The present invention relates to a composition for moisturizing the skin or improving wrinkles on the skin, and more specifically, to a food or cosmetic composition for moisturizing the skin or improving wrinkles on the skin containing an extract of red bean sprouts and an extract of spearmint.

The skin is the part that primarily protects the body from the external environment, and performs body temperature control, sensory functions, and immune functions. In addition, the skin is responsible for the aesthetic aspect, and clean and healthy skin gives a sense of aesthetic beauty, so interest in the skin is increasing. In order to maintain elastic, blemish-free, clear skin, skin moisturizing management is important, but if the skin lacks moisture, cell regeneration ability decreases, elasticity decreases, and wrinkles appear, leading to aging symptoms.

Recently, environmental pollution, global warming, and natural disasters caused by these have emerged as serious problems. The countless pollutants generated by this environmental pollution dry out the skin, damage the skin barrier, and accelerate skin aging.

It is known that the skin aging phenomenon caused by the external environment is mainly caused by external ultraviolet rays penetrating the epidermis and deep into the dermis layer, damaging collagen and elastin in the dermis, causing wrinkles on the skin, and excessively producing harmful active oxygen species, which accelerates skin aging.

In addition, it is known that external pollutants such as fine dust cause various chemical irritations to keratinocytes and lipid membranes, resulting in inflammation, dry skin, destruction of the skin barrier, and wrinkles.

Health functional foods or cosmetics using various materials are being researched and developed for skin wrinkle improvement, moisturizing, etc., and as the aversion to consuming synthetic compounds has spread recently, consumers' interest in materials made from natural substances is increasing.

Korean Patent Publication No. 1020170101876 (publication date 2017.09.06) describes a method for producing a whitening and wrinkle-improving substance containing an azuki bean skin extract, which contains an azuki bean skin extract as an effective ingredient, obtained by adding ethanol to dried azuki bean skin, filtering, concentrating under reduced pressure, and freeze-drying the extracted liquid. Korean Patent Publication No. 1020210060009 (publication date 2021.05.26) describes a whitening wash-off pack composition containing spearmint essential oil as one of several ingredients.

The present invention aims to develop and provide a food or cosmetic composition that is effective in moisturizing the skin and improving wrinkles by utilizing natural materials.

The present invention provides a skin moisturizing food composition characterized by containing a complex of red bean sprout extract and spearmint extract as an effective ingredient.

In the present invention, a compound was prepared by mixing the above red bean sprouts and spearmint, and then the skin efficacy was confirmed. As shown in the following experiment, it was confirmed that the compound had excellent efficacy for moisturizing the skin and improving wrinkles.

According to the experiments of the present invention below, it was confirmed that the complex of the present invention reduces the expression of MMP1, MMP3, and MMP9 genes in skin fibroblasts, which are markers for improving skin wrinkles.

Meanwhile, the red bean sprout extract used in the present invention is preferably an extract obtained by adding ethanol as an extraction solvent to red bean sprouts and extracting them. At this time, it is preferably preferable to use ethanol having an ethanol content of 35 to 45%. For example, the red bean sprout extract used in the present invention may be manufactured by adding 15 to 25 times the amount of ethanol having an ethanol content of 35 to 45% to powdered red bean sprout dried matter, extracting at a temperature of 55 to 65°C for 4 to 8 hours to obtain an extract, filtering and concentrating the extract, and then freeze-drying the extract.

Meanwhile, the spearmint extract used in the present invention is preferably an extract obtained by adding ethanol as an extraction solvent to spearmint leaves and extracting the same. At this time, it is preferably preferable to use ethanol having an ethanol content of 45 to 55%. For example, the spearmint extract used in the present invention may be manufactured by adding 5 to 15 times the ethanol content of 45 to 55% to powdered dried spearmint leaves, extracting at 55 to 65°C for 4 to 8 hours to obtain an extract, filtering and concentrating the extract, and then freeze-drying the extract.

Meanwhile, in the present invention, a complex was prepared by mixing red bean sprout extract and spearmint extract in various ratios of 1:4, 0.9:1.1, 1:1, 1.1:0.9, and 4:1 (w/w), and the skin moisturizing ability and skin wrinkle improvement ability of the complex were evaluated. It was confirmed that relatively excellent skin moisturizing ability and skin wrinkle improvement ability were achieved at a mixing ratio of preferably 0.9 to 1.1 (red bean sprout extract): 1.1 to 0.9 (sprout mint extract).

Meanwhile, in the food composition of the present invention, the complex of the present invention is preferably contained in an amount of 0.01 to 30.0 wt% based on the total weight of the food composition.

Meanwhile, the present invention can be manufactured into various foods by adding various excipients, and examples thereof include meat, grains, caffeinated beverages, general beverages, chocolate, bread, snacks, confectionery, pizza, jelly, noodles, gum, ice cream, alcoholic beverages, alcohol, vitamin complexes, and other health supplements. In the cosmetic composition of the present invention, the cosmetic composition can be manufactured in any formulation commonly manufactured in the art, and for example, can be formulated as a base cosmetic formulation selected from the group consisting of a solution, a suspension, an emulsion, a paste, a toner, a gel, a water-soluble liquid, a cream, an essence, a surfactant-containing cleanser, an oil, an oil-in-water (O/W) type and a water-in-oil (W/O) type; a formulation for a mask pack; a formulation for a body wash; a peeling gel; an oil-in-water or water-in-oil type makeup base, etc., but is not necessarily limited thereto.

