Abstract
Description
도 2는 Stau1과 NS1의 상호작용이 RBD3L의 투여량에 비례하여 억제됨을 보여준다. 배양된 HEK 293T 세포들을 NS1, 및 RBD3L을 포함하거나 포함하지 않는 Stau1 발현 벡터로 함께 트랜스펙션시켰다. 대조군 세포들은 1 의 빈 벡터로 트랜스펙션시켰다. (A) co-IP 분석의 대표적 결과. 아래 패널은 각각의 단백질의 발현 수준을 나타냄. (B) 데이터를 분석한 막대그래프. co-IP 어세이의 밴드 강도는 밀도계측분석(densitometric analysis)으로 정량하였다. RBD3L (1 ) 발현에 대한 밴드 강도는 각각의 대조군에 대한 백분율로 나타냈다(***p < 0.001, ns: not significant compared with controls).
도 3은 RBD3L 발현이 Stau1과 NS1이 공동으로 위치하는 것(colocalization)을 감소시킨다는 결과이다. 배양된 A549 세포들을 GFP-Stau1, Myc-NS1, 및 RBD3L 또는 RBD2(대조군)로 트랜스펙션시켰다. 배양 24시간 후에, 세포들을 고정하고, 항-Myc 항체로 면역 염색하였다. 핵을 염색하기 위해 DAPI를 사용하였다. 도 4는 RBD3L 발현이 바이러스 복제를 감소시킴을 보여준다. A549 세포들을 FLAG-표지 Stau1, RBD3L, RBD2, 또는 빈 플라스미드 벡터 대조군으로 트랜스펙션시키고, 24시간 동안 배양하였다. 그 후 A/WSN/1933 (H1N1) 바이러스로 감염시켰다(0.01 MOI). 감염된 세포 배양액에서 바이러스 역가(titers)는 다른 시간에(12, 24, 36, 및 72 h p.i.) - A형 인플루엔자 바이러스 단백질 NS1의 숙주 단백질인 Staufen1에 대한 결합을 방해하는 물질인 서열번호 2의 아미노산 서열로 이루어진 RBD3L폴리펩티드를 유효성분으로 포함하는 A형 인플루엔자 바이러스 감염 질환의 예방 및 치료용 약학적 조성물.
