Abstract
Description
도 2는 벼 유래 OsCP1 유전자가 포함된 재조합 벡터를 이용하여 벼 식물체의 형질전환 과정을 나타낸 사진이다. (A) 캘러스 유도 배지에서 벼 캘러스를 유도하는 단계, (B) 유도된 벼 캘러스에 재조합 벡터를 포함하는 아그로박테리움 튜머파시엔스(Agrobactrium tumefaciens) LBA4404를 공동배양하여 형질전환시키는 단계, (C) 형질전환된 벼 캘러스를 제초제 성분인 포스피노트리신(phosphinotricin)이 첨가된 배지에서 배양하는 단계, (D) 및 (E) 형질전환된 벼 캘러스로부터 재분화된 식물체(신초형성, 뿌리형성 및 신장).
도 3A는 벼(Oryza sativa), 애기장대(Arabidopsis thaliana) 및 배추(Brassica rapa) 유래 CP(cysteine proteinase) 단백질들간의 유연관계를 나타낸 모식도이고, 도 3B 및 도 3C는 CP 패밀리 단백질에 보존된 도메인(Peptidase_C1 superfamily) 및 상기 도메인의 아미노산 서열을 나타낸 그림이다.
도 4는 야생형 벼 식물체의 뿌리(root), 줄기(stem), 잎집(sheath), 잎(leaf), 지엽 잎(flag leaf), 이삭(panicle) 및 종자(grain)에서 OsCP1 유전자의 mRNA 발현량을 확인한 결과이다.
도 5는 OsCP1 유전자가 과발현된 벼 형질전환체로부터 단일 카피(single copy) 도입 형질전환체를 선발한 결과이다. HPT(hygromycin phosphotransferase): 선발 마커 유전자.
도 6은 OsCP1 유전자가 과발현된 벼 형질전환체의 유묘에 물(H2O), 벼 흰잎마름병균(Xanthomonas oryzae pv. oryzae, Xoo K3a), 살리실산(SA), 메틸 자스모네이트(meJA), 가뭄(5% soil moisture), 고온(42℃) 및 염분(NaCl)을 각각 처리한 후 OsCP1 유전자의 mRNA 발현량을 확인한 결과이다.
도 7은 염 스트레스에 대한 감수성 품종인 동진벼(S, WT), 저항성 품종인 석정벼(T) 및 본 발명의 OsCP1 과발현 벼 형질전환체(계통 1 및 2)에 염 스트레스를 처리하고 10일 후 식물체의 성장 상태를 나타낸 사진이다.

Claims (6)
- 서열번호 2의 아미노산 서열로 이루어진 벼 유래 OsCP1(Oryza sativa cysteine proteinase 1) 단백질을 코딩하는 유전자를 포함하는 재조합 벡터로 식물 세포를 형질전환하여 형질전환된 식물세포에서 OsCP1 유전자를 과발현시키는 단계를 포함하는 식물체의 염 스트레스 내성을 증가시키는 방법.
