Claims (12)
- 시료에서 핵산인 DNA를 추출하는 1단계;
RT-PCR 또는 PCR을 이용하여 설치류에 의한 오염여부를 확인하는 2단계;를 포함하는 식품의 오염여부를 판별하는 방법에 있어서,
상기 1단계는 a) 식품으로부터 샘플을 획득하는 단계;
b) 상기 획득한 샘플을 반응 용기로 옮겨 상기 샘플을 수-비혼화성상(water-immiscible phase)에 접촉시키는 단계;
c) 상기 수-비혼화성상에 접촉된 상기 샘플을 용해시키는 단계;
d) 상기 용해된 샘플로부터 DNA를 추출하는 단계;
를 포함하며,
상기 2단계는 서열번호 1 내지 5의 프라이머 세트를 이용하여 RT-PCR 또는 PCR을 수행하는 단계인 것을 특징으로 하는 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.
- 제1항에 있어서,
상기 용해된 샘플로부터 DNA를 추출하는 단계는 A260/A280 및 A260/A230이 1.7 내지 2.2 일 경우 PCR에 적합한 DNA로 판단하는 것을 포함하는 식품의 오염여부를 판별하는 방법.

- 제1항 또는 제2항에 있어서, 단계 a)의 식품으로부터 획득하는 샘플은 절임배추, 고춧가루, 가공식품으로 이루어진 군에서 선택된 어느 하나인 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.

- 제1항 또는 제2항에 있어서, 원심 분리단계를 더 포함하는 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.

- 제1항 또는 제2항에 있어서, 상기 단계b)의 수-비혼화성상(water-immiscible phase)은 유기상인 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.
- 제1항 또는 제2항에 있어서, 상기 단계b)의 수-비혼화성상(water-immiscible phase)은 알코올과 혼화되지 않는 것임을 특징으로 하는 식품의 오염여부를 판별하는 방법.
- 제1항 또는 제2항에 있어서, 상기 단계b)의 수-비혼화성상(water-immiscible phase)은 사이클릭, 포화 또는 불포화 탄화수소 또는 방향족 탄화수소를 포함하는 것임을 특징으로 하는 식품의 오염여부를 판별하는 방법.
- 제1항 또는 제2항에 있어서, 상기 단계c)는 열 처리가 수행되어 상기 수-비혼화성상에 접촉된 상기 샘플의 용해를 촉진시키는 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.
- 제11항에 있어서, 상기 열 처리는 80 ℃ 내지 89 ℃에서 3 내지 5분 동안 가열하는 것을 특징으로 하는 식품의 오염여부를 판별하는 방법.
