Claims (14)
- 치료용 세포의 무균 배양을 위한 밀폐형 세포 배양 시스템으로서,
(ⅰ) 세포 성장을 지원하고 배양 중에 가스가 세포로 전달되도록 하기에 적절한 가스 투과성 부분과, 기저부(base)에 접하는 적어도 하나의 벽을 포함하는 용기(vessel)로서, 세포 배양을 지원하기 위한 필수 매체량을 수용하기 위한 내부 체적을 한정하는 용기와,
(ⅱ) 서로 유체 연통되는 내부 오리피스와 이로부터 이격되는(distal therefrom) 외부 오리피스를 한정하는 도관(conduit)을 포함함으로써 가스가 배출될 수 있도록 상기 용기를 채우는 동안에 압력의 균형을 허용하면서 세포-비움 과정 동안에 가스가 상기 내부 체적내로 유입되도록 허용하는 배출구(vent)로서,
상기 도관은 상기 용기의 벽, 기저부 또는 뚜껑을 통해 밀폐형 시스템의 외부로부터 연장되고 상기 용기의 내부 체적 내로 연장되어 상기 내부 오리피스와 함께 내부 체적 내에서 종결됨으로써, 상기 내부 오리피스의 액체와의 노출은 액체 매체의 충전 및 비움 중에 최소화되도록 배열되며, 상기 외부 오리피스는 무균 필터에 부착될 수 있도록 배열된 배출구와,
(ⅲ) 유체와 세포가 상기 용기 내로 무균 도입을 위한 출입구(port) 또는 출입구들(ports)로서, 상기 시스템을 외부 환경에 노출시키지 않은 상태에서 유체가 상기 시스템에서 배출되도록 하기 위하여, 상기 시스템이 세포를 유출구(exit port)와 유체 연통된 상태에 두도록 배향될 때 내부에서 성장한 세포가 중력에 의해 상기 시스템에서 배출될 수 있도록 한 출입구(port) 또는 출입구들
을 포함하는 밀폐형 세포 배양 시스템. - 제1항에 있어서, 상기 용기는 일체형인 밀폐형 세포 배양 시스템.
- 제2항에 있어서, 상기 도관이 연장되는 상기 용기의 벽, 기저부 또는 뚜껑은 상기 용기의 벽인 밀폐형 세포 배양 시스템.
- 제2항에 있어서, 상기 용기의 일부는 상기 용기가 적절히 배향될 때 상기 유출구로 세포를 보내도록 배열되는 밀폐형 세포 배양 시스템.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 배출구와 상기 출입구들은 상기 용기의 동일한 벽, 기저부 또는 뚜껑에 배치되는 밀폐형 세포 배양 시스템.
- 제5항에 있어서, 상기 배출구와 상기 출입구들은 상기 기저부의 대향측에 배치되는 밀폐형 세포 배양 시스템.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 유출구는 상기 용기의 내부 체적의 중심축과 동축이 아닌 밀폐형 세포 배양 시스템.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 유출구는 상기 용기의 벽, 기저부 또는 뚜껑에 위치하면서 상기 출입구는 상기 내부 체적내로 0.15mm 이하로 연장되는 밀폐형 세포 배양 시스템.
- 치료용 세포의 무균 배양 방법으로서,
(a) 제1항 내지 제4항 중 어느 한 항에 청구된 밀폐형 시스템에 배양용 세포군을 무균 첨가하는 단계와,
(b) 상기 세포 배양을 위한 영양분을 상기 시스템에 무균 첨가하는 단계와,
(c) 표적(T) 세포군을 증식시키기 위해 상기 세포를 배양하는 단계와,
(d) 상기 세포를 적절한 컨테이너 내로 무균 수확하는 단계를 포함하면서,
단계(a)는 단계(b) 이전이나 이후, 또는 단계(b)와 동시에 수행될 수 있는 치료용 세포의 무균 배양 방법. - 제9항에 있어서, 수확 후에, 상기 증식된 세포군을 세척하는 추가 단계를 포함하는 치료용 세포의 무균 배양 방법.
- 제9항에 있어서, 수확 후에, 상기 증식된 세포군을 계수하는 추가 단계를 포함하는 치료용 세포의 무균 배양 방법.
- 제11항에 있어서, 상기 세포의 치료 투여량을 적절한 컨테이너 내로 분리하는 추가 단계를 포함하는 치료용 세포의 무균 배양 방법.
- 제9항에 있어서, DMSO와 같은 첨가제가 수확된 세포에 첨가되는 치료용 세포의 무균 배양 방법.
