Description
Claims (13)
- 적어도 하나의 항체를 제1 항원 결정기 및 제2 항원 결정기와 접촉시키는 단계를 포함하고, 여기서 적어도 하나의 항체는 순차적으로 제1 항원 결정기를 포함하는 제1 면역원성 조성물에 의해 유도되는 제1 면역학적 시험감염에 노출시킨 후에 제2 항원 결정기를 포함하는 제2 면역원성 조성물에 의해 유도되는 제2 면역학적 시험감염에 노출시킨, 인간을 제외한 동물로부터 수득된 것이고, 여기서 적어도 하나의 항체가 제1 및 제2 항원 결정기에 결합하였다는 것은 교차-반응성을 시사하는 것이고, 이로써 교차-반응성을 갖는 항원 결정기를 포함하는 분자가 결정되는 것인, 교차-반응성을 갖는 항원 결정기를 포함하는 분자를 결정하는 방법.
- 제1항에 있어서, 제1 면역원성 조성물이 추가로 제1 항원 결정기를 포함하는 제1 폴리펩티드를 포함하고, 제2 면역원성 조성물이 추가로 제2 항원 결정기를 포함하는 제2 폴리펩티드를 포함하고, 여기서 제1 폴리펩티드는 제1 미생물에 의해 발현되고, 제2 폴리펩티드는 제2 미생물에 의해 발현되고, 여기서 제1 및 제2 미생물은 동일 종의 다른 혈청형인 것을 특징으로 하는 것인 방법.
- 제2항에 있어서, 제1 및 제2 미생물이 세균인 방법.
- 제3항에 있어서, 세균이 H. 파라수이스(H. parasuis)인 방법.
- 제4항에 있어서, 제1 미생물이 혈청형 5이고, 제2 미생물이 혈청형 13인 방법.
- 제4항에 있어서, 제1 및 제2 폴리펩티드가 각각 헴-결합 단백질 또는 ABC형 수송체인 방법.
- a) 인간을 제외한 동물에서 기억 B 세포를 활성화시켜 적어도 하나의 항체를 생산하고, 여기서 활성화시킨다는 것은 기억 B 세포를 활성화시키는 면역학적 반응을 유도하는 제1 분자를 사용하여 상기 동물을 면역학적으로 시험감염시키는 것을 포함하는 것인 단계; 및
b) 적어도 하나의 항체를 제2 분자와 접촉시키고, 여기서 적어도 하나의 항체가 제1 및 제2 분자와 결합하였다면 제2 분자는 교차-반응성을 갖는 항원 결정기를 포함하는 것으로 결정되는 것인 단계
를 포함하고, 여기서 제1 분자 또는 제2 분자는
로 이루어진 군으로부터 선택되는 서열을 포함하는 것인, 교차-반응성을 갖는 항원 결정기를 포함하는 분자를 결정하는 방법. - 제8항에 있어서, 기억 B 세포를 활성화시킨 후에 이를 시험관내에서 배양하는 단계를 더 포함하는 방법.
- 예방학적 또는 치료학적 유효량의,
(i)
로 이루어진 군으로부터 선택되는 아미노산 서열을 포함하는 단리된 폴리펩티드; 및
(ii) 제약상 허용되는 비히클, 담체 또는 부형제
를 포함하고, 여기서 상기 단리된 폴리펩티드는 H. 파라수이스의 2개 이상의 혈청형에 의해 발현되는 단백질에 존재하는 교차-반응성을 갖는 항원 결정기를 추가로 포함하며,
상기 단리된 폴리펩티드는 N-숙시디미딜-3-(2-피리딜디티오)프로피오네이트 (SPDP), 6-말레이미도카프로산의 반응성 에스테르, 2-브로모아세트산의 반응성 에스테르, 2-요오도아세트산의 반응성 에스테르, 4-(N-말레이미도-메틸)사이클로헥산-1-카르복실산의 반응성 에스테르, 시스테인 잔기, 및 N-말단 His 태그로 이루어진 군으로부터 선택되는 캐리어 물질에 결합되어 있거나, 또는 상기 단리된 폴리펩티드는 펩티드의 유리 아미노기로 형성된 산 부가 염 또는 펩티드의 유리 카르복실기로 형성된 염기 부가 염인, 백신. - 제10항에 있어서, 동물의 H. 파라수이스 감염과 관련된 질환, 병태 또는 그의 증상을 치료 또는 예방하기 위한 백신.
