Abstract
Description
도 1b는 도 1a의 그래프로부터, 50% 생존율 유지 일수(ST50)를 구한 결과를 나타낸 막대 그래프이다.
도 2a는 도 1a와 동일하게, 싱글 셀 상태에서 4℃로 보존한 경우의, 보존 일수에 의한 생존율의 변화를 나타낸 그래프이다. 다만, 도 1a와 동일하게, 보존 개시 시의 생존율을 1.0로 맞추고 있다.
도 2b는 도 2a의 그래프로부터, 80% 생존율 유지 일수(ST80)를 구한 결과를 나타낸 막대 그래프이다.
도 2c는 6일간의 냉장 보존 후, 5일간의 배양에 의한 콜로니 형성능을 나타내는 막대 그래프이다.
도 3a는 3일간의 냉장 보존을 행한 직후에 일어나는, 아포토시스에 의한 세포사의 비율을 나타내는 막대 그래프이다.

도 9는 냉장 보존 배지로서, UW액과, MEM-alpha를 2/1로 혼합한 냉장 보존 배지 「HTM-alpha」(UW액+MEM-alpha, 2:1)를 사용하여, 7일간의 냉장 보존 후에 4일간 재배양했을 때의 콜로니수의 증가율에 대하여 나타내는, 도 7f 및 8과 동일하한 막대 그래프이다. 여기서는, 「HTM-alpha」 및 「HTM1」에, 5mM의 Trolox 및 5배 농도(x5; 10mM)의 GlutaMAX(등록상표, GIBCO)를 첨가했다. 또한, 시판하고 있는 냉장 보존 배지인 HTS FRS를 사용한 결과도 함께 나타낸다.
도 10은 마우스의 배반포기배아(체외 수정 후 3.5일)를, 도 9와 동일한 배지 중에서 3일간 냉장 보존한 후, 2일간의 배양 기간 중에 투명대(透明帶)를 탈출한 비율을 나타내는 막대 그래프이다.
도 11은 도 10의 실험와 동일한 조건에서, 아데닌염산염의 첨가 농도의 영향을 조사한 결과를 나타낸, 도 10과 동일한 막대 그래프이다.
도 12a는 도 6a의 실험와 동일한 조건에서, 하이드록시에틸스타치(HES) 대신에 폴리비닐알코올(PVA)을 사용한 경우의 보호 효과를 나타내는, 막대 그래프이다.

Claims (7)
- 칼륨 이온종의 함량이 30∼80 mmoL/L이며, 나트륨 이온종의 함량이 30∼80 mmoL/L이며, 나트륨 이온종의 양은, 칼륨 이온종에 대한 이온 베이스의 몰비(Na+/K+의 비)로, 0.5∼1.3이며,
트롤록스(Trolox)를, 0.5∼8 mM의 농도로 함유하고,
아데닌 또는 그의 염 혹은 유도체를, 0.1∼4.2 mM의 농도로 함유하고,
필수아미노산에 대하여, 전부, 또는, 1종, 2종 또는 3종을 제외하고 전부 함유하고, 필수아미노산의 토탈 함량은, 50∼200 mg/L이며,
비필수아미노산의 토탈 함량이 50∼200 mg/L인, 인간 또는 동물의 줄기세포 또는 배아를 비동결 상태로 보존하기 위한 냉장 보존액. - (i) 락토비온산 또는 그의 염을 락톤 환산으로 30∼100 mmoL/L, (ii) 라피노스수화물 10∼30 mmoL/L, (iii) 알로퓨리놀 0.3∼1 mmoL/L, (iv) 글루타티온(토탈 글루타티온) 1∼3 mmoL/L, (v) 아데노신 2∼10 mmoL/L, (vi) 리포산 0.1∼0.5 μmoL/L, (vii) 피루브산나트륨 0.1∼0.7 mmoL/L, (viii) 글루코오스 1∼5 mmoL/L, (ix) 아스코르브산 0.03∼0.3 mmoL/L, 및 트롤록스(Trolox) 5∼8 mM, (x) 아데닌 또는 그의 염 혹은 유도체 0.1∼4.2 mM, (xi) 그 외의 비타민 또는 그의 수용성 아날로그·유도체, (xii) 필수아미노산을 토탈로 50∼200 mg/L, (xiii) 비필수아미노산을 토탈로 100∼500 mg/L, (xiv) 칼륨 이온종 30∼80 mmoL/L, 및 (xv) 나트륨 이온종 20∼90 mmoL/L를 포함하는 생리완충액이며, 나트륨 이온종의 양은, 칼륨 이온종에 대한 이온 베이스의 몰비(Na+/K+의 비)로, 0.5∼1.3이고, 상기 비타민은 폴산, 니코틴아미드(나이아신아미드), 리보플라빈, B12, 콜린, 이노시톨, 판토텐산, 피리독살인산, 티아민을 포함하는, 인간 또는 동물의 줄기세포 또는 배아를 비동결 상태로 보존하기 위한 냉장 보존액.
- 제1항에 있어서,
칼륨 이온종에 대한 이온 베이스의 몰비(Na+/K+의 비)가 1보다 작은, 냉장 보존액. - 제1항에 있어서,
하이드록시에틸전분을 포함하지 않고, 폴리비닐알코올을 2.5∼25 g/L 포함하는, 냉장 보존액. - 제4항에 있어서,
만니톨을 20∼100 mmoL/L 더 포함하는, 냉장 보존액. - 제1항에 있어서,
L-알라닐-L-글루타민, 또는, 그 외의 디펩티드 대체품, 또는 그의 염을 2∼5 mM의 농도로 포함하는, 냉장 보존액. - 제1항 내지 제6항 중 어느 한 항에 기재된 냉장 보존액 중에, 인간 또는 동물의 iPS 세포 혹은 그 외의 인공줄기세포, 그 외의 줄기세포, 이것으로부터 얻어지는 오르가노이드, 또는 배아를, 부유 또는 침지시키고, 2∼8 ℃에서, 1∼10 일 보존하는, 냉장 보존 방법.
