도 2 내지 도 4는 탈세포화를 나타낸 실험결과이다.
도 5 내지 도 9는 탈세포화 과정에서 조직의 손상여부를 나타낸 실험결과이다.
Claims (11)
- 탈세포화 대상이 되는 포유류 조직을 준비하는 단계;
상기 조직에 포함된 세포를 제거하기 위하여, 양쪽성 계면활성제 및 류펩틴(leupeptin), 아프로티닌(aprotinin) 및 이들의 조합으로 이루어진 그룹에서 선택된 단백질분해 억제제를 포함하는 제1 탈세포액으로 상기 조직을 1차로 처리하는 제1 세포제거 단계; 및
상기 제1 세포제거 단계를 진행한 상기 조직에 포함된 DNA 및 RNA를 제거하기 위하여, DNA 분해효소 5 내지 15㎍/ml 및 RNA 분해효소 150 내지 250㎍/ml가 포함된 제2 탈세포액으로 상기 조직을 2차로 처리하는 제2 세포제거 단계;를 포함하는, 조직의 탈세포화 방법. - 삭제
- 제1 항에 있어서,
상기 양쪽성 계면활성제는 알킬디메틸아민 옥시드(alkyldimethylamine oxide), 알킬카르복시베타인(akylcarboxybetaine), 알킬아미드프로필베타인(alkylamidopropylbetaine), 알킬아미드프로필술포베타인(alkylamidopropylsulfobetaine), 알킬이미다졸베타인(alkylimidazoliumbetaine), 3-[(3-클라미드프로필)디메틸암모니오]-2-히드록시-1-프로판술폰산염(3-[(3-cholamidopropyl)dimethylammonio]-2-hydroxy-1-propanesulfonate, CHAPSO) 및 3-[(3-클라미드프로필)디메틸암모니오]-1-프로판술폰산염((3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate, CHAPS)에서 선택된 어느 하나 또는 이들의 조합인 조직의 탈세포화 방법. - 삭제
- 제1 항에 있어서,
상기 제1 탈세포액은 TNM(Tris NaCl MgCl2) 버퍼, PBS(Phosphate Buffered Saline) 버퍼, Tris-Cl 버퍼, SSC(Saline Sodium Citrate) 버퍼, HEN(Hepes EDTA Neocuproine) 버퍼, TEN(Tris EDTA NaCl) 버퍼 및 이들의 조합으로 이루어진 그룹에서 선택된 제1 버퍼액을 더 포함하는 조직의 탈세포화 방법. - 제1 항에 있어서,

상기 세척액은 증류수와 항생제를 포함하는 조직의 탈세포화 방법. - 제1 항에 있어서,
상기 제1 세포제거 단계와 상기 제2 세포제거 단계 사이에, 1차로 처리된 상기 조직을 식염수로 세척하는 제2 세척단계를 더 포함하는 조직의 탈세포화 방법.
