Abstract
Description
도 2는 실시예 2에서 설명되는 비아코어 기기에서 수행된 전형적인 결합 분석의 결과를 보여준다. 센서그램은 각각 고정된 이량체 Z 변이체 (Z05477, 서열 509) 상에 인간 C5 (hC5; 흑색 실선 곡선), 시노몰구스 C5 (cC5; 흑색 짧은-파선 곡선), 래트 C5 (rC5; 흑색 긴-파선 곡선), 인간 MG7 도메인 (hMG7; 회색 점선 곡선), 및 인간 이뮤노글로불린 G (hIgG; 회색 실선 곡선)의 주입에 의해 얻었다.
도 3은 선택된 성숙된 Z 변이체에 대해, 각각 hC5 및 rC5에 대한 ELISA에서의 반응을 보여주는 막대 그래프이다. 흑색 막대는 실시예 4에서 설명되는 바와 같이 각각의 Z 변이체에 대해 0.05 ㎍/ml hC5를 사용하여 얻은 450 nm에서의 흡광도 (각각의 군에서 좌측 막대) 및 4 ㎍/ml rC5를 사용하여 얻은 450 nm에서의 흡광도 (각각의 군에서 우측 막대)에 대응한다. Z 변이체 Z05363 (서열 510)에 대한 반응을 양성 대조군으로서 플로팅한다.

도 15는 실시예 10에서 설명되는 바와 같이 분석된, C5 결합 Z-ABD 융합 분자 (GS 링커에 의해 ABD094 (서열 759)에 융합된 Z06175a (서열 753)) 또는 OmCI (서열 761)과 조합된 염증유발 분자 지모산 (40 mg/kg i.p.) 및 그 단독 처리의 효과 (세척액 내의 C5a 농도)를 보여주는 도표이다. Z-ABD를 지모산을 사용한 유도 18 h 전에 20 nmol/kg (LD), 100 nmol/kg (MD) 및 500 nmol/kg (HD)으로 s.c. 투여하였다. OmCI (30 nmol/kg)를 지모산 처리 1 h 전에 i.p. 투여하고, 샘플을 지모산 유도 1 h 후에 채취하였다.
도 16a 및 16b는 실시예 11에서 설명되는 바와 같이 암컷 C57bl 마우스에게 500 nmol/kg의 기관내 투여 후에 Z-ABD (GS 링커에 의해 ABD094 (서열 759)에 융합된 Z06175a (서열 753))의 약동학적 프로파일을 보여준다. a) 각각의 동물의 혈청 농도 (각각의 시점에 대해 n=3, 총 27마리의 동물) 및 b) 1:5로 희석된 상기 혈청 샘플에 노출된 양 적혈구의 용혈.
Claims (32)
- (i) (a) 3개-나선 다발(bundle) 단백질 도메인의 일부를 형성하고 (b) 서열 1, 서열 2, 서열 3, 서열 4, 및 서열 5로부터 선택되는 아미노산 서열을 포함하는 C5 결합 모티프 BM, 및
(ii) 폴리펩티드의 생산, 정제, 생체내 또는 시험관내 안정화, 커플링 또는 검출을 개선하고, 시스테인 잔기, His6 태그, myc 태그, FLAG 태그, 및 반감기 연장 모이어티(moiety)로부터 선택되는, C 말단 및/또는 N 말단 아미노산
을 포함하는 C5 결합 폴리펩티드. - 제1항에 있어서, 서열 1 및 서열 4로부터 선택되는 아미노산 서열을 포함하는 C5 결합 폴리펩티드.
- 제1항에 있어서, 서열 1의 아미노산 서열을 포함하는 C5 결합 폴리펩티드.
- 제1항에 있어서, 서열 4의 아미노산 서열을 포함하는 C5 결합 폴리펩티드.
- 제1항에 있어서, 추가의 아미노산이 반감기 연장 모이어티를 포함하고, 반감기 연장 모이어티가 항체의 Fc 도메인인 C5 결합 폴리펩티드.
- 제1항에 있어서, 추가의 아미노산이 반감기 연장 모이어티를 포함하고, 반감기 연장 모이어티가 알부민 결합 도메인인 C5 결합 폴리펩티드.
- 제6항에 있어서, 알부민 결합 도메인이 스트렙토코쿠스(Streptococcus) 단백질 G의 알부민 결합 도메인인 C5 결합 폴리펩티드.
- 제7항에 있어서, 알부민 결합 도메인이 서열 759의 아미노산 서열을 포함하는 것인 C5 결합 폴리펩티드.
- 제3항에 있어서, 추가의 아미노산이 반감기 연장 모이어티를 포함하고, 반감기 연장 모이어티가 항체의 Fc 도메인인 C5 결합 폴리펩티드.
- 제3항에 있어서, 추가의 아미노산이 반감기 연장 모이어티를 포함하고, 반감기 연장 모이어티가 알부민 결합 도메인인 C5 결합 폴리펩티드.

- 제11항에 있어서, 알부민 결합 도메인이 서열 759의 아미노산 서열을 포함하는 것인 C5 결합 폴리펩티드.
- 제4항에 있어서, 추가의 아미노산이 반감기 연장 모이어티를 포함하고, 반감기 연장 모이어티가 항체의 Fc 도메인인 C5 결합 폴리펩티드.

- 제7항에 따른 C5 결합 폴리펩티드를 포함하고, 알부민 결합 도메인을 C5 결합 폴리펩티드의 C 또는 N 말단에 연결시키기 위한 연결 모이어티를 추가로 포함하는, C5 결합 화합물.

여기서, 서로 독립적으로, [CBP1]은 C5 결합 폴리펩티드이고, [L1]은 연결 모이어티이고, [ALBD]는 스트렙토코쿠스 단백질 G의 알부민 결합 도메인임. - 제17항에 있어서, 연결 모이어티가 G, GS; [G2S]n; [G3S]n; [G4S]n; GS[G4S]n; [S2G]m; [S3G]m; [S4G]m; 및 VDGS로부터 선택되는 것이고, 여기서 n이 0 내지 7이고 m이 0 내지 7인, C5 결합 화합물.
