Claims (20)
- 바실러스 (Bacillus) 속 미생물 유래의 α-1,2-푸코오스 전이효소를 암호화하는 핵산 서열과, 이와 작동 가능하게 연결된 항시 발현 프로모터를 포함하는 조절 서열을 포함하며,
바실러스 (Bacillus) 속 미생물에서 α-1,2-푸코오스 전이효소를 발현하는 유전자 발현 카세트 (gene expression cassette). - 제1항에 있어서, 상기 항시 발현 프로모터는 p3610, p12455, p24930, pTu, 및 pPDC로 이루어지는 군에서 선택된 1종 이상인, 유전자 발현 카세트.
- 제1항에 있어서, 상기 조절 서열은 서열번호 21의 핵산 서열을 포함하는 리보좀 결합 영역 (Ribosome Binding Site, RBS)을 포함하는, 유전자 발현 카세트.
- 제1항에 있어서, 상기 α-1,2-푸코오스 전이효소를 암호화하는 핵산 서열은 서열번호 2 내지 5로 이루어지는 군에서 선택된 1종 이상의 아미노산 서열을 암호화하는 핵산 서열을 포함하는 것인, 유전자 발현 카세트.
- 제1항에 있어서, 상기 α-1,2-푸코오스 전이효소를 암호화하는 핵산 서열은 서열번호 7 내지 10으로 이루어지는 군에서 선택된 1종 이상의 핵산 서열을 포함하는 것인, 유전자 발현 카세트.
- 제1항에 있어서, 상기 유전자 발현 카세트는 락토오스 막 수송 단백질을 암호화하는 핵산 서열을 추가로 포함하는 것인, 유전자 발현 카세트.
- 제6항에 있어서, 상기 락토오스 막 수송 단백질은 Lac12 및 LacY로 이루어지는 군에서 선택된 1종 이상인, 유전자 발현 카세트.
- 제6항에 있어서, 상기 락토오스 막 수송 단백질의 조절 서열을 추가로 포함하며, 상기 조절 서열은 프로모터 및 RBS로 이루어지는 군에서 선택된 1종 이상인, 유전자 발현 카세트.
- 제1항에 있어서, 상기 유전자 발현 카세트는 바실러스 메가테리움 (Bacillus megaterium), 바실러스 서브틸리스 (Bacillus subtilis), 바실러스 세레우스 (Bacillus cereus), 바실러스 리체니포르미스 (Bacillus licheniformis), 바실러스 아밀로리퀴파시엔스 (Bacillus amyloliquefaciens), 및 바실러스 투링기엔시스 (Bacillus thuringiensis) 로 이루어지는 군에서 선택된 1종 이상에서 α-1,2-푸코오스 전이효소를 발현하는 것인, 유전자 발현 카세트.
- 제1항 내지 제9항 중 어느 한 항에 따른 α-1,2-푸코오스 전이효소를 발현하는 유전자 발현 카세트를 포함하는 벡터.
- 제10항에 있어서, 상기 벡터는 도 1a의 개열지도를 가지는 재조합 벡터인, 벡터.
- 제1항 내지 제9항 중 어느 한 항에 따른 유전자 발현 카세트를 포함하거나, 상기 발현 카세트를 포함하는 벡터로 형질전환된 재조합 바실러스 속 미생물.
- 제12항에 있어서, 상기 벡터는 푸코오스 합성 유전자 발현 카세트를 추가로 포함하는 것, 또는 상기 미생물은 푸코오스 합성 유전자 발현 카세트를 포함하는 벡터를 추가로 포함하는 것인, 미생물.

- 제13항에 있어서, 상기 푸코오스 합성 유전자 발현 카세트는,
GTP 만노오스-1-포스페이트 구아닐릴트랜스퍼라아제를 암호화하는 핵산 서열, 포스포만노뮤타아제를 암호화하는 핵산 서열, GDP-L-푸코오스 신타아제 (WcaG)를 암호화하는 핵산 서열, 및 GDP-D-만노오스-4,6-데하이드라타아제를 암호화하는 핵산 서열로 이루어지는 군에서 선택된 1종 이상과, 이와 작동 가능하게 연결된 조절 서열을 포함하며,
상기 조절 서열은 항시 발현 프로모터를 포함하는 것인, 미생물. - 제15항에 있어서, 상기 푸코오스 합성 유전자 발현 카세트에 포함되는 조절 서열은 p3610, p12455, p24930, p0706, pTu, 및 pPDC로 이루어지는 군에서 선택된 1종 이상의 항시 발현 프로모터를 포함하는 것인, 미생물.
- 제13항에 있어서, 상기 푸코오스 합성 유전자 발현 카세트를 포함하는 벡터는 도 1b의 개열지도를 가지는 재조합 벡터인, 미생물.
- 제12항에 있어서, 상기 바실러스 속 미생물은 바실러스 메가테리움 (Bacillus megaterium), 바실러스 서브틸리스 (Bacillus subtilis), 바실러스 세레우스 (Bacillus cereus), 바실러스 리체니포르미스 (Bacillus licheniformis), 바실러스 아밀로리퀴파시엔스 (Bacillus amyloliquefaciens), 및 바실러스 투링기엔시스 (Bacillus thuringiensis)로 이루어지는 군에서 선택된 1종 이상인, 미생물.
- 제12항에 따른 재조합 바실러스 속 미생물을 이용하여 얻어진 α-1,2-푸코오스 전이효소, 상기 미생물의 균체, 상기 미생물의 배양물, 상기 미생물의 파쇄물, 및 상기 파쇄물 또는 배양물의 추출물로 이루어진 군에서 선택된 1종 이상을 포함하는, 2'-푸코실락토오스 생산용 조성물.
- 제1항 내지 제9항 중 어느 한 항에 따른 유전자 발현 카세트를 포함하거나, 상기 발현 카세트를 포함하는 벡터로 형질전환된 재조합 바실러스 속 숙주세포를 배양하는 단계를 포함하는, 2’-푸코실락토오스 (2’-fucolsylactose)의 제조방법.
