Abstract
변색된 치아를 미백시키고, 치석 등을 제거할 수 있는 치아용 미백기능성 물질 전달시스템이 개시된다. 상기 치아용 미백기능성 물질 전달시스템은 치아에 도포된 후, 치아표면을 신속하게 미백시킬 뿐만 아니라, 장기적으로 미백상태를 유지시키는 치아미백용 젤 및 액상 조성물을 의미한다. 상기 치아미백용 젤 조성물은 점착성 고분자, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 소수성 계면 활성제, 안정화제, 피로포스페이트(pyrophosphate)류 및 이산화규소(SiO2)를 포함하고, 또한 상기 치아미백용 액상 조성물은 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 안정화제, 치아 코팅용 점착성 고분자 및 휘발성 용매를 포함한다.

본 발명은 치아용 미백기능성 물질 전달시스템에 관한 것으로서, 더욱 상세하게는 변색된 치아를 미백시키고, 치석 등을 제거할 수 있는 치아용 미백기능성 물질 전달시스템에 관한 것이다. 본 발명에서 치아용 미백기능성 물질 전달시스템은 치아에 도포된 후, 치아표면을 신속하게 미백시킬 뿐만 아니라, 장기적으로 미백상태를 유지시키는 치아미백용 젤 및 액상 조성물을 의미한다.
치아는 내부의 상아질 층과 치아의 보호벽이 되는 외부의 범랑질층으로 구성되어 있다. 치아의 범랑질층의 색은 원래 불투명 흰색 또는 약간 황색이다. 상기 범랑질은 자연적인 공간이나 구멍이 있으며, 여기에 약간의 착색물질이나 변색제가 침투하여 치아를 변색하게 한다. 개인이 매일 접촉하는 많은 물질이 치아를 착색시키거나 치아의 미백을 감소시킬 수 있다. 특히, 음식물 및 담배 등은 치아의 범랑질층에 축적되어 치아에 얇은 막과 같은 것을 형성시키며, 이들 착색변질 물질이 범랑질층을 통과할 수 있다. 이런 일이 여러 해 동안 반복적으로 발생하면서, 개인 치아의 범랑질을 두드러지게 변색시킬 수 있다. 또한, 어떤 질병이나 환경적 요인이 치아의 변색에 또한 영향을 끼칠 수 있다. 물론 변색된 치아도 건강을 나타낼 수 있고 어떤 건강상의 위험이나 문제를 발생시키지는 않으나, 최근 치아가 투명하거나 흰색이 되기를 갈망하는 사람들이 지속적으로 증가하는 추세이다.
현재 사용되고 있는 치아미백제 중 불소함유 치약은 음식물 찌꺼기나 니코틴, 커피 및 홍차 등이 치아표면에 흡착하거나 치면 상의 치석이나 여성 침전물 등이 색소성분에 의해 염색됨으로써 발생된 치아의 얼룩을 화학 작용에 의해 제거하는 작용을 하는 것으로, 입안에 깨끗한 느낌을 주거나 입 냄새를 줄이는 등의 용도로 일반적으로 사용되고 있다. 그러나 치약을 사용하여 칫솔질을 통해 치아미백 효과를 달성하기는 어려웠다.
따라서, 치과의사들에 의해 제공되는 전문적인 치아미백 프로그램인 진료실내 미백 처리와 진료실 외 미백 처리 방법이 나왔다. 진료실 내 처리는 통상적으로 과산화수소인 미백 약품이 구강연 조직과 접촉하는 것을 방지하기 위하여 입안에 특별하게 끼워 맞춰지는 고무댐을 제조하는 것으로부터 시작한다. 진료실 외 미백프로그램은 환자가 장기간, 통상적으로 몇 주 동안 하루에 몇 시간에 걸쳐서 저 강도의 미백약품을 사용하여 환자가 본인의 치아에 미백제를 접촉시킨다는 점에서 차이가 있다. 진료실 외 프로그램에서는 통상적으로 특정 환자에게 특정한 기구를 치과 진료소에서 초기에 끼워 맞출 필요가 있다. 상기 도구는 환자의 치아에 정교하게 끼워 맞춰지도록 제조되어 환자의 미백 젤을 전달하는 장치이다. 그러나 상기 방법들은 비용이 많이 들뿐만 아니라 일상생활을 유지하는데 어려움이 있었다.
미국특허 US 5,425,953에서는 치아 미백용 액상 폴리머 조성물을 개시하였고, 상기 액상 폴리머 조성물은 치아에 적용 전에는 액상이고, 치아에 적용 후 고형 필름상으로 변환되는 시스템을 이용하여 젤 상 제품의 단점을 개선하고자 하였다. 또한 미국특허 US 5,891,453은 휨강성이 낮은 스트립 재질을 사용하여 미백물질을 치아에 전달하는 시스템에 대해 개시하고 있으며, 상기 기술을 이용한 치아미백물질 전달 시스템은 일상생활이 용이하나, 스트립 재질 즉, 방수성 스트립 재질은 치아에 적용 후 일정시간이 경과한 다음 떼어내어야 하는 불편함이 있으며, 휨 강성을 조절하여 치아 부착 시 휘어지기 쉽도록 하였지만 방수성 필름 자체의 이물감이 존재하는 문제점이 있다. 또한 이는 기존 젤 상의 미백 젤을 필름에 도포한 것에 불과하므로, 손에 묻어나거나 끈적거림이 심하여 미백활성 물질의 전달이 용이하지 않은 단점이 있다.
