도 2는 탈세포화된 바이오 스캐폴드의 화학적 특성을 나타낸 도이다 (A: DNA 함량; B: 콜라겐, GAG 및 엘라스틴 함량)
도 3은 본 발명의 재세포화된 바이오 스캐폴드의 세포의 부착, 증식 및 기능유지 확인한 결과이다 (A: 본 발명의 재세포화를 위한 관류 시스템의 모식도; B: 재세포화된 바이오 스캐폴드의 조직 검사; C: 재세포화된 바이오 스캐폴드의 면역 염색 결과; 항-Ki 67 항체 (적색); 항-알부민 항체 (녹색); DAPI (청색)).
도 4는 본 발명의 재세포화된 바이오 스캐폴드의 특성을 평가한 도이다 (A: 재세포화된 바이오 스캐폴드의 알부민 생산능 확인; B 및 C: 대사 및 종양관련 유전자 발현양 확인; D: MTX (methotrexate)의 처리에 따른 재세포화된 바이오 스캐폴드의 저항성 확인)
Claims (10)
- (1) 계면활성제를 인간을 제외한 동물의 간에 관류시켜 간을 탈세포화 하는 단계; 및
(2) 상기 탈세포화된 간에 간암세포를 관류시켜 간을 재세포화 하는 단계;를 포함하는 재세포화된 바이오 스캐폴드의 제조방법. - 제 1항에 있어서, 상기 계면활성제는 SDS (sodium dodecyl sulfate), Triton-X, DNase 및 CHAPS (3-(3-cholamidopropyl)dimethylammonio)-1-propane sulfonate)로 이루어진 군 중에서 선택된 1 종 이상인 것을 특징으로 하는, 재세포화된 바이오 스캐폴드의 제조방법.
- 제 1항에 있어서, 상기 계면활성제가 SDS인 경우, 0.01 내지 0.5 % (v/v)의 SDS를 장기에 관류시키는 것을 특징으로 하는, 재세포화된 바이오 스캐폴드의 제조방법.
- 제 1항에 있어서, 상기 관류는 계면활성제를 1 내지 100 mL/분의 유속으로 관류시키는 것을 특징으로 하는, 재세포화된 바이오 스캐폴드의 제조방법.
- 제 1항에 있어서, 상기 동물은 개과 동물, 고양이과 동물, 멧돼지과 동물, 소과 동물, 사슴과 동물, 기린과 동물, 페커리과 동물, 낙타과 동물, 하마과 동물, 말과 동물, 맥과 동물, 코뿔소과 동물, 족제비과, 토끼과, 설치류 및 인간을 제외한 영장류로 이루어진 군 중에서 선택된 1종 이상인 것을 특징으로 하는, 재세포화된 바이오 스캐폴드의 제조방법.
- 제 1항에 있어서, 상기 (1) 탈세포화 단계 전에,
1) 헤파린을 인간을 제외한 동물에 적용하는 단계;
2) 상기 1)단계의 동물의 문맥을 통하여 헤파린을 포함하는 PBS (phosphate buffered saline)를 동물에 관류시켜 혈액을 제거하는 단계;
3) 상기 2)단계의 동물의 하대 정맥 (inferior vena cava; IVC) 및 상대 정맥 (superior vena cava; SVC)을 절단하고, 상기 문맥을 통하여 동물의 간에 헤파린을 포함하는 PBS를 관류시켜 혈액을 제거하는 단계; 및
4) 상기 3)단계의 동물로부터 간을 분리하는 단계를 포함하는, 재세포화된 바이오 스캐폴드의 제조방법. - 제 1항에 있어서, 상기 (1)단계 후 (2)단계 전에,
a) PBS를 탈세포화된 간에 관류시켜 계면활성제를 제거하는 단계;
b) 상기 a) 단계의 간에 살균제를 관류시켜 간을 살균하는 단계;
c) 상기 b)단계의 살균된 간에 PBS를 관류시켜 잔여 화학물질을 제거하는 단계;를 포함하는, 재세포화된 바이오 스캐폴드의 제조방법. - 제 1항에 있어서, 상기 (2)단계의 재세포화하는 단계는,
(2-1) 세포 배양액을 인간을 제외한 동물의 간문맥을 통하여 관류시키는 단계;
(2-2) 간세포를 간문맥을 통하여 관류시키는 단계; 및
(2-3) 세포 배양액을 간문맥을 통하여 관류시키는 단계;를 포함하는 것을 특징으로 하는, 재세포화된 바이오 스캐폴드의 제조방법. - 제 1항 내지 제 8항 중 어느 한 항의 방법으로 제조된, 재세포화된 바이오 스캐폴드.
- 제 9항의 재세포화된 바이오 스캐폴드에 항암제 후보물질을 투여하는 단계;를 포함하는 재세포화된 바이오 스캐폴드를 이용한 항암제의 스크리닝 방법.
