Abstract
Description
도 2는 Actin 6 유전자를 증폭대상으로 한 질편모충의 LAMP 반응의 감도를 측정한 결과를 나타낸 도이다. 질편모충의 게놈 DNA를 연속으로 희석한 후 LAMP 반응을 수행한 결과 질편모충의 게놈 DAN가 1pg만 있어도 검출이 가능함을 알 수 있었다.
도 3은 Actin 6 유전자를 타겟으로 한 LAMP 반응 증폭 산물의 실시간 검출 결과를 나타낸 도이다. 반응액 내에서 1pg만 있어도 40분 내에 증폭이 일어남을 확인하였다.
도 4는 LAMP 반응 산물의 전기영동과 제한효소 처리 분석 결과 및 Actin 6 유전자를 표적으로 한 질편모충의 LAMP 반응과 PCR의 검출 감도의 비교 결과를 나타낸 도이다. LAMP 반응에서 Actin 6 유전자가 증폭되었는지를 확인하기 위하여 Actin 6을 절단하는 제한효소 Hind Ⅲ 제한효소를 처리한 결과 예상한 DNA 절편이 관찰되었다. 또한, LAMP와 PCR의 검출 감도를 비교한 결과 PCR의 경우 DNA 농도가 10pg이상일 때 겸출되는 반면 LAMP 반응의 경우 DNA 농도가 1pg (10-12g)만 있어도 검출되는 것을 확인하였다.
도 5는 질편모충의 DNA를 정제까지 상당한 시간이 소요되는 일반적인 방법을 사용하여 분리하지 않고, 단순히 끓여서 확보한 후 DNA를 포함하는 세포용해액에 대한 LAMP 반응과 PCR의 검출 감도의 비교 결과를 나타낸 도이다. 질편모충 102개를 단순히 끓여 준비한 세포용해액에 대해서 LAMP 반응의 경우 102, 101, 100 (1) 개 모두 균일하게 증폭된 반응 양상이 관찰되었으나 기존 PCR 방법의 경우 증폭반응이 매우 희석 정도에 따라 약해져서 100 (1) 개의 경우 양성반응이 매우 미약하게 관찰되었다.
도 6는 질염이 있는 환자 10명을 대상으로 LAMP 반응을 수행한 결과를 나타낸 도이다. 정확하게 질편모충이 감염된 환자에서만 양성반응이 나타나는 것을 확인하였고 질염을 유발하는 대표적인 곰팡이류인 칸디다균(Candida albicans)와 교차반응이 없는 것을 확인하였다.

Claims (8)
- 서열번호 1 및 2의 염기서열로 이루어진 제 1프라이머 쌍;
서열번호 3 및 4의 염기서열로 이루어진 제 2프라이머 쌍; 및
서열번호 5 및 6의 염기서열로 이루어진 제 3프라이머 쌍으로 이루어진 질편모충 검출용 프라이머 세트.
- 제1항의 프라이머 세트를 포함하는 질편모충 검출용 조성물.
- 제2항에 있어서, 상기 조성물은 고리매개등온증폭용인 것을 특징으로 하는 조성물.
- 제2항 또는 제3항의 조성물을 포함하는 질편모충 검출용 키트.
- 제2항 또는 제3항의 조성물에 시료를 혼합하여 염기서열을 증폭하는 단계를 포함하는 질편모충의 검출 방법.
- 제5항에 있어서, 상기 방법은 1)상기 프라이머를 포함하는 조성물과 시료를 혼합하는 단계, 2)상기 혼합물에 대하여 DNA 서열 증폭 방법을 이용하여 서열을 증폭하는 단계, 및 3)상기 증폭물을 측정하는 단계로 이루어지는 것을 특징으로 하는, 질편모충의 검출 방법.
- 제5항에 있어서, 상기 서열 증폭은 고리매개등온증폭법을 이용하는 것을 특징으로하는, 질편모충의 검출 방법.
- 제5항에 있어서, 상기 시료는 여성의 자궁경부, 항문 인후의 분비액, 남성의 요도 분비물 전립선액, 인후 및 항문의 분비액으로 이루어진 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 질편모충의 검출 방법.
