Abstract
Description
도 2는 패닝법을 이용하여 항체 라이브러리부터 항체를 선별하는 방법을 도식화한 것이다.
도 3은 패닝법을 이용하여 선별한 파아지에 디스플레이된 다클론 Fab 항체 (polyclonal Fab antibody)의 RNA에 대한 결합력을 ELISA로 분석한 결과를 나타낸 것이다.
도 4는 패닝법을 이용하여 선별한 파아지에 디스플레이된 Fab 형태의 단일 클론 항체의 RNA에 대한 결합력을 닷 블랏으로 분석한 결과를 나타낸 것이다.
도 5는 동물 세포주에서 발현된 항체를 정제한 후 SDS-PAGE로 분석할 결과를 나타낸 것이다.
도 6은 동물 세포주에서 발현된 항체를 정제한 후 ELISA로 분석한 결과를 나타낸 것이다.
도 7은 단일 클론 항체와 RNA와의 필터 결합법을 도식화한 것이다.
도 8은 전체 IgG 항체의 RNA에 대한 결합력을 필터 결합법으로 분석한 결과를 나타낸 것이다.

Claims (12)
- (a) 고정상 지지체에 표적 RNA를 고정하는 단계;
(b) 상기 고정화된 표적 RNA에 파지 디스플레이-인간 항체 라이브러리를 결합시키는 단계;
(c) 상기 표적 RNA에 결합되지 않은 항체를 발현하는 파아지를 제거하는 단계 및
(d) 효모 전체 RNA(yeast total RNA)를 넣어 상기 표적 RNA에 결합한 항체 발현 파아지를 분리하고 항체를 얻는 단계를 포함하고,
상기 (b) 결합단계, (c) 제거단계 및 (d) 항체를 얻는 단계를 반복하는,
RNA에 특이적으로 결합하는 항체의 선별방법.
- 제1항에 있어서, 상기 (b) 결합단계, (c) 제거단계 및 (d) 항체를 얻는 단계를 1 내지 5회 반복하는 것인 선별방법.
- 제1항에 있어서, 상기 표적 RNA는 BC200 RNA인 선별방법.
- BC200 RNA에 특이적으로 결합하는, 항체 또는 이의 항원 결합 단편으로서,
상기 항체는 서열번호 1의 아미노산 서열을 갖는 상보성결정영역 1 (CDR1), 서열번호 2의 아미노산 서열을 갖는 CDR2 및 서열번호 3의 아미노산 서열을 갖는 CDR3을 함유하는 중쇄 가변영역(VH) 및,
서열번호 4의 아미노산 서열을 갖는 CDR1, 서열번호 5의 아미노산 서열을 갖는 CDR2 및 서열번호 6의 아미노산 서열을 갖는 CDR3을 함유하는 경쇄 가변영역(VL)을 포함하는 항체 또는 항원 결합 단편 또는;
상기 항체는 서열번호 7의 아미노산 서열을 갖는 CDR1, 서열번호 8의 아미노산 서열을 갖는 CDR2 및 서열번호 9의 아미노산 서열을 갖는 CDR3을 함유하는 중쇄 가변영역(VH) 및,
서열번호 10의 아미노산 서열을 갖는 CDR1, 서열번호 11의 아미노산 서열을 갖는 CDR2 및 서열번호 12의 아미노산 서열을 갖는 CDR3을 함유하는 경쇄 가변영역(VL)을 포함하는 항체 또는 항원 결합 단편.

- 제4항에 있어서, 상기 항체 또는 항원 결합 단편은 scFv, (scFv)2, Fab, Fab' 및 F(ab')2로 이루어진 군으로부터 선택되는 항체 또는 항원 결합 단편.
- 서열번호 1, 서열번호 2 및 서열번호 3으로 이루어진 군으로부터 선택되는 아미노산 서열을 갖는 중쇄 상보성결정영역 (CDR)을 코딩하는 뉴클레오티드 서열; 및 상기 뉴클레오티드 서열의 상보적 서열을 포함하고, BC200 RNA에 특이적으로 결합하는 항체의 중쇄 상보성결정영역(CDR)을 코딩하는 핵산분자.
- 서열번호 4, 서열번호 5 및 서열번호 6로 이루어진 군으로부터 선택되는 아미노산 서열을 갖는 경쇄 상보성결정영역 (CDR)을 코딩하는 뉴클레오티드 서열; 및 상기 뉴클레오티드 서열의 상보적 서열을 포함하고, BC200 RNA에 특이적으로 결합하는 항체의 경쇄 상보성결정영역(CDR)을 코딩하는 핵산분자.
- 서열번호 7, 서열번호 8 및 서열번호 9로 이루어진 군으로부터 선택되는 아미노산 서열을 갖는 중쇄 상보성결정영역 (CDR)을 코딩하는 뉴클레오티드 서열; 및 상기 뉴클레오티드 서열의 상보적 서열을 포함하고, BC200 RNA에 특이적으로 결합하는 항체의 중쇄 상보성결정영역(CDR)을 코딩하는 핵산분자.
- 서열번호 10, 서열번호 11 및 서열번호 12로 이루어진 군으로부터 선택되는 아미노산 서열을 갖는 경쇄 상보성결정영역 (CDR)을 코딩하는 뉴클레오티드 서열; 및 상기 뉴클레오티드 서열의 상보적 서열을 포함하고, BC200 RNA에 특이적으로 결합하는 항체의 경쇄 상보성결정영역(CDR)을 코딩하는 핵산분자.
- 제4항 및 제7항 중 어느 한 항의 항체 또는 항원 결합 단편을 포함하는 BC200 RNA 탐지용 조성물.
