Abstract
Description
Claims (16)
- 콘드로이틴을 생산하는 재조합 미생물로서,
상기 콘드로이틴이, β1-3(GlcUA → GalNAc) 결합 및 β1-4(GalNAc → GlcUA) 결합에 의해 결합된 D-글루쿠론산(GlcUA) 잔기 및 N-아세틸-D-갈락토사민(GalNAc) 잔기를 교대로 함으로써 구성된 선형 글리코사미노글리칸으로 정의되며,
상기 미생물은 K 항원 그룹에 속하는 미생물로부터 유래되고, 상기 미생물에서, 선형 콘드로이틴 골격에 대한 프럭토스 잔기의 첨가를 담당하는 효소를 암호화하는 원래 존재하는 kfoE 유전자가, 상기 유전자의 전부 또는 일부의 결실 또는 치환, 또는 추가의 뉴클레오타이드 서열의 삽입에 의한 파괴에 의해 불활성화되고,
상기 kfoE 유전자가
(A) 서열번호 2의 아미노산 서열을 포함하는 단백질; 및
(B) 하나 이상의 아미노산의 결실, 치환 또는 삽입에 의해 변형된 서열번호 2의 아미노산 서열을 포함하고 프럭토실-트랜스퍼라제 활성을 갖는 단백질
로 이루어진 그룹으로부터 선택된 단백질을 암호화하는, 콘드로이틴을 생산하는 재조합 미생물. - 제1항에 있어서, 상기 불활성화된 kfoE 유전자가,
(a) 서열번호 1의 뉴클레오타이드 서열을 포함하는 DNA인, 콘드로이틴을 생산하는 재조합 미생물. - 제1항 또는 제2항에 있어서, 상기 kfoE 유전자가 이의 전부 또는 일부를 카나마이신 내성 카세트로 치환하고 이를 후속적으로 제거하여 kfoE 유전자의 전부 또는 일부를 결실시킴으로써 불활성화되는, 콘드로이틴을 생산하는 재조합 미생물.
- 제1항 또는 제2항에 있어서, 상기 미생물이, 에스케리키아(Escherichia), 헤모필러스(Haemophilus), 캄필로박터(Campylobacter), 글로에오캅사(Gloeocapsa) 및 비브리오(Vibrio)로부터 선택된 속에 속하는 박테리아로부터 유래되는, 콘드로이틴을 생산하는 재조합 미생물.
- 제4항에 있어서, 상기 미생물이, 에스케리키아 콜라이 종에 속하는 박테리아로부터 유래되는, 콘드로이틴을 생산하는 재조합 미생물.
- 제5항에 있어서, 상기 미생물이, K 항원의 그룹 2에 속하는 에스케리키아 콜라이 종의 혈청형으로부터 유래되는, 콘드로이틴을 생산하는 재조합 미생물.
- 제6항에 있어서, 상기 미생물이 에스케리키아 콜라이 05:K4:H4U1-41 균주로부터 유래되는, 콘드로이틴을 생산하는 재조합 미생물.
- 제1항 또는 제2항에 있어서, 상기 미생물이 에스케리키아 콜라이 DSM23578 또는 에스케리키아 콜라이 DSM23644인, 콘드로이틴을 생산하는 재조합 미생물.
- 제8항에 있어서, 상기 미생물이 에스케리키아 콜라이 DSM23644인, 콘드로이틴을 생산하는 재조합 미생물.
- 콘드로이틴의 생물공학적 생산방법으로서,
에스케리키아 콜라이 DSM23578 및 에스케리키아 콜라이 DSM23644로부터 선택된 재조합 미생물이 사용되는, 콘드로이틴의 생물공학적 생산방법. - 제10항에 있어서,
상기 재조합 미생물이 에스케리키아 콜라이 DSM23644인, 콘드로이틴의 생물공학적 생산방법. - 콘드로이틴의 생물공학적 생산방법으로서,
(a) 제1항 또는 제2항의 재조합 미생물을 적합한 배지에서 배양하는 단계; 및
(b) 상기 미생물 배양액에 존재하는 콘드로이틴을 회수하고 정제하는 단계를 포함하는, 콘드로이틴의 생물공학적 생산방법. - 제12항에 있어서, 상기 재조합 미생물이 에스케리키아 콜라이 DSM23578 또는 에스케리키아 콜라이 DSM23644인, 콘드로이틴의 생물공학적 생산방법.
- 제13항에 있어서, 상기 재조합 미생물이 에스케리키아 콜라이 DSM23644인, 콘드로이틴의 생물공학적 생산방법.
