Abstract
Description
도 2는 본 발명의 핵산 추출방법의 과정을 간략하게 나타낸 모식도이다. 용해 완충액에 의해 생물학적 시료가 용해되면서 핵산과 부산물이 분리되고 이온 전하 결정체가 양이온성 부산물을 부착시킴과 동시에 물을 흡수하면서 핵산이 농축된 완충액을 얻는 과정을 보여준다.
도 3은 Oropharyngeal sample(구인두암 샘플)에 본 발명의 핵산 추출방법(Test 2)과 용해 완충액(DLB)만을 사용한 추출방법(Test 1)을 각각 적용한 후, Target 유전자(GAPDH)의 Real-time PCR 증폭 효율을 비교한 결과를 도시한 것이다.
도 4는 Nasopharyngeal sample test(비인두암 샘플)에 본 발명의 핵산 추출방법(Test 2)과 용해 완충액(DLB)만을 사용한 추출방법(Test 1)을 각각 적용한 후, Target 유전자(GAPDH)의 Real-time PCR 증폭 효율을 비교한 결과를 도시한 것이다.

도 9는 다양한 형태의 고분자 흡수체를 사용하여 핵산을 분리 및 농축 한 후, 각 방법의 핵산 분리효능 PCR 검출 시험을 통해 확인한 결과를 도시한 것이다.
Claims (13)
- 핵산 분리용 다공성 이온 전하 결정체로
상기 다공성 이온 전하 결정체는 폴리아크릴레이트(polyacrylate), 폴리아크릴산나트륨(sodium polyacrylate)으로 이루어진 군에서 선택되는 1종 이상의 음이온 고분자전해질(polyelectrolyte)이며, 용매 흡수 전 약 0.1 내지 1cm의 평균 직경을 가지고 포어(pore) 사이즈가 1 내지 5nm인 것인, 다공성 이온 전하 결정체. - 삭제

- 상기 청구항 제1항의 핵산 분리용 다공성 이온 전하 결정체를 포함하는, 핵산 추출 튜브.
- 핵산을 포함하는 시료를 용해 완충액(Direct lysis buffer)과 핵산 추출 튜브에 투입하여 혼합하는 단계;
상기 혼합물에 상기 청구항 제1항의 다공성 이온 전하 결정체(Ion charge particle)를 투입하고 5분 내지 30분 동안 반응시키는 단계; 및
상기 반응을 완료한 이온 전하 결정체를 제거하고 핵산이 농축된 완충액을 얻는 단계를 포함하는, 핵산 추출방법. - 삭제

상기 혼합물에 상기 청구항 제1항의 다공성 이온 전하 결정체(Ion charge particle)를 투입하고 5분 내지 30분동안 반응시키는 단계;
상기 반응을 완료한 이온 전하 결정체를 제거하여 핵산이 농축된 완충액을 얻는 단계;
상기 핵산이 농축된 완충액에 PCR 혼합제를 투입하고 중합효소 연쇄반응(PCR)을 수행하는 단계; 및
상기 PCR 과정에서 또는 완료된 시료를 인-시츄(in-situ)로 검출하는 단계를 포함하는, 핵산 검출 방법.