따라서, 당업계에서는 기존 미백제의 사용감을 그대로 유지하면서, 치아미백 투과율이 뛰어나고, 치아를 미백시키기에 충분한 시간 동안 침식되지 않아, 미백 효과를 장기적으로 유지할 수 있으며, 미백 효과는 더 뛰어난 치아미백용 조성물의 개발이 요구되고 있다.
이에, 본 발명자들은 종래 치아미백제에 비해 치아표면 투과율 및 미백 효과가 뛰어난 치아미백용 조성물을 개발하기 위하여 예의 노력한 결과, 점착성 고분자, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 소수성 계면 활성제, 안정화제, 피로포스페이트(pyrophosphate)류 및 이산화규소(SiO2)를 포함하는 치아미백용 젤 조성물이, 기존 미백제의 사용감은 그대로 유지하면서 미백 효과는 신속하고, 장기적으로 유지할 수 있음을 확인하고, 본 발명을 완성하게 되었다.
또한, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 안정화제, 치아 코팅용 점착성 고분자 및 휘발성 용매를 포함하는 치아미백용 액상 조성물이, 기존 미백제의 사용감은 그대로 유지하면서 미백효과는 신속하고, 장기 적으로 유지할 수 있음을 확인하고, 본 발명을 완성하게 되었다.
결국 본 발명의 목적은, 기존 치아미백제의 사용 감을 그대로 유지하면서, 신속하게 미백시킬 뿐만 아니라, 장기적으로 미백상태를 유지시킬 수 있는 치아미백 조성물을 제공하는데 있다.
상기 목적을 달성하기 위하여, 본 발명은, 일 관점에서 점착성 고분자, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 소수성 계면 활성제, 안정화제, 피로포스페이트(pyrophosphate)류 및 이산화규소(SiO2)를 포함하는 치아미백용 젤 조성물 을 제공한다.
본 발명에 있어서, 상기 치아미백용 젤 조성물의 상기 점착성 고분자는 하이드록시프로필메틸셀룰로오스, 하이드록시에틸셀룰로오스, 폴리비닐알코올, 폴리비닐피롤리돈류, 하이드록시프로필셀룰로오스, 폴리아크릴산 유도체인 카보머, 메틸 비닐 에테르와 무수 말레인산의 공중합체 및 폴리에틸렌옥사이드로 구성된 군으로부터 선택되는 어느 하나 또는 2 이상인 것을 특징으로 한다.
본 발명은, 다른 관점에서, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 안정화제, 치아 코팅용 점착성 고분자 및 휘발성 용매를 포함하는 치아미백용 액상 조성물을 제공한다.
본 발명에 따른 상기 치아미백용 액상 조성물의 상기 치아 코팅용 점착성 고 분자로는 12 내지 120의 K값을 갖는 폴리비닐피롤리돈(Polyvinylpyrrolidone)을 사용할 수 있으며, K값이 90인 폴리비닐피롤리돈을 사용하는 것이 바람직하다.
이하, 첨부된 도면을 참조하여 본 발명을 상세히 설명하면 다음과 같다.
본 발명에 따른 치아미백용 젤 조성물은 점착성 고분자, 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 소수성 계면 활성제, 안정화제, 피로포스페이트(pyrophosphate)류 및 이산화규소(SiO2)를 포함한다.
본 발명의 일실시예에 따른 상기 점착성 고분자는 치아 점착성을 갖는 역할을 하고, 수화(水和)시 치아 점착성을 나타내는 고분자로서, 하이드록시프로필메틸셀룰로오스, 하이드록시에틸셀룰로오스, 폴리비닐알코올, 폴리비닐피롤리돈류, 하이드록시프로필셀룰로오스, 폴리아크릴산 유도체인 카보머, 메틸 비닐 에테르와 무수 말레인산의 공중합체 및 폴리에틸렌옥사이드로 구성된 군으로부터 선택되는 어느 하나 또는 2 이상인 것을 특징으로 한다.
상기 점착성 고분자의 중량평균 분자량은 100,000 내지 4,000,000인 것이 바람직하며, 더욱 바람직하게는 100,000 내지 2,000,000이다. 만일 상기 점착성 고분자의 분자량이 1000,000 미만이면 통상적인 사용량에서 도포에 적합한 점도에 도달하기 어려우며, 4,000,000을 초과하면 저 농도에서도 점도가 높아 적용이 어려운 문제점이 있다.
상기 점착성 고분자의 함량은, 전체 젤 조성물 100중량부에 대하여, 5 내지 80중량부인 것이 바람직하며, 더욱 바람직하게는 5 내지 60중량부이다. 만일 상기 점착성 고분자의 함량이 5중량부 미만이면 미백성분 층의 점도가 너무 낮아 미백성분 층이 흘러서 시술시 적용하기 어려운 문제가 있고, 80중량부를 초과하면 점도가 너무 높아 작업이 곤란하다.
