Abstract
Description
도 2는 본 발명의 DNA 결합 자성 나노입자의 핵산 분해 효소(Endonuclease, DNase) 반응성 확인 결과를 보여준다.
도 3은 발색반응을 이용한 DNA 결합 자성 나노입자의 E.coli 검출 결과를 보여준다.
도 4는 발색반응을 이용한 DNA 결합 자성 나노입자의 양에 따른 E.coli 검출 결과를 보여준다.
도 5는 발색반응을 이용한 DNA 결합 자성 나노입자의 Acinetobacter baumannii 검출 결과를 보여준다.
도 6은 발색반응을 이용한 DNA 결합 자성 나노입자의 Staphylococcus aureus 검출 결과를 보여준다.
Claims (17)
- 자성 나노입자 표면-고정화된 올리고뉴클레오타이드 프로브를 포함하는, 살아있는 미생물 현장 오염 검출용 조성물로서,
상기 올리고뉴클레오타이드 프로브는, 이의 3'-말단이 자성 나노입자 표면에 고정화되고, 이의 5'-말단은 검출가능한 시그널을 발생시키는 효소 표지를 포함하며;
상기 효소 표지는 미생물 뉴클레아제의 핵산 절단 반응에 의해 절단되어 방출되면서 상기 검출가능한 시그널을 발생시키고;
상기 시그널은 발색 반응이며;
상기 효소 표지는 바이오틴에 의해 연결되고;
상기 바이오틴은 스트렙타아비딘, 아비딘(avidin), 트랩타비딘(traptavidin) 및 뉴트라비딘(neutravidin)으로 이루어진 군에서 선택되는 1 종 이상의 바이오틴-친화성 단백질과 결합하며;
상기 효소는 HRP(Horseradish peroxidase), ALP(Alkaline phosphatase), 루시퍼라이제(luciferase), 혈당 분해 효소, 인버타제(invertase), 글루코스 옥시다아제(Glucose Oxidase), 베타-디-갈락토시다아제(β-D-galactosidase), MDH(malate dehydrogenase) 및 아세틸콜린에스터라아제(acetylcholinestrerase)로 이루어진 군에서 선택되는 것을 특징으로 하는, 조성물.

(c) 상기 시료에서 방출되는 시그널을 검출하는 단계;
를 포함하는 살아있는 미생물 현장 오염 진단 방법으로서,
상기 시그널의 검출은 상기 미생물의 존재를 나타내는 것을 특징으로 하는, 방법.