In the cosmetic composition of the present invention, the cosmetic composition can be used in combination with other cosmetic compositions other than those of the present invention. In addition, the cosmetic composition according to the present invention can be used according to a conventional method of use, and the number of times it is used can be varied according to the skin condition or preference of the user.

Hereinafter, the contents of the present invention will be described in more detail through the following examples and experimental examples. However, the scope of the rights of the present invention is not limited to the following examples and experimental examples, but includes modifications of technical ideas equivalent thereto.

[Example 1: Confirmation of skin moisturizing effect and skin wrinkle improvement effect of red bean sprout and spearmint complex]

1. Purpose of the experiment

In this example, a composite was prepared by mixing red bean sprouts and spearmint in various mixing ratios, and the skin moisturizing effect and skin wrinkle improvement effect of the composite were confirmed.

2. Experimental method

(1) Preparation of extracts

1) Red bean sprout extract: Add 20 times the amount of alcohol (40% ethanol content) to the powdered dried material and extract at 60℃ for 6 hours. After filtration, the extract was concentrated in a rotary vacuum evaporator at 50℃ and freeze-dried for 4 days to obtain the red bean sprout extract.

2) Spearmint extract: Add 10 times the amount of alcohol (50% ethanol content) to the powdered dried material and extract at 60℃ for 6 hours. After filtration, the product was concentrated in a rotary vacuum evaporator at 50℃ and freeze-dried for 4 days to obtain the spearmint extract.

3) Manufacturing of composites:

The red bean sprout and spearmint extracts prepared above were mixed in ratios of 1:4, 0.9:1.1, 1:1, 1.1:0.9, and 4:1 (w/w) to prepare a composite.

(2) Reagent

DMEM, IMDM, and FBS were purchased from Gibco (Grand island, NY, USA). FavorPrep™ Blood/Cultured Cell Total RNA Mini Kit for RNA extraction was purchased from Favorgen (Taipei, Taiwan). AccuPower® CycleScript™ RT PreMix (dN6) for cDNA synthesis was purchased from Bioneer (Daejeon, Korea). PowerUp™ SYBR™ Green Master Mix for qPCR for real-time PCR was purchased from Applied biosystems (Waltham, MA, USA).

(3) Cell culture and UV irradiation

HaCaT cells, which are human skin keratinocytes, were cultured in DMEM medium containing 1% penicillin-streptomycin and 10% FBS, and CCD-986sk cells, which are human skin fibroblasts, were cultured in IMDM containing 1% penicillin-streptomycin and 10% FBS at 37°C in a 5% _CO2 incubator, and passaged once every 3 days.

UVB irradiation was performed by seeding cells at a density of ^5x105 cells/well in 6-well plates, culturing them for 24 hours, replacing the medium with serum-free medium, and starving the cells for 24 hours using a UV irradiator BLX-312 (Vilber Lourmat, Collegien, France). After washing twice with PBS, 1 mL of PBS was dispensed, and then UVB was irradiated at 40 mJ/ ^cm2 .

(4) Gene expression evaluation

Real-time PCR was used for gene expression evaluation. HaCaT and CCD-986sk cells were seeded in 6-well plates at a concentration of 5 x 10 ⁵ cells/well, cultured for 24 hours, replaced with serum-free medium, and starved for 24 hours. Cells were collected after 24 hours by treating with 50 μg/mL of single or complex diluted in serum-free medium. The supernatant was removed by centrifugation at 13,000 rpm for 3 minutes, and RNA was extracted using FavorPrep™ Blood/Cultured Cell Total RNA Mini Kit, and cDNA was synthesized at a concentration of 1000 ng using AccuPower® CycleScript™ RT PreMix (dN6). The synthesized cDNA was subjected to real-time PCR using PowerUp™ SYBR™ Green Master Mix for qPCR. At this time, the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) gene, a housekeeping gene, was used as an internal control, and the gene sequence was as follows.

3. Experimental Results

(1) Measurement of skin moisturizing effect of red bean sprout and spearmint complex

HAS1, HAS2, and HAS3 genes are marker genes for skin moisturizing ability. When human skin keratinocytes were treated with a complex of red bean sprout and spearmint, the expression of HAS1, HAS2, and HAS3 genes was significantly increased in the complex ratios of 0.9:1.1, 1:1, and 1.1:0.9 (red bean sprout: spearmint), compared to the control group. (* p <0.05, ** p <0.01, *** p <0.001 vs. CON) There was no significant difference between the complex ratios of 0.9:1.1, 1:1, and 1.1:0.9 (red bean sprout: spearmint) (NS: Not significant).

When human skin fibroblasts were treated with a red bean sprout and spearmint complex, the expression of MMP1, MMP3, MMP9 genes significantly decreased when the sample was treated compared to the UV group.
