Description
도 2는 도 1의 신속한 ATF 유가식 수확 수행 방법에 관한 흐름도이다.

Claims (37)
- 세포가 배양 배지 중에서 세포 생성물을 원하는 수확 농도로 생산할 때까지 세포를 유가식 또는 농축 유가식 배양물 중에서 배양하는 단계; 및
배양 배지를 교번 접선 유동 (ATF) 장치를 통과시킴으로써 세포 생성물을 수확하여 세포 생성물을 포함하는 투과물을 수득하는 단계이며, 여기서
생존가능한 세포 밀도 (VCD)가 미리 결정된 값 초과이면, 적어도 90%의 세포 생성물이 투과물 내에 존재할 때까지 5 내지 30 ℓ/㎡/시간 (LMH)의 플럭스로 정용여과 모드에서 상기 ATF 장치를 작동시키거나, 또는
VCD가 상기 미리 결정된 값 미만이면, VCD가 상기 미리 결정된 값에 도달하거나 배양 배지가 출발 부피보다 낮은 미리 결정된 부피에 도달할 때까지 농축 모드에서 10 내지 30 LMH의 플럭스로 상기 ATF 장치를 작동시키고, 그 이후 적어도 90%의 세포 생성물이 투과물 내에 존재할 때까지 정용여과 모드에서 작동시키는 것인 단계
를 포함하는, 세포 배양물로부터 세포 생성물을 수확하는 방법. - 제1항에 있어서, 상기 정용여과 모드가 단계식 정용여과 또는 연속 정용여과인 것인 방법.
- 제2항에 있어서, 정용여과가 보충액으로 포스페이트 완충 염수를 사용하는 것인 방법.
- 제2항에 있어서, 정용여과가 보충액으로 배양 배지를 사용하는 것인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 수확이 500 ℓ 내지 2,000 ℓ 부피에 대하여 18시간 미만이 소요되는 것인 방법.
- 제5항에 있어서, 수확이 6.0시간 미만이 소요되는 것인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 미리 결정된 부피가 출발 부피의 5% 내지 30%인 방법.
- 제7항에 있어서, 미리 결정된 부피가 출발 부피의 10% - 20%인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 세포 배양물이 농축 유가식 배양물을 포함하고, 여기서 미리 결정된 부피가 출발 부피의 50%인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, ATF 장치가 중공 섬유 필터를 포함하는 것인 방법.
- 제10항에 있어서, 필터가 공극 크기 0.1 내지 5.0 ㎛, 또는 500 내지 1,000 kD을 갖는 것인 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 세포 배양물이 유가식 세포 배양물인 방법.

- 유입구 및 배출구를 포함하는 생물반응기;
생물반응기 유입구에 연결된, 세포 생성물을 함유하지 않는 유체 배지 공급원;
생물반응기 배출구에 연결된 교번 접선 유동 (ATF) 장치;
ATF 장치의 배출구에 연결되고, ATF 장치로부터 유체가 나오도록 구성된 펌프; 및
세포가 배양 배지 중에서 세포 생성물을 원하는 수확 농도로 생산할 때까지 세포를 유가식 또는 농축 유가식 배양물 중에서 배양하고,
배양 배지를 ATF 장치를 통과시킴으로써 세포 생성물을 수확하여 세포 생성물을 포함하는 투과물을 수득하며, 여기서
생존가능한 세포 밀도 (VCD)가 미리 결정된 값 초과이면, 적어도 90%의 세포 생성물이 투과물 내에 존재할 때까지 5 내지 30 ℓ/㎡/시간 (LMH)의 플럭스로 정용여과 모드에서 상기 ATF 장치를 작동시키거나, 또는
