Abstract
Description
도 2는 뇌 신경교종 줄기세포 표지마커 CD133 ECD의 단백질 발현 능력 및 단백질 정제를 나타낸다. 도 2의 (a)에서 B.I(before induction)는 IPTG를 넣기 전, O/N(overnight incubation)은 18℃에서 과발현 시킨 상태의 단백질을 나타낸다. 도 2의 (a)를 통해 발현된 단백질의 수용성(Sol, soluble form protein)과 불용성(InS, insoluble form of protein)의 비율을 알 수 있다. CD133 단백질이 불용성 형태로 발현되는 것을 확인 하였다. 도 2의 (b)는 불용성 단백질 형태로 발현된 CD133를 정제 해내기 위해 변성 조건(denature condition) 정제법을 수행하여, 친화성 크로마토그래피를 통해 CD133 단백질을 선별한 것을 나타낸다. F3-F11은 친화성 크로마토그래피를 통해 모아진 단백질을 나타낸다.
도 3은 CD133 단백질과 결합하는 페이지 펩타이드를 스크리닝하기 위한 M13 페이지 스크리닝의 계획도를 나타낸다. 플레이트의 각 웰에 CD133 을 고정시키고(도 3의 (1)), M13 페이지 표면에 12개의 아미노산으로 구성된 펩타이드들이 발현된 페이지 라이브러리를 첨가한 후(도 3의 (2)), 이에 다양한 세척 조건과 결합 시간을 적용시키면 CD133 단백질에 극한 조건에서도 결합하는 페이지 펩타이드가 웰 에 남는다(도 3의 (3)). 마지막으로 이를 추출해 내면 CD133 단백질에 특이적으로 결합하는 페이지 펩타이드만이 최종적으로 확보 된다(도 3의 (4)). 도 3에서 (1)-(4)까지를 한 라운드(round)라고 보고, 상기와 같은 과정을 반복하는 것을 바이오 패닝(biopanning)이라 칭한다. 본 발명자들은 3 라운드의 바이오 패닝을 실시하였다.
도 4는 CD133 촉매도메인 단백질에 대하여 높은 특이성 및 결합력을 페이지 펩타이드를 검출하기 위한 각 라운드(round)의 바이오 패닝 조건을 나타낸다. 각 라운드 당 NaCl과 Tween-20의 비율을 높여주는 반면 페이지 펩타이드 결합시간은 짧게 하여 극한 조건에서도 CD133 단백질에 결합하는 페이지 펩타이드를 검출해 내었다.
도 5는 CD133 단백질에 결합하는 총 35개의 페이지 플라그를 확보한 결과를 나타낸다. 35개의 페이지 플라그는 4종류의 서로 다른 서열의 펩타이드로 분류되었다.
도 6은 CD133 단백질에 결합하는 총 35개의 페이지 플라그 확보: 35개의 페이지 플라그는 4종류의 서로 다른 서열의 펩타이드로 분류되었다. 네 개의 펩타이드 모두 picomolarity 수준으로 CD133 단백질에 효과적으로 결합하는 것을 확인하였다.
도 7은 CD133 단백질에 결합하는 페이지 중, 이차적으로 선별된 펩타이드 서열을 형광 프로브와 합성하였으며, CD133 단백질과 결합력 측정을 수행한 결과를 나타낸다. CD133-GCGK-FITC는 대상물질과 나노몰(nM) 수준의 강한 결합을 이루는 것을 확인하였다.
도 8은 CD133을 발현하는 U87 세포에 대한 선별 펩타이드 서열의 결합력을 분석한 결과를 나타낸다. CD133 항체(antibody)-PE, sc-펩타이드 및 선별된 펩타이드를 이용하여 결합력을 측정한 결과 선별된 펩타이드가 특이적으로 CD133에 강한 결합력으로 결합하는 것을 확인하였다.
