Abstract
Description
미성숙 랫트에 10 IU PMSG를 연속 4일 동안 투여하고, 그 다음 30 IU hCG를 투여함. 대조군 랫트는 10 IU PMSG를 4일 연속 투여하고, 5일째에 살린을 투여함.
도 2는 고나도트로핀 자극 후 랫트 난소에서 IL-11 유전자 발현을 측정한 결과이다.
A, 랫트 IL-11 cDNA 프로브를 이용하여 노던 블롯팅을 수행하여 난소 총 RNA(20 μg)를 분석함. 28S rRNA의 발현은 내부 표준 대조군으로 사용됨. 결과는 3번의 독립적인 실험에 대하여 나타냄. B, 배란 전 난포로부터 분리한 난포막 세포(TC) 및 과립막 세포(GC)에서 IL-11의 발현 정도를 실시간 PCR 방법으로 측정함. C, IL-11 mRNA의 인 시투 위치를 관찰한 결과임. 35S-표지 랫트 IL-11 cRNA 프로브와 난소 절편을 혼성화함. 난포막 세포에서의 IL-11 발현은 난포 크기와 관계없음을 나타냄. 현미경사진은 명 시야 및 암 시야 조건에서 촬영함(P, 배란 전 난포; G, 성장하는 난포; 화살표, 난포막 세포).
도 3은 인 비보에서 배란-억제 억제제에 의한 hCG-유도 IL-11 발현 조절을 측정한 결과이다.
억제제(RU486, 프로게스테론 수용체 안타고니스; 인도메타신, 사이클로옥시제나제 억제제; NDGA, 리폭시게나제 억제제; GW9662, PPARγ 안타고니스트)는 hCG 투여와 동시에 주사함. 난소 내 IL-11 발현 정도는 실시간 PCR로 측정함. 결과는 2번의 독립적인 실험에 대하여 ‘평균±표준오차’로 나타냄.
도 4는 인 비트로에서 배양한 배란 전 난포에서 LH-자극 IL-11 발현에 대한 신호전달 과정을 규명한 결과이다.
배란 전 난포는 LH가 존재하거나(200 ng/ml) 존재하지 않는 조건(C, 대조군)에서 H-89(10 μM; PKA 억제제), MDL-12,330A (30 μM; PKA 억제제), U73122 (10 μM; PLC 억제제), U0126 (10 μM; MEK 억제제), 스타우로스포린(100 nM; PKC 억제제) 및 LY294002(10 μM; PI3K 억제제)를 포함하는 신호전달 억제제를 처리 또는 처리하지 않으면서 6시간 동안 배양함. 결과는 2번의 독립적인 실험에 대한 값으로 나타냄.
도 5는 난소과자극증후군 랫트에서 IL-11의 난소 발현을 측정한 결과이다.
고용량으로 고나도트로핀을 투여하여 난소과자극증후군을 유도하고, 실시간 PCR 방법으로 발현 정도를 분석함. 난소과자극증후군 유도를 평가하기 위해 VEGF의 발현(A) 정도를 측정함. 결과는 3번의 독립적인 실험에 대하여 ‘평균±표준오차’로 나타냄.
도 6은 과립막 세포에서 IL-11에 의한 유전자 조절을 측정한 결과이다.
PMSG를 48시간 동안 투여한 미성숙 랫트의 난소로부터 수득한 배란 전 난포의 과립막 세포를 무혈청 배지에서 95% O2 , 5% CO2, 37℃에서 LH가 존재(200 ng/ml) 또는 존재하지 않고(C, 대조군), 재조합 인간 IL-11을 투여(100 ng/ml) 또는 투여하지 않는 조건에서 6시간 동안 배양함
도 7은 배란 전 난포에서 스테로이드 생성에 대한 IL-11의 효과를 측정한 결과이다.
배란 전 난포를 LH(200 ng/ml)가 존재 또는 존재하지 않는 조건(C, 대조군)에서 재조합 인간 IL-11을 농도별로 처리하여 12시간 동안 배양함. 배양 배지의 스테로이드 농도(평균±표준오차)는 RIA로 결정함. 결과는 2번의 독립적인 실험에 대하여 나타냄.
도 8은 인 비트로에서 배양한 배란 전 난포의 난구 확장에 있어서 IL-11이 미치는 영향에 대해 측정한 결과이다.
eCG를 투여한 랫트로부터 수득한 비확장 난구세포복합체를 EGF(100 μg/ml), FSH (100 ng/ml) 또는 IL-11 (100 ng/ml)를 포함하는 배지에서 배양함. 결과는 2번의 독립적인 실험에 대하여 나타냄.
도 9는 체외 수정(in vitro fertilization)을 진행 중인 환자의 난포액(follicular fluid, FF)에서 IL-11 레벨을 ELISA로 측정한 결과이다.
A, 난포액에서 IL-11의 레벨과 나이와의 관계는 선형 회귀 분석으로 확인함. B, 난소를 자극시킨 불임 여성(n = 38)의 난포액에서 IL-11 농도를 측정하고, 불임 인자와 난포액의 IL-11 농도를 플로팅(plotting) 함(C). 괄호안의 숫자는 샘플 수를 의미함. PCOS(다낭성난소증후군, polycystic ovarian syndrome), Tubal(난관이상), Endometriosis(자궁내막증)

- 난소과자극증후군(ovarian hyperstimulation syndrome)에 필요한 정보를 제공하기 위하여, 체외로 분리된 인간의 난포액에서 인터루킨-11(interleukin-11)의 양을 측정하는 방법.
- 제 1 항에 있어서, 상기 방법은 항원-항체 반응 방식으로 실시되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 방법은 마이크로어레이 방식으로 실시되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 방법은 유전자 증폭 방식으로 실시되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 인터루킨-11은 정상 난포액에서의 수준에 비해 난소과자극증후군 난포액에서 수준이 증가되어 있는 것을 특징으로 하는 방법.
- (a) 인터루킨-11 유전자 서열에 특이적으로 결합하는 프라이머 또는 프로브, 또는 (b) 인터루킨-11 단백질에 특이적으로 결합하는 항체 또는 앱타머를 포함하는 난소과자극증후군 진단용 키트.
- 제 6 항에 있어서, 상기 키트는 면역분석(immunoassay)용 키트인 것을 특징으로 하는 키트.
- 제 6 항에 있어서, 상기 키트는 마이크로어레이 키트인 것을 특징으로 하는 키트.
- 제 6 항에 있어서, 상기 키트는 유전자 증폭 키트인 것을 특징으로 하는 키트.
