Abstract
Description
도 2는 실험백서에 LPS를 농도별, 시간별로 처리한 후 혈장에서 검출된 APE1/Ref-1의 단백질 검출양 변화를 보여주는 전기영동 결과 사진이다.
도 3는 LPS를 처리한 실험백서의 혈장에서 검출된 APE1/Ref-1 단백질을 시간에 따라 정량화한 결과를 보여주는 도표이다.
도 4는 대조군으로 정상적인 APE1/Ref-1인 단백질로써 LPS를 처리한 실험백서의 간조직에서 APE1/Ref-1 단백질을 검출한 전기영동 결과 사진이다.
도 5는 LPS를 처리한 실험백서의 혈장에서 검출된 APE1/Ref-1 단백질이 정상 APE1/Ref-1에 비해 분절된 형태로 검출됨을 보여주는 전기영동 결과 사진이다.
Claims (6)
- 염증질환 및 패혈증 진단에 필요한 정보를 제공하기 위하여 잠재 환자로부터 채취한 혈액 시료로부터 항원·항체 반응을 통해 APE1/Ref-1 단백질을 정성 또는 정량분석하는 방법.
- 제 1 항에 있어서,
(A) 상기 혈액을 전기영동하는 단계;
(B) 전기영동 결과 겔에 1차 APE1/Ref-1 항체를 반응시킨 후 세척하는 단계;
(C) 표지체가 접합된 2차 APE1/Ref-1항체를 반응시킨 후 세척하는 단계;
(D) 발색제를 가하여 발색반응을 유도한 후 발색정도를 측정하는 단계;
를 포함하는 것을 특징으로 하는 APE1/Ref-1 단백질을 정성 또는 정량분석하는 방법.
- 제 2 항에 있어서,
상기 단계(A)에서 혈액에서 혈장만을 분리하여 전기영동하는 것을 특징으로 하는 APE1/Ref-1 단백질을 정성 또는 정량분석하는 방법.
- 제 2 항에 있어서,
상기 단계(C)에서 전기영동 결과 겔로부터 전개된 단백질을 멤브레인으로 옮긴 상태에서 항체반응시키는 것을 특징으로 하는 APE1/Ref-1 단백질을 정성 또는 정량분석하는 방법.
- 제 2 항 내지 제 4 항 중 어느 한 항에 있어서,
상기 2차 항체의 표지체는 HRP(horseradish peroxidase), 염기성 탈인산화효소(alkaline phosphatase), 콜로이드 골드(coloid gold), FITC(poly L-lysine-fluorescein isothiocyanate) 또는 RITC(rhodamine-B-isothiocyanate)인 것을 특징으로 하는 APE1/Ref-1 단백질을 정성 또는 정량분석하는 방법.
