Abstract
Description

Claims (125)
- 서열번호 16 또는 14와 적어도 98% 동일하고 서열번호 16 또는 14의 220개 이상의 인접한 아미노산인, 단리된 아미노아실-tRNA 신테타아제 (AARS) 폴리펩타이드 또는 이의 단편을 포함하는, 염증성 장애, 고콜레스테롤혈증, 고지혈증, 1형 및 2형 당뇨병, 대사 증후군, 또는 혈관 질환의 치료를 위한 치료 조성물로서,
상기 폴리펩타이드는 세포외 신호전달 활성을 가지고 적어도 5 mg/mL의 용해도를 가지며, 상기 조성물은 단백질 기준으로 적어도 95%의 순도 및 단백질 mg 당 10EU 미만의 내독소를 갖는, 치료 조성물. - 제1항에 있어서, 상기 AARS 폴리펩타이드는 서열번호 16 또는 14로 이루어지거나, 세포외 신호전달 활성을 보유하면서 1, 2, 3, 4, 또는 5개의 아미노산의 치환, 결실, 부가 또는 이들의 조합에 의해 서열번호 16 또는 14와 상이한 것인, 치료 조성물.
- 제1항에 있어서, 상기 AARS 폴리펩타이드는 이종 폴리펩타이드에 융합된 것인, 치료 조성물.
- 제3항에 있어서, 상기 이종 폴리펩타이드는 정제 태그, 에피토프 태그, 표적 서열, 신호 펩타이드, 막 전좌 서열, 및 PK 수식인자로 이루어진 그룹으로부터 선택된 것인, 치료 조성물.
- 세포외 신호전달 활성을 가지는, 서열번호 16 또는 14와 적어도 98% 동일하고 서열번호 16 또는 14의 220개 이상의 인접한 아미노산인, 단리된 아미노아실-tRNA 신테타아제 (AARS) 폴리펩타이드 또는 이의 단편.
- 제5항에 있어서, 상기 AARS 폴리펩타이드는 서열번호 16 또는 14로 이루어지거나, 세포외 신호전달 활성을 보유하면서 1, 2, 3, 4, 또는 5개의 아미노산의 치환, 결실, 부가 또는 이들의 조합에 의해 서열번호 16 또는 14와 상이한 것인, 단리된 AARS 폴리펩타이드.
- 제5항에 있어서, 상기 AARS 폴리펩타이드는 이종 폴리펩타이드에 융합된 것인, 단리된 AARS 폴리펩타이드.
- 제7항에 있어서, 상기 이종 폴리펩타이드는 정제 태그, 에피토프 태그, 표적 서열, 신호 펩타이드, 막 전좌 서열, 및 PK 수식인자로 이루어진 그룹으로부터 선택된 것인, AARS 폴리펩타이드.
- 제5항 내지 제8항 중 어느 한 항의 단리된 아미노아실-tRNA 신테타아제 (AARS) 폴리펩타이드를 포함하는, 염증성 장애, 고콜레스테롤혈증, 고지혈증, 1형 및 2형 당뇨병, 대사 증후군, 또는 혈관 질환의 치료를 위한 조성물로서,
상기 폴리펩타이드는 실온 또는 37℃의 물 또는 생리학적 pH의 생리학적 완충액에서 적어도 5mg/mL의 용해도를 가지며, 상기 조성물은 단백질 기준으로 적어도 95%의 순도 및 단백질 mg 당 10 EU 미만의 내독소를 가지는, 조성물. - 제9항에 있어서, 상기 폴리펩타이드에 적어도 하나의 모이어티 또는 고형 기질이 공유결합 또는 비공유결합으로 부착된 것인, 조성물.
- 제5항 내지 제8항 중 어느 한 항의 아미노아실-tRNA 신테타아제 (AARS) 폴리펩타이드, 및 적어도 하나의 세포가 상기 AARS 폴리펩타이드를 인코딩하는 폴리뉴클레오타이드를 포함하는, 무혈청 배지에서 성장할 수 있는 세포의 제작된 군집을 포함하는, 세포 조성물.

- 제5항 내지 제8항 중 어느 한 항의 아미노아실-tRNA 신테타아제 (AARS) 폴리펩타이드에 특이적으로 결합하는 화합물을 확인하는 방법으로서, 상기 방법은,
a) 적당한 조건 하에서 AARS 폴리펩타이드를 적어도 하나의 시험 화합물과 조합하는 단계, 및
b) 시험 화합물에 대한 AARS 폴리펩타이드의 결합을 검출하고, 이로써 AARS 폴리펩타이드에 특이적으로 결합하는 화합물을 확인하는 단계를 포함하는, 방법. - 제5항 내지 제8항 중 어느 한 항의 AARS 폴리펩타이드 및 약제학적으로 허용가능한 담체를 포함하는, 염증성 장애, 고콜레스테롤혈증, 고지혈증, 1형 및 2형 당뇨병, 대사 증후군, 또는 혈관 질환의 치료를 위한 약제학적 조성물.
