Abstract
Description
도 2는 본 발명의 일 실시예에 있어서, 세포 배양층 표면 거칠기의 변화를 온도 및 시간에 따라 나타낸 것이다.
도 3(a)는 본 발명의 일 실시예에 있어서, 줄기세포 시트를 제조하는 방법에 의해 프라이밍되는 줄기세포를 역주기형광현미경(phase-contrast inverted fluorescence microscope)의 X100배율로 촬영한 이미지를 나타내며, 도 3(b)는 프라이밍이 완료되어 수확된 줄기세포 시트를 나타낸다.
도 4(a)는 본 발명의 일 실시예에 있어서, 줄기세포 시트를 제조하는 방법에 사용되는 공배양 세포의 종류에 따라, 프라이밍된 줄기세포를 역주기형광현미경의 X40배율로 촬영한 이미지를 나타내며, 도 4(b)는 사용되는 공배양 세포의 종류에 따라, 최종적으로 수확된 줄기세포 시트 내에 저장되는 측분비인자의 양 및 종류를 나타낸다.
Claims (12)
- 다공성 멤브레인 및 상기 다공성 멤브레인 표면의 세포 배양층을 포함하는 세포 배양용 멤브레인의 일면에 줄기세포를 접종하는 단계;
상기 줄기세포가 접종된 면의 반대면에 공배양 세포를 접종하는 단계;
상기 공배양 세포를 접종하는 단계에 후속적으로, 5 내지 10일 동안 32℃ 초과 내지 40℃ 이하의 온도에서 줄기세포를 프라이밍하는 단계; 및
상기 줄기세포를 프라이밍한 후, 0℃ 이상 내지 32℃ 미만의 온도에서 상기 줄기세포를 탈착하는 단계를 포함하고,
상기 줄기세포는 중간엽 줄기세포((mesenchymal stem cell))이고,
상기 공배양 세포는 HUVEC(인간 제정맥 내피세포), ASMC(대동맥평활근세포), 및 hDF(인간 섬유아세포)로 이루어진 그룹으로부터 선택되는 적어도 하나인, 줄기세포 시트를 제조하는 방법.
- 제1항에 있어서, 상기 세포 배양층은 N-이소프로필아크릴아마이드 단량체, 가교제, 중합 개시제 및 잔부의 물을 포함하는 코팅 조성물을 이용하여 중합된 폴리이소프로필아크릴아마이드를 포함하는, 줄기세포 시트를 제조하는 방법.
- 제2항에 있어서,
상기 가교제는 N-이소프로필아크릴아마이드 단량체 및 물의 혼합물 100중량부를 기준으로 1중량부 이상 내지 5중량부 미만인, 줄기세포 시트를 제조하는 방법.
- 제1항에 있어서,
상기 세포 배양층은 32℃ 초과 내지 40℃ 이하에서는 세포가 부착되고, 0℃ 이상 내지 32℃ 미만에서는 세포가 탈착되는, 줄기세포 시트를 제조하는 방법.
- 제1항에 있어서,
상기 세포 배양층은 32℃ 초과 내지 40℃ 이하에서 표면 거칠기가 4 내지 37nm이고, 0℃ 이상 내지 32℃ 미만에서 표면 거칠기가 4㎛ 이상인, 줄기세포 시트를 제조하는 방법.
- 제3항에 있어서,
상기 가교제는 N,N'-메틸렌비스아크릴아미드(N,N-Methylenebisacrylamide;MBAAm), 테트라메틸에틸렌디아민(tetramethylethylenediamine,TEMED) 또는 이들의 혼합인, 줄기세포 시트를 제조하는 방법.
- 제1항에 있어서,
상기 다공성 멤브레인의 공극은 평균 직경 100nm 내지 20㎛의 기공 사이즈를 갖는 것인, 줄기세포 시트를 제조하는 방법.

상기 줄기세포를 접종하는 단계 및 공배양 세포를 접종하는 단계는 32℃ 초과 내지 40℃ 이하의 온도에서 수행되는, 줄기세포 시트를 제조하는 방법.

상기 줄기세포를 접종하는 단계에서 줄기세포는 1X104 cells/cm2 내지 1X105 cells/cm2의 양으로 접종되며,
