1. 실험 목적 1. Purpose of the experiment

본 실시예에서는 팥순 및 스피아민트를 다양한 배합비로 혼합하여 복합물을 제조한 후, 이 복합물의 피부 보습효과 및 피부 주름 개선 효과를 확인하고자 하였다. In this example, a composite was prepared by mixing red bean sprouts and spearmint in various mixing ratios, and the skin moisturizing effect and skin wrinkle improvement effect of the composite were confirmed.

2. 실험방법2. Experimental method

(1) 추출물 제조(1) Preparation of extracts

1) 팥순 추출물 : 분말화한 건조물에 주정 (40% 에탄올 함량) 20 배수를 넣고 60℃에서 6시간 추출하였다. Filter 여과 후 50℃에서 회전감압농축기로 농축하고 4일 동안 동결건조하여 팥순 추출물을 취득하였다.1) Red bean sprout extract: Add 20 times the amount of alcohol (40% ethanol content) to the powdered dried material and extract at 60℃ for 6 hours. RNA 추출을 위한 FavorPrep™ Blood/Cultured Cell Total RNA Mini Kit는 Favorgen (Taipei, Taiwan)에서 cDNA 합성을 위한 AccuPower® CycleScript™ RT PreMix (dN6)는 Bioneer (Daejeon, Korea)에서 실시간 PCR을 위한 PowerUp™ SYBR™ Green Master Mix for qPCR는 Applied biosystems (Waltham, MA, USA)에서 구입을 하였다.DMEM, IMDM, and FBS were purchased from Gibco (Grand island, NY, USA). FavorPrep™ Blood/Cultured Cell Total RNA Mini Kit for RNA extraction was purchased from Favorgen (Taipei, Taiwan). AccuPower® CycleScript™ RT PreMix (dN6) for cDNA synthesis was purchased from Bioneer (Daejeon, Korea). PowerUp™ SYBR™ Green Master Mix for qPCR for real-time PCR was purchased from Applied biosystems (Waltham, MA, USA).

(3) 세포 배양 및 자외선 조사(3) Cell culture and UV irradiation

인간 피부 각질 세포인 HaCaT 세포는 1% 페니실린-스트렙토마이신과 10% FBS이 함유된 DMEM 배지를 사용하고, 인간 피부 섬유아 세포인 CCD-986sk 세포는 1% 페니실린-스트렙토마이신과 10% FBS이 함유된 IMDM 37℃, 5% CO2 인큐베이터에서 배양하였으며 3일에 한 번씩 계대 배양을 시행하였다. HaCaT cells, which are human skin keratinocytes, were cultured in DMEM medium containing 1% penicillin-streptomycin and 10% FBS, and CCD-986sk cells, which are human skin fibroblasts, were cultured in IMDM containing 1% penicillin-streptomycin and 10% FBS at 37°C in a 5% CO2 incubator, and passaged once every 3 days.

UVB 조사는 세포를 5x105 cell/well 농도로 6 웰 플레이트에 분주하고 24시간 배양하고, 무혈청 배지로 교환한 후 24시간 cell starvation 한 뒤 자외선 조사기 BLX-312(Vilber Lourmat, Collegien, France)를 이용하였다. PBS 2회 세척한 뒤 1 mL의 PBS를 분주한 후 UVB를 40 mJ/cm2를 조사하였다. UVB irradiation was performed by seeding cells at a density of 5x105 cells/well in 6-well plates, culturing them for 24 hours, replacing the medium with serum-free medium, and starving the cells for 24 hours using a UV irradiator BLX-312 (Vilber Lourmat, Collegien, France). After washing twice with PBS, 1 mL of PBS was dispensed, and then UVB was irradiated at 40 mJ/ cm2 .

(4) 유전자 발현 평가(4) Gene expression evaluation

유전자 발현 평가를 위해 real-time PCR을 이용하였다. HaCaT 및 CCD-986sk 세포를 5x105 cell/well 농도로 6 웰 플레이트에 분주하고 24시간 배양하고, 무혈청 배지로 교환한 후, 24시간 cell starvation 하였다. 무혈청 배지에 희석한 단일물 또는 복합물 50 μg/mL을 처리하고 24시간 후에 세포를 수집하였다. 13,000 rpm, 3분 동안 원심 분리하여 상등액을 제거하고 FavorPrep™ Blood/Cultured Cell Total RNA Mini Kit를 사용하여 RNA 추출을 하였으며 AccuPower® CycleScript™ RT PreMix (dN6)를 이용하여 1000 ng 농도로 cDNA를 합성했다. 합성된 cDNA는 PowerUp™ SYBR™ Green Master Mix for qPCR을 사용하여 real-time PCR을 수행했다. 이때, housekeeping 유전자인 glyceraldehyde-3-phosphate dehydrogenase (GAPDH) 유전자를 포함하여 internal control로 사용하였으며 유전자 서열은 다음과 같았다. At this time, the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene, a housekeeping gene, was used as an internal control, and the gene sequence was as follows.

HAS1, HAS2, HAS3 유전자는 피부 보습능에 대한 마커 유전자인데, 인간 피부 각질세포에 팥순 및 스피아민트 복합물을 처리할 때 0.9:1.1, 1:1, 1.1:0.9 (팥순:스피아민트) 비율의 복합물이, 대조군 대비, HAS1, HAS2, HAS3 유전자 발현 유의적으로 증가했다. (*p<0.05, **p<0.01, ***p<0.001 vs. CON) 0.9:1.1, 1:1, 1.1:0.9 (팥순:스피아민트)의 복합물 간의 유의적 차이는 나타나지 않았다 (N.S. : Not significant). HAS1, HAS2, and HAS3 genes are marker genes for skin moisturizing ability. MMP1, MMP3, MMP9 유전자는 피부 주름 개선에 관한 마커 유전자인데, 인간 피부 섬유아세포에 팥순 및 스피아민트 복합물을 처리할 때, UV 군 대비 시료 처리 시 MMP1, MMP3, MMP9 유전자 발현이 유의적으로 감소하였다. 이때, 0.9:1.1, 1:1, 1.1:0.9 (팥순:스피아민트) 비율 복합물 처리 시 유전자 발현이 가장 많이 감소됐다. (#p<0.05, ##p<0.01 vs. UV, *p<0.05, **p<0.01, ***p<0.001 vs. CON) 0.9:1.1, 1:1, 1.1:0.9 (팥순:스피아민트)의 복합물 간의 유의적 차이는 나타나지 않았다 (N.S. : Not significant).
