본 발명의 레스베라트롤을 함유하는 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자는 유효성분인 레스베라트롤을 다량 포함할 수 있을 뿐 아니라, 입자 크기가 작고 균일하며 약물전달 효율이 높은 것으로 알려진 탄산칼슘 결정 형태인 배터라이트 및 칼사이트를 다수 포함할 수 있다. 또한 상기 나노입자는 안정성이 높고, 유효성분인 레스베라트롤의 안정성, 세포 보호 및 항산화 효능, 피부흡수율 및 경피투과율을 높일 수 있고, 레스베라트롤이 높은 농도에서 나타내는 세포 독성을 낮추어 안전성을 높일 수 있으며, 레스베라트롤이 갖는 항염증 효과를 가지는 것으로 나타나, 고기능성 화장품 소재로서의 높은 유용하게 사용될 수 있다.
Description
도 1은 레스베라트롤 원액(a) 및 실시예 1에 따라 제조된 레스베라트롤(RSV)을 함유하는 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘(PVP-PAA-CaCO3) 나노입자(b)의 이미지이다.
도 2는 실시예1.1에 따른 나노입자 제조 시 PAA-CaCl2와 Na2CO3간 비율 조정에 따른 RSV의 PVP-PAA-CaCO3 나노입자 내 봉입율을 분석한 결과이다.
도 3은 레스베라트롤 단독(RSV), 폴리아크릴산(PAA), 폴리비닐피롤리돈(PVP) 및 실시예1.1에 따라 제조된 RSV를 함유하는 PVP-PAA-CaCO3 나노입자 (도면 내 RSV-PAA,PVP로 표기)에 대한 FT-IR 비교분석(a) 및 PVP-PAA-CaCO3 나노입자 단독 FT-IR 분석(b)한 결과이다.
도 4는 실시예1.1에 따라 제조된 RSV를 함유하는 PVP-PAA-CaCO3 나노입자의 주사전자현미경(SEM) (a) 및 투과전자현미경(TEM) (b) 이미지(일부 입자 크기 표기)이다.

상기 혼합물에 탄산칼슘의 전구체를 혼합하는 단계;
를 포함하고,
상기 레스베라트롤, 폴리비닐피롤리돈 및 폴리아크릴산의 혼합물에 탄산칼슘의 전구체를 혼합하는 단계는,
상기 레스베라트롤, 폴리비닐피롤리돈 및 폴리아크릴산의 혼합물에 칼슘염을 혼합한 제3 혼합물; 및 탄산염을 혼합하는 것이고,
상기 제3 혼합물과 탄산염은 1:8 내지 1:10의 부피비율로 혼합되는 것이고,
상기 칼슘염과 탄산염은 3:1 내지 5:1의 몰비로 혼합되는 것인, 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자의 제조방법.
- 제4항에 있어서,
상기 레스베라트롤, 폴리비닐피롤리돈 및 폴리아크릴산을 혼합하는 단계는,
레스베라트롤을 유기용매에 혼합한 제1혼합물; 및
폴리비닐피롤리돈 및 폴리아크릴산을 수계용매에 혼합한 제2혼합물;을 각각 준비한 후 제1혼합물 및 제2혼합물을 섞어 제조되는 것인, 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자의 제조방법.
- 제5항에 있어서,
상기 제1혼합물 내 레스베라트롤은 0.5 내지 10중량% 포함되는 것인, 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자의 제조방법.
- 제5항에 있어서,
상기 제2혼합물 내 폴리비닐피롤리돈은 0.1 내지 10중량% 포함되고,
상기 제2혼합물 내 폴리아크릴산은 0.05 내지 5중량% 포함되는 것인, 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자의 제조방법.
- 제5항에 있어서,
상기 제1혼합물과 제2혼합물의 비율(v/v)은 1:99 내지 20:80인 것인, 폴리비닐피롤리돈-폴리아크릴산-탄산칼슘 나노입자의 제조방법.
