Abstract
(1) 시료 중의 FGF-23 을, 수성 배지 중에서, 자성 입자, FGF-23 에 결합하는 제 1 항체 또는 이의 단편, 및 FGF-23 에 결합하는 제 2 항체 또는 이의 단편과 반응시켜, 자성 입자 상에 FGF-23 에 결합하는 제 1 항체 또는 이의 단편, FGF-23, 및 FGF-23 에 결합하는 제 2 항체 또는 이의 단편을 포함하는 면역복합체를 생성시키는 단계;
(2) 단계 (1) 후 반응 혼합물 중의 자성 입자를 자력에 의해 수집하고, 자력에 의해 수집된 자성 입자와 그 이외의 성분을 분리하는 단계; 및
(3) 단계 (2) 에서 분리된 자성 입자 상의 면역복합체를 검정하는 단계.
이에 의해, 고감도이며 측정 범위가 넓은, 시료 중의 FGF-23 의 검정 방법이 개시된다.
Description
도 2 는 실시예 2 의 FGF-23 의 측정 방법의 측정 범위를 나타내는 그래프를 보여주며, 시료 중의 FGF-23 농도와 발광 수준 간의 관계를 보여준다. 가로축은 FGF-23 농도 (pg/mL) 를 나타내며, 세로축은 발광 수준 (RLU) 을 나타낸다.
도 3 은 투석 환자-유래 혈청 (환자 혈청 D) 을 사용하는, 실시예 3 의 측정 방법에 있어서의 희석 직선성을 나타내는 그래프를 보여주며, 희석률과 FGF-23 농도 간의 관계를 보여준다. 가로축은 상기 혈청의 희석률을 나타내며, 세로축은 측정된 FGF-23 농도 (pg/mL) 를 나타낸다.
도 4 는 투석 환자-유래 혈청 (환자 혈청 E) 을 사용하는, 실시예 3 의 측정 방법에 있어서의 희석 직선성을 나타내는 그래프를 보여주며, 희석률과 FGF-23 농도 간의 관계를 보여준다. 가로축은 상기 혈청의 희석률을 나타내며, 세로축은 측정된 FGF-23 농도 (pg/mL) 를 나타낸다.
도 5 는 플레이트를 사용하는 비교예 1 의 FGF-23 측정 방법을 사용하여 최소 측정가능 농도를 나타내는 그래프를 보여주며, 시료 중의 FGF-23 농도와 흡광도 간의 관계를 보여준다. 가로축은 FGF-23 농도 (pg/mL) 를 나타내고, 세로축은 흡광도 (Abs) 를 나타낸다. "I" 표시는 평균치 ± 2 SD 의 범위를 나타낸다. 또한, 점선은 블랭크 + 2 SD 의 흡광도를 나타낸다.
도 6 은 플레이트를 사용하는 비교예 2 의 FGF-23 측정 방법의 측정 범위를 나타내는 그래프를 보여주며, 시료 중의 FGF-23 농도와 흡광도 간의 관계를 보여준다. 가로축은 FGF-23 농도 (pg/mL) 를 나타내며, 세로축은 흡광도 (Abs) 를 나타낸다.
Claims (12)
- 하기 단계를 포함하는, 시료 중의 섬유아세포 증식 인자-23 (이하, FGF-23 으로 표시함) 의 측정 방법:
(1) 시료 중의 FGF-23 을, 수성 배지 중에서, 자성 입자, FGF-23 에 결합하는 제 1 항체 또는 이의 단편, 및 FGF-23 에 결합하는 제 2 항체 또는 이의 단편과 반응시켜, 자성 입자 상에 FGF-23 에 결합하는 제 1 항체 또는 이의 단편, FGF-23, 및 FGF-23 에 결합하는 제 2 항체 또는 이의 단편을 포함하는 면역복합체를 형성시키는 단계;
(2) 단계 (1) 후 반응 혼합물 중의 자성 입자를 자력에 의해 수집하고, 자력에 의해 수집된 자성 입자와 그 이외의 성분을 분리하는 단계; 및
(3) 단계 (2) 에서 분리된 자성 입자 상의 면역복합체를 측정하는 단계. - 제 1 항에 있어서, 제 2 항체가 표지 항체인 방법.
- 제 2 항에 있어서, 표지 항체가 알칼리 포스파타아제-표지 항체인 방법.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 단계 (3) 에서의 자성 입자 상 면역복합체의 측정이 화학발광의 측정에 의해 실행되는 방법.
- 제 1 항 내지 제 3 항 중 어느 한 항에 있어서, 시료가 혈청 또는 혈장인 방법.
- 제 4 항에 있어서, 시료가 혈청 또는 혈장인 방법.
- 제 1 항에 기재된 측정 방법에 사용되는, 시료 중의 섬유아세포 증식 인자-23 (이하, FGF-23 으로 표시함) 측정 시약으로서, 자성 입자, FGF-23 에 결합하고 또한 자성 입자에 결합하고 있지 않은 제 1 항체 또는 이의 단편, 및 FGF-23 에 결합하는 제 2 항체 또는 이의 단편을 포함하는, 시료 중의 FGF-23 측정 시약.
- 제 7 항에 있어서, 제 2 항체가 표지 항체인 시약.
- 제 8 항에 있어서, 표지 항체가 알칼리 포스파타아제-표지 항체인 시약.
- 제 7 항 내지 제 9 항 중 어느 한 항에 있어서, 화학발광 측정 시약을 추가로 포함하는 시약.
- 제 7 항 내지 제 9 항 중 어느 한 항에 있어서, 시료가 혈청 또는 혈장인 시약.
- 제 10 항에 있어서, 시료가 혈청 또는 혈장인 시약.
