Abstract
본 발명에 따른 폴리감마글루탐산-키토산 나노입자를 함유하는 면역보강제 조성물은 독성 및 부작용이 거의 없으며, 바이러스 및 세균성 감염증에 대한 백신과 암에 대한 인체용 또는 동물용 예방 및 치료용 백신에 첨가되어 항체생성율을 증가시키는 효과가 있다.
Description
도 2는 항원 단백질의 혼합순서에 따른 단백질 도입률을 확인하기 위하여, FITC 표지된 OVA가 도입된 폴리감마글루탐산-키토산 나노입자의 공초점 레이저 형광 현미경 사진이다.
도 3은 OVA 항원에 대한 체액성 면역반응에 미치는 영향을 관찰하기 위하여, OVA와 PGA 분자량별로 제조된 폴리감마글루탐산-키토산 나노입자를 혼합하여 마우스에 주사 후 OVA 특이 항체 생성 정도를 OVA-specific serum IgG를 측정하여 확인한 결과를 나타낸 것이다.
도 4는 OVA와 전하에 따른 폴리감마글루탐산-키토산 나노입자를 혼합하여 마우스에 주사 후 OVA 특이 항체 생성 정도를 확인하고 Freund adjuvant와의 비교를 위해 OVA-specific serum IgG 생성 정도를 측정한 결과를 나타낸 것이다.
도 5는 폴리감마글루탐산-키토산 나노입자에 의한 마우스 비장 내 T세포의 OVA에 대한 세포성 면역반응을 조사하기 위하여, 분자량별 PGA를 이용하여 제조된 PGA-chitosan nanoparticle에 의한 IFN-γ를 분비하는 CD8+ T 세포의 활성화 정도를 FACS로 분석한 결과를 나타낸 것이다.
도 6는 혼합순서에 따라 제조된 폴리감마글루탐산-키토산 나노입자에 의한 수지상세포와 T 세포의 혼합비율에 따라 활성화된 T 세포를 확인하기 위하여 IFN-γ를 분비하는 CD8+ T 세포의 분포를 FACS로 분석한 결과를 나타낸 것이다.
도 7는 AI 단백질과 폴리감마글루탐산-키토산 나노입자를 혼합하여 마우스에 피하주사 또는 비강 투여 후 AI 단백질 특이 항체 생성 정도를 확인하기 위해 AI-specific serum IgG를 측정한 결과를 나타낸 것이다.
도 8은 폴리감마글루탐산-키토산 나노입자의 중화항체 유도능을 확인하기 위해 HI (Haemagglutination Inhibition) 테스트법을 사용하여 마우스 혈청에서의 항체 역가를 측정한 결과를 나타낸 것이다.
도 9는 폴리감마글루탐산-키토산 나노입자의 항체 생성능을 확인하기 위해 피하주사 또는 비강투여 후 바이러스를 감염시킨 마우스의 감염 경과일에 따른 사망 개체수 변화를 나타낸 그래프이다.
도 10은 인플루엔자 백신 항원과 폴리감마글루탐산-키토산 나노입자를 혼합하여 마우스에 근육주사한 후 인플루엔자 백신 항원 특이 항체 생성 정도를 확인하기 위하여 vaccine antigen-specific serum IgG를 측정한 결과를 나타낸 것이다.
도 11은 인플루엔자 백신 항원과 폴리감마글루탐산-키토산 나노입자의 항체 생성능을 확인하기 위해 근육주사 후 바이러스를 감염시킨 마우스의 감염 경과일에 따른 몸무게 변화를 나타낸 그래프이다.

Claims (13)
- 폴리감마글루탐산과 키토산이 이온결합으로 결합되어 있는 폴리감마글루탐산-키토산 나노입자를 함유하는 면역보강제(adjuvant) 조성물.
- 삭제
- 제1항에 있어서, 상기 폴리감마글루탐산-키토산 나노입자의 표면전하는 음전하인 것을 특징으로 하는 면역보강제 조성물.
- 제1항의 면역보강제 조성물을 인간을 제외한 동물에 항원과 함께 투여하여, 항원에 대한 항체 생성률을 증가시키는 방법.
- 제4항에 있어서, 투여는 피하주사, 근육내 주사, 피하내 주사, 복막내 주사, 비강투여, 구강투여, 경피투여 및 경구투여로 구성된 군에서 선택된 어느 하나의 방법으로 수행되는 것을 특징으로 하는 방법.
- 항원을 포함하는 백신용 조성물에 있어서, 폴리감마글루탐산 나노입자와 키토산이 이온결합으로 결합되어 있는 폴리감마글루탐산-키토산 나노입자와 항원을 포함하는 백신용 조성물.
- 제6항에 있어서, 상기 항원은 단백질, 팹타이드, 뉴클레오시드, 뉴클레오티드, 바이러스, 항바이러스제, 항종양제, 항생물질 및 항염증제로부터 선택된 것을 특징으로 하는 백신용 조성물.
- 제6항에 있어서, 조류 인플루엔자 바이러스, 돼지 인플루엔자 바이러스 및 신종 인플루엔자 바이러스로 선택된 군에서 어느 하나 이상의 바이러스에 의한 질병의 예방용 또는 치료용인 것을 특징으로 하는 백신용 조성물.
- 제6항에 있어서, 자궁경부암, 피부흑색종암, 전립선암, 결장암, 폐암, 유방암, 난소암, 두경부암, 외음부암, 방광암, 뇌암 및 신경교종으로 선택된 군에서 어느 하나 이상의 질병의 예방용 또는 치료용인 것을 특징으로 하는 백신용 조성물.
- 다음 단계를 포함하는 폴리감마글루탐산-키토산 나노입자와 표면전하가 음전하인 항원을 포함하는 백신의 제조방법:
(a) 표면전하가 음전하인 항원과 키토산을 이온결합시키는 단계; 및
(b) 상기 키토산과 결합한 항원에 폴리감마글루탐산을 첨가하여, 키토산과 폴리감마글루탐산을 이온결합시켜, 폴리감마글루탐산-키토산 나노입자와 항원이 결합되어 있는 백신을 제조하는 단계.
- 다음 단계를 포함하는 폴리감마글루탐산-키토산 나노입자와 표면전하가 양전하인 항원을 포함하는 백신의 제조방법:
(a) 표면전하가 양전하인 항원과 폴리감마글루탐산을 이온결합시키는 단계; 및
(b) 상기 폴리감마글루탐산과 결합한 항원에 키토산을 첨가하여, 폴리감마글루탐산과 키토산을 이온결합시켜, 폴리감마글루탐산-키토산 나노입자와 항원이 결합되어 있는 백신을 제조하는 단계.
- 제6항의 백신용 조성물을 인간을 제외한 동물에 투여하여, 항원에 대한 항체 생성률을 증가시키는 방법.
- 제12항에 있어서, 투여는 피하주사, 근육내 주사, 피하내 주사, 복막내 주사, 비강투여, 구강투여, 경피투여 및 경구투여로 구성된 군에서 선택된 어느 하나의 방법으로 수행되는 것을 특징으로 하는 방법.
