통상적인 염분 제거 과정으로는 물에 끓이거나, 세척하거나, 오랜시간 동안 물에 넣어 두는 방법 등이 있다. 하지만 이와 같은 과정들은 염생식물 내에 유효 성분들의 손실을 가져오는 결과를 낳을 수 있다. 따라서, 본 발명에서는 분체의 혼합, 연합, 과립 과정을 목적으로 이용되는 분체혼합기를 이용하여 제염을 진행한다. 분체 혼합기에 증류수를 넣은 후 제염을 진행할 미역을 넣고 5 내지 10분 동안 가동하면 분체혼합기의 블레이드가 회전하면서 미역 내의 염분이 증류수에 용해되어 제염이 진행된다. 그 후 추출 효율을 저하시키는 미역 내의 수분을 제거하기 위해 정제나 환제에 당의를 입히는 과정을 목적으로 이용되는 당의기(환코팅기)를 이용하여 미역에 과한 열을 주지 않고 수분을 날려 건조한다.Conventional salt removal processes include boiling in water, washing, or leaving in water for a long time. 1. 항산화 활성 평가1. Antioxidant activity evaluation
상기 얻어진 솔루블 및 인솔루블 성분을 DPPH (1,1-diphenyl-2-picrylhydrazyl) 반응을 통해 확인하였다. The obtained soluble and insoluble components were confirmed through DPPH (1,1-diphenyl-2-picrylhydrazyl) reaction.
DPPH (1,1-diphenyl-2-picrylhydrazyl) 2 mg을 ethanol (EtOH) 15 mL에 녹여 만든 DPPH액 1.2 mL과 3 mL의 EtOH와 0.5 mL의 dimethyl sulfoxide (DMSO)를 혼합한 용액을 동일한 비율로 혼합하여 DPPH radical solution을 준비한다. 준비된 DPPH radical solution을 cuvette에 넣고 518 nm의 파장에서 흡광도를 측정하여 그 측정값이 0.95~0.98이 되도록 농도를 조절하였다. 농도가 조절된 DPPH radical solution 900 μL에 고체상태의 솔루블 및 인솔루블 성분을 에탄올에 녹여 100 μg/mL의 농도로 제조한 것 100 uL을 각각 가하여 voltex한 후 10분 후에 518 nm의 파장에서 흡광도를 측정하였다.Prepare a DPPH radical solution by mixing 1.2 mL of DPPH solution prepared by dissolving 2 mg of DPPH (1,1-diphenyl-2-picrylhydrazyl) in 15 mL of ethanol (EtOH) and a solution of 3 mL of EtOH and 0.5 mL of dimethyl sulfoxide (DMSO) in equal ratios. 측정 결과, 솔루블과 인솔루블 성분 모두에서 녹차와 대황보다 뛰어난 항산화 활성을 보였으며, 솔루블은 알로에와 비타민 C와 유사한 항산화 활성을 보였다. 이는 내·외부로터 받는 활성 산소종에 대하여 피부를 방어할 수 있는 능력을 가졌다는 의미로서, 본 발명의 후코이단 추출물의 화장품 원료로서의 사용 가능성을 기대할 수 있었다.As a result of the measurement, both the soluble and insoluble components showed antioxidant activity superior to green tea and rhubarb, and the soluble component showed antioxidant activity similar to aloe and vitamin C. 피부에 수분을 공급하고 피부결을 매끄럽게 해주는 성분으로 알려진 히알루론산(hyaluronic acid)는 우리 몸에서 생합성되는 히알루론산 분해효소(hyaluronidase)에 의해서 분해된다. 후코이단 추출물의 보습 화장품 원료로서 효능을 확인하기 위하여 히알루론산 분해효소 저해 활성을 평가하였다.Hyaluronic acid, known as an ingredient that provides moisture to the skin and smoothens skin texture, is broken down by hyaluronidase, an enzyme that is synthesized in our bodies. 100 μL의 히알루론산 분해효소 용액과 100 μL의 소듐포스페이트 버퍼(sodium phosphate buffer)를 첨가한 뒤, 상기 평가 1에서 제조된 솔루블 성분 및 인솔루블 성분 50 μL을 첨가했다. 37℃에서 10분간 반응시킨 뒤 100 μL의 히알루론산 용액(히알루론산 0.03%, 소듐포스페이트 300 mM, pH 5.35)을 첨가하여 37℃에서 45분간 반응시켰다. 마지막으로 1 mL의 알부민산 용액(BSA 0.1%, 소듐아세테이트 24 mM, 아세트산 79 mM, pH 3.75)을 첨가한 후, 실온에서 10분간 반응시켰고, 600 nm의 파장에서 측정했다. 대조군은 항산화 실험과 동일하게 설정했다(도 3). 상기 추출물 1 및 추출물 2를 혼합하여 후코이단을 포함하는 추출물을 수득 및 농축하는 단계를 포함하며,
상기 미역을 발효시키는 단계에서는 미역 조직을 분해하면서 후코이단의 글리코시드 결합은 끊지 않는 단백질 분해효소(protease)를 사용하는 것을 특징으로 하는, 미역으로부터 후코이단을 포함하는 추출물을 제조하는 방법. A step of fermenting seaweed to obtain a fermented product;
