Claims (10)
- 태아를 임신한 여성 대상체의 생물학적 샘플에서 태아 DNA의 분획 농도를 결정하는 방법으로서, 생물학적 샘플이 여성 대상체 및 태아로부터의 핵산 분자를 포함하고, 여성 대상체가 제1 대립유전자에 대한 제1 유전자좌에서 동형접합성이고, 태아가 제1 대립유전자 및 상기 제1 대립유전자와 상이한 제2 대립유전자에 대한 제1 유전자좌에서 이형접합성이며, 하기를 포함하는 방법:
생물학적 샘플로부터의 핵산 분자를 포함하는 제1 다수의 반응으로부터 제1 데이터를 수득하는 단계로서, 반응이 두 개의 관심있는 폴리뉴클레오티드 서열의 존재 또는 부재를 나타내고, 두 개의 관심있는 폴리뉴클레오티드 서열이 제1 유전자좌에서의 제1 대립유전자 및 제2 대립유전자이고, 제1 다수의 반응이 디지털 중합효소 연쇄 반응이고, 제1 데이터가
(1) 제1 대립유전자의 존재에 대해 양성인 반응의 제1 수를 나타내는 제1 세트의 정량 데이터; 및
(2) 제2 대립유전자의 존재에 대해 양성인 반응의 제2 수를 나타내는 제2 세트의 정량 데이터를 포함하는 단계; 및
태아 DNA의 분획 농도를 결정하기 위해 제2 수에 대한 제1 수의 분획을 계산하는 단계. - 제1항에 있어서, 태아 DNA의 분획 농도가 제1 수 및 제2 수를 이용하여 백분율로 결정되는 것인 방법.
- 제1항에 있어서, 하기를 더 포함하는 방법:
생물학적 샘플로부터의 핵산 분자를 포함하는 제2 다수의 반응으로부터 제2 데이터를 수득하는 단계로서, 제2 데이터가
(1) 임상적으로 관련된 핵산 서열의 제3 양(amount)을 나타내는 제3 세트의 정량 데이터; 및
(2) 임상적으로 관련된 핵산 서열과 상이한 배경 핵산 서열의 제4 양을 나타내는 제4 세트의 정량 데이터를 포함하고, 배경 핵산 서열이 공지된 임상적으로 관련된 핵산 서열에 대해 정상 비율을 갖고, 임상적으로 관련된 핵산 서열 및 배경 핵산 서열이 태아의 제1 유형의 세포 및 임신한 여성의 하나 이상의 제2 유형의 세포로부터 유래하는 단계;
제3 세트의 정량 데이터 및 제4 세트의 정량 데이터로부터 파라미터를 결정하는 단계로서, 파라미터가 제3 양과 제4 양 사이의 비율을 포함하는 단계;
태아 DNA의 분획 농도로부터 제1 분별값(cutoff value)을 유도하는 단계;
파라미터를 제1 분별값과 비교하는 단계; 및
