhIL-1를 선별하고;(4) 피키아 파스토리스/rhIL-1를 연속배양하여 인터루킨-2를 세포외로 발현시키고; 및(5) 발현된 재조합 인터루킨-2를 정제하여 수득하는 것을 포함하는 재조합 인터루킨-2의 제조방법.
