Claims (13)
- 음전하를 띠는 다공성의 제1 종이기판;
상기 제1 종이기판의 일 단면에 적층되고, 세파로스 및 유리비드의 혼합물 을 포함하는 분리층; 및
상기 분리층의 일 단면에 적층되고, 음전하를 띠는 다공성 제2 종이기판;으로 구성된 종이기반 핵산검출용 센서.
- 제1항에 있어서,
상기 제1 종이기판 및 제2 종이기판은 동일하거나 서로 상이한 크기의 기공을 가지는 것을 특징으로 하는 종이기반 핵산검출용 센서.
- 제2항에 있어서,
상기 기공의 크기는 1 내지 5 ㎛인 것을 특징으로 하는 종이기반 핵산검출용 센서.
- 제1항에 있어서,
상기 제1 종이기판 및 제2 종이기판은 서로 동일하거나 상이한 재질이고, 각각 독립적으로 glass fiber Millipore G041, Millipore GFDX, cellulosic Millipore C083, Millipore C048, Millipore C068, Millipore C083, Millipore C248, Healthcare CF1, CF3, CF4 cotton linter, Whatman Fusion 5, Std 14 및 Std 15로 이루어진 군 중에서 선택되는 어느 하나인 것을 특징으로 하는 종이기반 핵산검출용 센서.
- 제1항에 있어서,
상기 제1 종이기판의 전체 면적보다 제2 종이기판의 전체 면적이 2 배 내지 10 배 더 작은 것을 특징으로 하는 종이기반 핵산검출용 센서.
- 제1항에 있어서,
상기 분리층의 두께는 10 내지 100 ㎛인 것을 특징으로 하는 종이기반 핵산검출용 센서.
- a) 아비딘(avidin), 스트렙타비딘(streptavidin), 프라이머에 결합된 항원에 항원-항체 반응을 통해 결합될 수 있는 항체, 프라이머에 결합된 표적분자에 특이적으로 결합하는 앱타머(aptamer) 및 프라이머에 결합된 올리고뉴클레오티드에 상보적인 올리고뉴클레오티드로 이루어진 군에서 선택되는 어느 하나로 코팅 처리된 포획 나노입자(capture nanoparticle)가 분산된 제1 용액; 및
b) 제1항 내지 제5항 중 어느 한 항의 종이 기반 핵산 검출용 센서;를 포함하는 종이기반 핵산검출용 키트.
- 제7항에 있어서,
상기 나노입자는 자성입자(magnetic bead), 금(Au) 나노입자, 은(Ag) 나노입자, 백금(Pt) 나노입자, 양자점(Quaantum dot), 상방전환 나노입자(upconversion nanoparticle, UCNP) 그래핀(graphene)-나노입자 복합체, 색 염색 나노입자(color dyed particles) 및 라텍스(latex) 나노입자로 이루어진 군으로부터 선택되는 종이기반 핵산검출용 키트.
- 제7항에 있어서,
상기 나노입자는 스트렙타비딘(streptavidin)이 코팅된 자성비드인 것을 특징으로 하는 종이기반 핵산검출용 키트.
- 제7항에 있어서,
a-1) 검출하고자 하는 표적 핵산에 특이적으로 결합하는, 프라이머 세트(primer set)를 더 포함하고,
상기 프라이머 세트는 아비딘(avidin), 비오틴(biotin), 아비딘-비오틴, 항원, 표적분자, 올리고뉴클레오티드로 이루어진 군에서 선택되는 어느 하나로 표지된 정방향 프라이머(forward primer), 및 형광 표지된 역방향 프라이머(reverse primer)를 포함하는 것을 특징으로 하는 종이기반 핵산검출용 키트
- (a) 제7항에 따른 종이기반 핵산검출용 키트의 제1 용액과 아비딘(avidin), 비오틴(biotin), 아비딘-비오틴, 항원, 표적분자, 올리고뉴클레오티드로 이루어진 군에서 선택되는 어느 하나로 표지된 PCR 증폭산물을 혼합하여 반응물을 수득하는 단계; 및
(b) 제7항에 따른 종이기반 핵산검출용 키트의 센서에 상기 반응물을 적하하는 단계;를 포함하는 PCR 증폭산물을 분석하기 위한 방법.
- 제11항에 있어서,
상기 (b) 단계 후에, (c) 상기 센서의 제1 종이기판 표면 상에 퍼진 반응물의 범위를 통해 PCR 증폭산물 내에 표적 핵산이 포함되어 있었는지 여부를 판별하는 단계를 더 포함하는 것을 특징으로 하는, 방법.
- 제11항에 있어서,
a) 단계는
a-1) 검체로부터 분리된 생물학적 시료로부터 핵산을 추출하는 단계; 및
a-2) 목적 핵산에 특이적으로 결합하는, 아비딘(avidin), 비오틴(biotin), 아비딘-비오틴, 항원, 표적분자 및 올리고뉴클레오티드로 이루어진 군에서 선택되는 어느 하나로 표지된 정방향 프라이머(forward primer), 및 Cy3, Cy5, TAMRA, TEX, TYE, HEX, FAM, TET, JOE, MAX, ROX, VIC, Cy3.5, Texas Red, Cy5.5, BHQ, Iowa Black RQ 및 IRDye로 이루어지는 군에서 선택되는 어느 하나로 표지된 역방향 프라이머(reverse primer)를 포함하는 프라이머 세트를 처리하여 목적 핵산이 증폭된 PCR 증폭산물을 수득하는 단계;를 포함하는 것을 특징으로 하는, 방법.
