Abstract
Description
도 2는, 산소유도 망막병증 동물 모델의 망막에서 ERR-γ의 발현을 확인한 것으로, A는 대조군을, B는 산소유도 망막병증 동물 모델에 대한 실험 결과를 나타낸다.
도 3은, hVEGF 프로모터 및 ERR-γ 발현 벡터를 공동-형질감염(co-transfection)한 C2C12 세포에 화학식 1로 표시되는 화합물을 처리한 다음, VEGF 프로모터 활성을 측정한 결과를 나타낸 것이다.
도 4에서, A는 생후 7일부터 12일까지 고산소 처리한 다음, 생후 12일부터 17일까지 정상산소로 복귀시켜 망막병증을 유도한 산소유도 망막병증 쥐에서 안구를 적출한 후, 망막을 분리하여 mRNA를 추출한 다음, PCR을 진행하여 VEGF mRNA의 발현을 확인하여 수치화한 결과를 나타내는 것이다. 또한, B는 생후 7일부터 12일까지 고산소 처리한 다음, 생후 12일에 정상 산소로 복귀시킨 쥐가 생후 14일이 되었을 때, 화학식 1로 표시되는 화합물을 망막 내 주입하고, 같은 개체의 반대쪽 눈에는 30% PEG(polyethylene glycol)을 주입하여 화학식 1로 표시되는 ERR-γ 저해제의 효과를 VEGF mRNA 측정으로 확인한 결과를 나타낸 것이다.
도 5에서, RGC-5 세포에 데페록사민(Deferoxamine)을 100μM의 농도로 처리하고 ERR-γ 저해제를 도면에 표시된 농도로 동시에 처리한 다음, 12시간 후 mRNA를 추출하여 VEGF 및 액틴(actin)에 대한 PCR을 수행한 결과를 A에 나타내었다. 상기 도 5의 A 결과를 수치화한 결과를 B에 나타내었다.
Claims (7)
- ERR-γ(estrogen-related receptor gamma)의 발현 또는 활성을 저해하는 물질을 포함하는, 망막병증의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 ERR-γ의 발현 또는 활성을 저해하는 물질은 ERR-γ mRNA의 발현을 억제하는 올리고 뉴클레오티드, 항체, 또는 화합물인 것인 조성물.
- 제1항에 있어서, 상기 조성물은 안구 투여용인 것인 조성물.
- ERR-γ의 발현 또는 활성을 저해하는 물질을 포함하는, 망막병증의 예방 또는 개선을 위한 건강기능식품 조성물.
- (a) 망막병증이 유발된 분리된 망막 시료에 망막병증 치료 후보 물질을 처리하는 단계; 및
(b) 상기 후보 물질이 처리된 망막 시료에서 ERR-γ 유전자의 mRNA 또는 이의 단백질 수준 또는 이의 활성을 측정하는 단계를 포함하는, 망막병증 치료제의 스크리닝 방법.
- 제6항에 있어서, 상기 방법은 (c) 상기 (b) 단계에서 측정된 ERR-γ 유전자의 mRNA 또는 이의 단백질 수준; 또는 이의 활성이 후보 물질이 처리되지 않은 시료에 비해 감소하면 망막병증 치료제로 판단하는 단계를 추가로 포함하는 것인, 방법.
