Abstract
Description
도 2는 본 명세서에 개시된 기술의 일 구현 예에 따른 세포배양검사 장치의 웰 유닛의 단면을 나타낸 것이다.
도 3은 제1 유체 및 제2 유체를 웰 유닛에 도입하여 항생제 감수성 검사를 하는 과정을 나타낸다.

Claims (14)
- 복수의 웰 유닛들을 구비한 세포배양검사 장치로서,
상기 웰 유닛은 고형화제를 함유한 액상 매질과 생물학적 물질의 혼합용액인 제1 유체 및 물, 세포배양용 배지, Dimethylformamide(DMF) 또는 Dimethyl sulfoxide (DMSO)를 포함하는 용매인 제2 유체를 주입하기 위한 입구부; 및
상기 제1 유체를 수용하기 위한 제1 서브웰 및 상기 제2 유체를 수용할 수 있으며, 항생제를 특정 위치에 적재하기 위한 제2 서브웰을 포함하는 바닥부를 포함하되,
상기 웰 유닛의 내벽의 둘레를 따라 상기 웰 유닛의 중심방향으로 돌출된 제1 단차부가 존재함으로써, 상기 웰 유닛이 상기 입구부 쪽의 상단부보다 좁은 내부 너비를 갖는 하단부를 가지며,
상기 제1 서브웰의 직경 및 상기 제2 서브웰의 직경은 상기 웰 유닛의 상기 바닥부 직경보다 작게 제작되어 제2 단차부를 형성하며,
상기 제1 단차부는 상기 제2 유체의 주입 시 상기 바닥부로부터 수직방향 전진을 막을 수 있도록 상기 제2 유체의 전진방향에 대해 90도 이상의 각도를 가지며,
상기 제2 단차부는 상기 제1 유체의 주입 시 상기 바닥부로부터 수직방향 전진을 막을 수 있도록 상기 제1 유체의 전진방향에 대해 90도 이상의 각도를 가지며,
상기 제1 서브웰 및 상기 제2 서브웰의 내부모서리는 곡률반경이 0.1~1mm이며, 높이가 0.1~1mm인 곡선형을 갖는 것인 세포배양검사 장치. - 삭제

상기 제1 단차부는 상기 내벽의 둘레를 따라 연결된 모서리의 형상을 갖는 것인 세포배양검사 장치. - 제1 항에 있어서,
상기 제1 단차부의 높이는 상기 바닥부의 내부 모서리로부터 상기 바닥부 중심까지의 거리보다 작은 것인 세포배양검사 장치. - 제4 항에 있어서,
상기 제1 단차부의 높이는 0.2mm 이상, 4mm 이하인 것인 세포배양검사 장치. - 제1 항에 있어서,
상기 바닥부와 유닛 내벽이 형성하는 바닥부의 내부모서리는 상기 내벽의 둘레를 따라 연결된 모서리의 형상을 갖는 것인 세포배양검사 장치. - 제6 항에 있어서,
상기 바닥부와 유닛 내벽이 형성하는 바닥부의 내부모서리는 바닥부를 기준으로 70~120°의 각도를 가지는 세포배양검사 장치. - 삭제

- 복수의 웰 유닛들이 정렬되어 형성된 어레이 구조를 가지는 복수의 웰 유닛들을 구비한 세포배양검사 장치를 이용한 세포의 분석 방법으로서,
상기 웰 유닛은 고형화제를 함유한 액상 매질과 생물학적 물질의 혼합용액인 제1 유체 및 물, 세포배양용 배지, Dimethylformamide(DMF) 또는 Dimethyl sulfoxide (DMSO)를 포함하는 용매인 제2 유체를 주입하기 위한 입구부; 및 상기 제1 유체를 수용하기 위한 제1 서브웰과 상기 제2 유체를 수용할 수 있으며 항생제를 특정 위치에 적재하기 위한 제2 서브웰을 포함하는 바닥부; 상기 배양액인 제2 유체의 주입 시 상기 바닥부로부터 수직방향 전진을 막을 수 있도록 상기 제2 유체의 전진방향에 대해 90도 이상의 각도를 갖는 제1단차부; 상기 제1서브웰의 직경 및 상기 제2 서브웰의 직경은 상기 웰 유닛의 상기 바닥부 직경보다 작게 제작되며, 상기 제1 유체의 주입 시 상기 바닥부로부터 수직방향 전진을 막을 수 있도록 상기 제1 유체의 전진방향에 대해 90도 이상의 각도로 형성되는 제2 단차부; 상기 웰 유닛의 내벽의 둘레를 따라 상기 웰 유닛의 중심방향으로 돌출된 제1 단차부가 존재함으로써, 상기 웰 유닛이 상기 입구부 쪽의 상단부보다 좁은 내부 너비를 갖는 하단부;를 포함하며,
상기 제1 서브웰 및 상기 제2 서브웰의 내부모서리는 곡률반경이 0.1~1mm이며, 높이가 0.1~1mm인 곡선형을 형성하며,
(a) 상기 제1 유체를 상기 제1 서브웰에 주입하고, 상기 혼합용액을 고화하여 고체 박막을 형성하는 단계;
(b) 상기 제2 유체를 상기 제2 서브웰에 투입하여 항생제를 분산 또는 용해하며, 상기 제2 유체를 상기 제2 서브웰 상부의 상기 웰 유닛에 설치된 상기 제1 단차부까지 투입하여 상기 제1 서브웰 내부의 상기 제1 유체의 상기 고체 박막과 접촉시키는 것으로 상기 항생제를 상기 고체 박막으로 확산시키는 단계; 및
(c) 상기 제1 유체의 상기 고체 박막과 상기 제2 유체가 서로 만나 형성된 계면에서 일어나는 생물학적 물질의 변화를 단일세포 단위로 관찰하는 단계;를 포함하는 세포의 분석 방법. - 제13 항에 있어서,
(d) 상기 항생제에 대한 생물학적 물질의 변화를 단일 세포 단위로 관찰하여 최소 억제 농도(Minimum Inhibitory Concentration, MIC) 또는 최소 바이오필름 박멸 농도(Minimum Biofilm Eradication Concentration, MBEC)를 측정하는 단계를 더 포함하는 세포의 분석 방법.
