Abstract
Description
도 2는 40℃ 및 75% 상대 습도에서의 8주 후의 모놀리식 정제 (비교), 본 발명의 이중층 정제, 삼중층 정제 (비교), 및 코비시스타트 단일 작용제 정제 (비교)에 대한 코비시스타트 불순물을 보여주는 그래프이다.
도 3은 코비시스타트의 산화적 분해를 도시한 개략도이다.
도 4는 아타자나비르 술페이트 및 코비시스타트 층을 함유하며, 불활성 부형제 층이 상기 두 층 사이에 개재되어 있는 것인, 비교 삼중층 정제의 내부를 보여주는 측면도이다.

Claims (47)
- HIV에 대해 유용한 2종의 항레트로바이러스 약물의 이중층 정제 제제이며, 상기 2종의 항레트로바이러스 약물이
a) 아타자나비르; 및
b) 코비시스타트가 이산화규소 담체 상에 담지된 형태의, 코비시스타트이고,
여기서 아타자나비르 및 코비시스타트가 서로 다른 층에 존재하고, 상기 정제가 불활성 부형제 층을 중간층으로서 갖는 삼중층 정제가 아닌 이중층 정제 제제. - 제1항에 있어서, 상기 아타자나비르가 아타자나비르 술페이트 형태인 이중층 정제 제제.
- 삭제
- 제2항에 있어서, 상기 이중층 정제가 시각적 균열을 보이지 않는 것인 이중층 정제 제제.
- 제4항에 있어서, 상기 이중층 정제가 유리 염기로서의 아타자나비르 술페이트 300 mg 및 코비시스타트 150 mg을 전달하는 것인 이중층 정제 제제.
- 제5항에 있어서, 스테아르산, 스테아르산마그네슘, 미세결정질 셀룰로스, 소듐 전분 글리콜레이트, 크로스포비돈, 히드록시프로필 셀룰로스, 및 크로스카르멜로스 소듐으로 이루어진 군으로부터 선택되는 1종 이상의 부형제를 추가로 포함하는 이중층 정제 제제.
- 제1항에 있어서, 상기 아타자나비르 및 상기 코비시스타트가 함께 상기 제제의 35-85% (w/w)를 구성하는 이중층 정제 제제.
- 제1항에 있어서, 제약상 허용되는 담체를 추가로 포함하며, 0.2% (w/w) 이하의 분해물 BMT-115982 또는 BMT-089290을 포함하는 이중층 정제 제제.
- 제1항에 있어서, 40℃ 및 75% 상대 습도에서 8주 경과 후 3% (w/w) 미만의 전체 코비시스타트 불순물을 갖거나, 또는 실온에서 보관되었을 때, 30℃/75%RH에서 보관되었을 때, 25℃/60%RH에서 보관되었을 때 또는 25℃/75%RH에서 보관되었을 때 12개월째에 2% (w/w) 미만의 코비시스타트 불순물을 갖는 이중층 정제 제제.
- 제9항에 있어서, 정제 프레스로부터의 방출 24시간 이내에 실온 조건하에서 균열되지 않는 이중층 정제 제제.
- 제9항에 있어서, 28-39 SCU 범위 이내의 경도 값을 갖는 이중층 정제 제제.
- 제1항, 제2항 및 제4항 내지 제11항 중 어느 한 항에 있어서, 환자에서 HIV 감염의 치료에 사용하기 위한 이중층 정제 제제.
- a) 별개로 아타자나비르를 1종 이상의 부형제와 함께 혼합하는 단계이며, 여기서 상기 부형제 중 적어도 1종이 미세결정질 셀룰로스이고, 여기서 상기 아타자나비르를 먼저 상기 1종 이상의 부형제의 제1 부분과 혼합하여 제1 아타자나비르 혼합물을 제조하고, 추가로, 이어서 상기 제1 혼합물을 물로 습식 과립화한 후, 습식 밀링하고, 크기별로 분류하고, 건조시키고, 이어서 상기 부형제의 제2 부분을 상기의 건조된 제1 혼합물과 혼합하여 제2 아타자나비르 혼합물을 제조하는 것인 단계;
b) 별개로 코비시스타트가 이산화규소 담체 상에 담지된 형태의 코비시스타트를 1종 이상의 부형제와 함께 혼합하는 단계이며, 여기서 상기 부형제 중 적어도 1종이 미세결정질 셀룰로스이고, 여기서 상기 코비시스타트를 먼저 상기 1종 이상의 부형제의 제1 부분과 혼합하여 제1 코비시스타트 혼합물을 제조하고, 이어서 상기 제1 혼합물을 롤러 압착시키고, 과립으로 밀링하고, 이어서 상기 부형제의 제2 부분을 상기의 과립화된 제1 혼합물과 혼합하여 제2 코비시스타트 혼합물을 제조하는 것인 단계; 및
c) 상기 제2 아타자나비르 혼합물 및 상기 제2 코비시스타트 혼합물을 이중층 정제로 압축시키는 단계
를 포함하는, 아타자나비르/코비시스타트 정제 제제를 제조하는 방법. - 제13항에 있어서, 단계 a)에서, 상기 부형제의 상기 제1 부분이 과립내 부형제이고, 상기 부형제의 상기 제2 부분이 과립외 부형제이고; 추가로, 단계 b)에서, 상기 부형제의 상기 제1 부분이 과립내 부형제이고, 상기 부형제의 상기 제2 부분이 과립외 부형제인 방법.
