Abstract
Description
도 2는 덱스트린 유래 고중합 이소말토올리고당 제조효소(Tt-LIMO)를 암호화하는 염기 서열이 클로닝된 덱스트린 유래 고중합 이소말토올리고당 제조효소(Tt-LIMO) 재조합 벡터를 나타내는 도면이다.
도 3은 전기영동 분석을 통해 덱스트린 유래 고중합 이소말토올리고당 제조효소가 성공적으로 제조되었음을 확인한 결과를 나타내는 도면이다.
도 4는 고중합 이소말토올리고당의 제조를 위한 기질 탐색을 위해 시간 경과에 따른 TG(시중효소, 실선) 및 TtLIMO(점선)에 의한 말토오스(M2, 왼쪽 그래프) 및 말토테트라오스(M4, 오른쪽 그래프)와의 반응 생성물의 상대적 조성을 분석한 결과를 나타내는 도면이다.

2) 상기 배양물로부터 당전이효소를 수득하는 단계; 및
3) 상기 당전이효소를 덱스트린(Dextrin)에 첨가하여 반응시키는 단계;를 포함하는 덱스트린 유래 중합도 7 내지 13인 고중합 이소말토올리고당의 제조 방법으로 제조되고,
상기 덱스트린은 3 내지 25인 중합도(degree of polymerization, DP)를 가지는 포도당이 α-1,4 형태로 결합된 단일물질 또는 혼합물이고,
상기 고중합 이소말토올리고당(LIMO)은 전체 중량의 33 내지 46 중량 %이고,
지소화성이 있는 덱스트린 유래 중합도 7 내지 13인 고중합 이소말토올리고당(LIMO)을 유효성분으로 포함하는, 당뇨 개선용 조성물.
상기 당전이효소는 써모안아에로박터 써모코프리애 (Thermoanaerobacter thermocopriae) 균주 유래인 것을 특징으로 하는, 당뇨 개선용 조성물.
상기 당전이효소는 고리형 이소말토올리고당 당전이효소(Cycloisomaltooligosacharride glucanotransferase, TPCITase)의 아미노산 서열을 암호화하는 전체 염기서열 서열번호 1에서 아미노산 C- 말단 서열을 암호화하는 서열번호 2를 포함하며, 덱스트린을 기질로하여 당전이 생성물을 제조하는 것을 특징으로 하는, 당뇨 개선용 조성물.
단계 1)의 재조합 벡터는 pET28a, pET, pGEX, pQE, pDEST 및 pCOLD로 이루어진 군에서 선택되는 하나의 벡터인 것을 특징으로 하는, 당뇨 개선용 조성물.
단계 1)의 형질전환체는 BL21(DE3), Rosetta(DE3), Rosetta2(DE3), ArcticExpress(DE3), STAR(DE3), C41(DE3) 및 C43(DE3)로 이루어진 군에서 선택되는 하나의 대장균에 형질전환된 것인 것을 특징으로 하는, 당뇨 개선용 조성물.
