UVB에 의한 광노화 억제 효능 시험에서는 추출물 농도 2% (v/v)에서 음성대조군 대비 22.7% 향상된 광노화 억제 효능이 있었다(도 17). 미백 효능 시험 결과에서도 추출물은 농도 의존적으로 L-DOPA 저해 활성을 나타내었으며, 5.14%의 IC50 (50% Inhibition concentration)값을 확인하였다(도 18). 추출물 농도 0.5% (v/v)에서 보습 인자인 HAS2 및 AQP3 발현은 음성대조군 대비 각각 81.5% 및 50.5% 증가되는 효과가 있는 것으로 나타났다(도 19).As a result of the test, in the antioxidant efficacy test using DPPH, the complex plant hot water extract showed DPPH scavenging activity as the concentration increased, and a SC 50 (50% Scavenging concentration) value of 1.63% (v/v) was confirmed (Figure 14 ). At an extract concentration of 0.5% (v/v), the expression of moisturizing factors HAS2 and AQP3 was found to be increased by 81.5% and 50.5%, respectively, compared to the negative control group (FIG. 19).

시험 결과, DPPH를 이용한 항산화 효능 시험에서 복합식물 열수 추출물은 농도가 증가함에 따라 DPPH 소거 활성을 나타내었으며, 0.18% (v/v)의 SC50 (50% Scavenging concentration) 값을 확인하였다(도 20). 또한 HDFn를 이용한 항산화 효능 시험 결과 추출물 농도 2% (v/v)에서 음성대조군(추출물 무처리) 대비 69.5% 향상된 항산화 효능이 있었고(도 21), 추출물 농도 0.5% (v/v)에서 항산화 인자인 Nrf2, AP1 및 TRX 발현이 음성대조군 대비 각각 111.4%, 141.2% 및 90.5% 증가되는 효과가 있었다(도 22). UVA에 의한 광노화 억제 효능 시험에서는 추출물 농도 2% (v/v)에서 음성대조군 대비 15.4% 향상된 광노화 억제 효능이 있었고(도 23), UVB에 의한 광노화 억제 효능 시험에서는 25.7% 향상된 효능이 있었으며(도 24), 48.7% 향상된 항염 효능이 있었다(도 25). 미백 효능 시험 결과에서도 추출물은 농도 의존적으로 L-DOPA 저해 활성을 나타내었으며, 0.89%의 IC50 (50% Inhibition concentration)값을 확인하였다(도 26). 추출물 농도 0.5% (v/v)에서 보습 인자인 HAS2 및 AQP3 발현은 음성대조군 대비 각각 184.8% 및 44.4% 증가되는 효과가 있는 것으로 나타났다(도 27).As a result of the test, in the antioxidant efficacy test using DPPH, the complex plant hot water extract showed DPPH scavenging activity as the concentration increased, and a SC 50 (50% Scavenging concentration) value of 0.18% (v/v) was confirmed (Figure 20 ). 범부채뿌리, 인디언구스베리열매, 또는/및 층층나무 식물체의 열수 또는 고주파 추출물의 피부 개선 효과에 대한 상기 실험결과 1 내지 4를 종합하면, 범부채뿌리 단일 열수 추출물은 항산화 및 항염 효과가 있었으나, 미백 및 광노화(UVA 및 UVB)에는 효과가 없었다. 인디언구스베리 단일 열수 추출물은 항산화, 항염, 미백, 및 UVB에 의한 광노화에서 모두 효과가 있었으나, UVA에 의한 광노화는 억제 효능이 없었다. 층층나무 단일 열수 추출물은 항산화 및 미백에는 효과가 있었으나, 항염 및 광노화(UVA 및 UVB)에는 효과가 없는 것으로 나타났다. 상기 범부채뿌리, 인디언구스베리, 또는 층층나무의 단일 열수 추출물에서 보습 효과는 시험되지 아니하였다. 범부채뿌리, 인디언구스베리, 및 층층나무를 복합으로 열수 추출한 경우에는 항산화, 미백, 보습, 및 UVB에 의한 광노화에 효과가 있는 것으로 나타났으나, 항염과 UVA에 의한 광노화에는 효과가 없었다. 범부채뿌리 단일 추출물, 및 인디언구스베리 단일 추출물이 각각 항염 효과가 있음에도 불구하고, 이들이 층층나무 추출물과 함께 복합으로 이용될 때 항염 효능이 소실되는 것은 예측하기 어려운 효과로서, 추출물을 복합적으로 이용하는 것이 항상 가산 효과를 야기하는 것은 아님을 의미한다. 반면, 범부채뿌리, 인디언구스베리, 및 층층나무를 복합으로 고주파 추출한 경우에는 항산화, 미백, 보습뿐만 아니라 항염 및 광노화(UVA 및 UVB)에도 효과가 있는 것으로 나타나, 동일 성분일지라도 추출 방법에 의하여 효과가 상이하며, 고주파 추출한 경우 열수 추출에 비해 피부 개선 효과가 우수함을 알 수 있었다. 그러나 범부채뿌리, 또는 층층나무를 단일 추출 시에는 열수 추출의 경우가 고주파 추출보다 항산화 효과가 우수함을 고려하면, 항상 고주파 추출이 열수 추출에 비해 우수한 효과를 지니는 것은 아니며, 식물체 종류와 추출법의 조합의 특징이 있는 것으로 해석된다. 결론적으로, 본 발명을 통해 범부채뿌리, 인디언구스베리, 및 층층나무를 복합으로 고주파 추출하여 이용하는 경우에 가장 우수한 피부 개선 효과가 기대된다.Summarizing the above experimental results 1 to 4 on the skin improvement effects of hot water or high-frequency extracts of Panfanasi root, Indian gooseberry fruit, or/and dogwood plant, the single hot water extract of Panfanart root had antioxidant and anti-inflammatory effects, but whitening and photoaging effects. A combined hot water extraction of pansy root, Indian gooseberry, and dogwood was found to be effective in antioxidant, whitening, moisturizing, and UVB-induced photoaging, but was not effective in anti-inflammatory and UVA-induced photoaging. Although the single extract of pansy root and the single extract of Indian gooseberry each have anti-inflammatory effects, when they are used in combination with dogwood extract, the loss of anti-inflammatory efficacy is an effect that is difficult to predict, so using the extract in combination is always an added benefit. This means that it does not cause any effect. On the other hand, high-frequency extraction of a combination of pansy root, Indian gooseberry, and dogwood was found to be effective not only in antioxidants, whitening, and moisturizing, but also in anti-inflammatory and photoaging (UVA and UVB) effects. Even if the ingredients are the same, the effects differ depending on the extraction method. It was found that high-frequency extraction had a superior skin improvement effect compared to hot water extraction.
