Claims (15)
- 락테이트, 3-하이드록시부티레이트 및 중간사슬길이(MCL)의 3-하이드록시알카노에이트를 반복단위로 함유하는 3-하이드록시부티레이트-MCL 3-하이드록시알카노에이트-락테이트 삼중합체 [poly(3-hydroxybutyrate-co-MCL 3-hydroxyalkanoate-co-lactate)].
- 제1항에 있어서, 상기 MCL의 3-하이드록시알카노에이트는 3-하이드록시헥사노에이트 (3-hydroxyhexanoate, 3HHx), 3-하이드록시헵타노에이트 (3-hydroxyheptanoate, 3HHp), 3-하이드록시옥타노에이트 (3-hydroxyoctanoate, 3HO), 3-하이드로노나노에이트 (3-hydrononanoate, 3HN), 3-하이드록시데카노에이트 (3-hydroxydecanoate, 3HD), 3-하이드록시운데칸노에이트 (3-hydroxyundecanoate, 3HUD) 및 3-하이드록시도데카노에이트 (3-hydroxydodecanoate, 3HDD)로 구성된 군에서 선택되는 것을 특징으로 하는 공중합체.
- 락테이트(lactate)를 락틸-CoA(lactyl-CoA)로 전환하고 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자 및 폴리하이드록시알카노에트(polyhydroxyalkanoate:PHA) 합성효소 유전자를 동시에 가지는 세포 또는 식물을 배양 또는 재배하는 것을 특징으로 하는 3-하이드록시부티레이트-MCL 3-하이드록시알카노에이트-락테이트 삼중합체 [poly(3-hydroxybutyrate-co-MCL 3-hydroxyalkanoate-co-lactate)] 의 제조방법.
- 제3항에 있어서, 상기 MCL의 3-하이드록시헥사노에이트 (3-hydroxyhexanoate, 3HHx), 3-하이드록시헵타노에이트 (3-hydroxyheptanoate, 3HHp), 3-하이드록시옥타노에이트 (3-hydroxyoctanoate, 3HO), 3-하이드로노나노에이트 (3-hydrononanoate, 3HN), 3-하이드록시데카노에이트 (3-hydroxydecanoate, 3HD), 3-하이드록시운데칸노에이트 (3-hydroxyundecanoate, 3HUD) 및 3-하이드록시도데카노에이트 (3-hydroxydodecanoate, 3HDD)로 구성된 군에서 선택되는 것을 특징으로 하는 방법.
- 제3항에 있어서, 세포 또는 식물은 상기 두 유전자를 모두 가지지 않는 세포 또는 식물을 락테이트(lactate)를 락틸-CoA(lactyl-CoA)로 전환하고 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자와 lactyl-CoA를 기질로 사용하는 PHA 합성효소의 유전자로 형질전환하여 수득되는 것임을 특징으로 하는 방법.
- 제3항에 있어서, 세포 또는 식물은 락틸-CoA(lactyl-CoA)를 기질로 사용하는 PHA 합성효소의 유전자를 가지는 세포 또는 식물을 락테이트(lactate)를 락틸-CoA(lactyl-CoA)로 전환하고 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자로 형질전환하여 수득되는 것임을 특징으로 하는 방법.
- 제3항에 있어서, 세포 또는 식물은 락테이트(lactate)를 락틸-CoA(lactyl-CoA)로 전환하고 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자를 가지는 세포 또는 식물을 lactyl-CoA를 기질로 사용하는 PHA 합성효소의 유전자로 형질전환하여 수득되는 것임을 특징으로 하는 방법.
- 제3항에 있어서, 상기 락테이트(lactate)를 락틸-CoA(lactyl-CoA)로 전환하고 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자는 프로피오닐-CoA 트랜스퍼라아제 유전자(pct)인 것을 특징으로 하는 방법.
- 제3항에 있어서, 폴리하이드록시알카노에트(polyhydroxyalkanoate:PHA) 합성효소 유전자는 슈도모나스 속(Pseudomonas sp.) 6-19 유래의 phaC1ps6 -19인 것을 특징으로 하는 방법.
- 제3항에 있어서, 폴리하이드록시알카노에트polyhydroxyalkanoate:PHA) 합성효소 유전자는 서열번호 7의 아미노산 서열에서 E130D, S325T 및 Q481M가 변이된 아미노산 서열을 코딩하는 유전자인 것을 특징으로 하는 방법.
- 제3항에 있어서, 세포 또는 식물은 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소의 유전자를 추가로 함유하는 것을 특징으로 하는 방법.
- 제11항에 있어서, 상기 3-하이드록시알카노에트(3-hydroxyalkanoate)를 3-하이드록시알카노일-CoA(3-hydroxyalkanoyl-CoA)로 전환하는 효소는 α-케토티올레 이즈(PhaA) 또는 아세토아세틸-CoA 리덕테이즈(acetoacetyl-CoA reductase: PhaB)인 것을 특징으로 하는 방법.
- 제3항에 있어서, 세포는 미생물인 것을 특징으로 하는 방법.
- 제13항에 있어서, 미생물은 대장균인 것을 특징으로 하는 방법.
- 제3항에 있어서, 배양 또는 재배는 3-하이드록시부티레이트(3-HB) 및MCL 3-하이드록시알카노에이트(MCL 3-HA)를 함유하는 환경에서 수행되는 것을 특징으로 하는 방법.
