Abstract
Description
도 2는 도 1에서 나타내어진 제 1 불용성 담체(면역크로마토그래프용 스트립)의 일형태의 종단면을 나타내는 모식도이다.
도 1 및 도 2에 있어서 1은 백 점착 시트, 2는 표식물질 유지 패드, 3은 크로마토그래프 담체(항체 고정화 멤브레인), 3a는 포착 부위, 31은 검출 부위, 32는 컨트롤 부위, 4는 흡수 패드, 5는 시료 첨가 패드, 10은 면역크로마토그래프용 스트립을 나타낸다. 피검물질을 함유하는 피검시료가 크로마토그래프 담체 상을 전개하는 방향을 화살표 A로 나타낸다. 본 발명에 있어서는 화살표 A의 근원 방향을 상류, 화살표 A의 선단 방향을 하류라고 정의한다.
도 3은 본 발명에서 사용할 수 있는 면역크로마토그래프 키트의 일형태를 모식적으로 나타낸다. 51은 도 1 및 도 2에서 나타낸 제 1 불용성 담체(면역크로마토그래프용 스트립), 52는 액체 저장 포드, 53은 제 2 불용성 담체(세정액 첨가 패드), 54는 제 3 불용성 담체, 55는 제 1 디바이스 부품, 56은 제 2 디바이스 부품, 57은 점착 구멍을 나타낸다.
Claims (11)
- 피검물질과,
상기 피검물질에 대한 제 1 결합물질로 수식한 금속을 포함하는 표식물질의 복합체를 형성시킨 상태에서 상기 복합체를 단백질의 프로테아제 분해물의 존재 하에서 불용성 담체 상에 전개하고,
상기 피검물질에 대한 제 2 결합물질, 또는 상기 피검물질에 대한 상기 제 1 결합물질로의 결합성을 갖는 물질을 포함한 불용성 담체 상의 검출 부위에 있어서 상기 피검물질과 상기 표식물질을 포착하며,
포착한 상기 표식물질을 증폭시켜서 상기 피검물질을 검출하는 것을 포함하는 면역크로마트그래프 방법으로서,
상기 단백질의 프로테아제 분해물은 산성 조건 하에서 불용 성분으로서의 석출물이 존재하지 않는 분해물인 것을 특징으로 하는 면역크로마토그래프 방법. - 제 1 항에 있어서,
상기 단백질의 프로테아제 분해물의 분자량의 범위는 0.1kD∼15kD인 것을 특징으로 하는 면역크로마토그래프 방법. - 삭제
- 제 1 항 또는 제 2 항에 있어서,
상기 단백질의 프로테아제 분해물은 상기 단백질의 프로테아제 분해물에 산을 첨가해서 pH를 4 이하로 하고, 석출된 석출물을 제거함으로써 얻어지는 단백질 분해물인 것을 특징으로 하는 면역크로마토그래프 방법. - 제 1 항 또는 제 2 항에 있어서,
상기 단백질의 프로테아제 분해물은 카제인의 프로테아제 분해물인 것을 특징으로 하는 면역크로마토그래프 방법. - 제 5 항에 있어서,
상기 카제인은 αS1 카제인, αS2 카제인, β 카제인, 또는 κ 카제인으로부터 선택되는 1개 이상의 카제인인 것을 특징으로 하는 면역크로마토그래프 방법. - 제 1 항 또는 제 2 항에 있어서,
상기 제 1 결합물질 및 제 2 결합물질 중 적어도 어느 한쪽이 항체인 것을 특징으로 하는 면역크로마토그래프 방법. - 제 1 항 또는 제 2 항에 있어서,
상기 포착한 표식물질을 증폭시키는 공정을, 은을 포함하는 화합물 및 은이온을 위한 환원제를 포함하는 증폭액을 이용하여 행하는 것을 특징으로 하는 면역크로마토그래프 방법. - 제 1 항 또는 제 2 항에 있어서,
상기 피검물질과, 상기 피검물질에 대한 상기 제 1 결합물질로 수식한 금속을 포함하는 상기 표식물질의 복합체를 형성시킨 상태에서,
상기 복합체를 상기 단백질의 프로테아제 분해물의 존재 하에서 불용성 담체 상에 전개한 후에 세정액을 불용성 담체 상에 전개하고,
그 후에 상기 포착한 표식물질을 증폭시키는 것을 특징으로 하는 면역크로마토그래프 방법. - 피검물질에 대한 제 1 결합물질로 수식한 금속을 포함하는 표식물질과,
상기 피검물질에 대한 제 2 결합물질, 또는 상기 피검물질에 대한 상기 제 1 결합물질로의 결합성을 갖는 물질을 포함한 불용성 담체와,
단백질의 프로테아제 분해물을 구비하고,
상기 단백질의 프로테아제 분해물은 산성 조건 하에서 불용 성분으로서의 석출물이 존재하지 않는 분해물인 것을 특징으로 하는 면역크로마토그래프용 키트. - 제 10 항에 있어서,
상기 제 1 결합물질 및 제 2 결합물질 중 적어도 어느 한쪽이 항체인 것을 특징으로 하는 면역크로마토그래프용 키트.