본 발명의 일실시예에 따른 상기 치아미백 활성물질은 과산화물, 아염소산 금속, 과붕산염, 과탄산염 및 과산화산으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 할 수 있다. 상기 과산화물은 과산화수소, 과산화칼슘 및 카르바마이드 퍼옥사이드(carbamide peroxide)로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 할 수 있으며, 상기 아염소산 금속은 아염소산 칼슘, 아염소산 바륨, 아염소산 마그네슘, 아염소산 리튬, 아염소산나트륨 및 아염소산 칼륨으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 할 수 있다.
상기 치아미백 활성물질의 함량은, 전체 젤 조성물 100중량부에 대하여, 0.1 내지 50중량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 30중량이다. 만일 상기 치아미백 활성물질의 함량이 0.1중량부 미만이면 원하는 미백 효과를 얻을 수 없으며, 50중량부를 초과하면 치아표면 부식, 구강 점막 손상 등의 안전성에 문제가 있게 된다.
본 발명의 일실시예에 따른 상기 치아미백 활성물질의 투과를 촉진하는 물질은 양전하(+)를 띠는 펩타이드인 것을 특징으로 한다. 상기 양전하(+)를 띠는 펩타이드는 LMWP(Low Molecular Weight Protamine)(서열번호1), HIV 단백질의 단백수송 도메인(Protein Transduction Domain, PTD)으로 부터 분리된 TAT(Trans Activator of Transcription)(서열번호2), 페너트라틴(penetratin)(서열번호3), 폴리아르기닌(polyarginine)(서열번호4) 폴리라이신(polylysine)(서열번호5) 및 프로타민(protamine)절편으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 할 수 있다.
대부분의 세포내 약물 전달 기법은 세포 표면의 리셉터를 이용한 엔도사이토시스(endocytosis)에 의존하고 있으나 상기 펩타이드들은 기존의 세포내 물질 수송기전의 하나인 엔도사이토시스와는 전혀 다르게 양전하(+)를 띠고 있으므로 음전하(-)인 세포막의 전하에 의존하여 부착, 전달되는 과정을 거치며 세포내로 이동시키고자 하는 물질들을 직접적으로 단시간 내에 효율적인 도입을 시킬 수 있다는 장점을 가지고 있다.
상기 치아미백 활성물질의 투과를 촉진하는 물질의 함량은 전체 젤 조성물 100중량부에 대하여, 0.1 내지 50중량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 20중량부이다. 만일 상기 치아미백 활성물질의 투과를 촉진하는 물질의 함량이 0.1중량부 미만이면 원하는 투과효과를 얻을 수 없으며, 50중량부를 초과하면 오히려 투과를 방해해 점막을 자극할 위험이 있는 등의 안정성에 문제가 있게 된다.
본 발명의 일실시예에 따른 상기 소수성 계면 활성제는 치아미백용 조성물의 고분자 활강성을 증가시키기 위한 것으로서, 치아미백제에 통상적으로 사용되는 것 을 제한없이 사용할 수 있으며, 바람직하게는 폴리에틸렌글리콜, 프로필렌글리콜, 글리세린, 트리에틸시트레이트, 폴리소르베이트 및 스팬으로 구성된 군으로부터 선택되는 어느 하나 또는 2 이상인 것을 예시 할 수 있다.
상기 소수성 계면 활성제의 함량은 전체 젤 조성물 100중량부에 대하여, 5.0 내지 50중량부인 것이 바람직하다. 만일, 상기 소수성 계면 활성제의 함량이 5.0중량부 미만이면 유연성이 떨어져 사용시 이물감을 주며, 50중량부를 초과하면 제조된 치아미백용 젤의 안정성이 떨어지는 문제점이 있다.
본 발명의 일실시예에 따른 상기 안정화제는 치아미백 활성물질의 안정성을 향상시키기 위한 것으로서, 치아미백제에 통상적으로 사용되는 것을 제한없이 사용할 수 있으며, 바람직하게는 에틸렌디아민 테트라아세트산(ethylenediamine tetra acetic acid)과 그의 염, 소듐 스테네이트(sodium stannate) 및 포타슘 스테네이트(potassium stannate)로 구성된 군에서 선택되는 어느 하나 또는 2 이상의 킬레이트화제 또는 부틸레이티드 하이드록시 톨루엔(butylated hydroxy toluene), 노디하이드로구아이아레틱에시드(nordihydroguaiaretic acid), 프로필 갈레이트(propyl gallate), 트리하이드록시부티로페논(trihydroxybutyrophenone) 및 부틸레이티드 하이드록시아니솔(butylated hydroxyanisol)로 구성된 군에서 선택되는 어느 하나 또는 2 이상의 산화방지제를 예시할 수 있다.
상기 안정화제의 함량은 전체 젤 조성물 100중량부에 대하여, 0.01 내지 5.0중량부인 것이 바람직하다. 만일 상기 안정화제의 함량이 0.01중량부 미만이면 만족스러운 안정화 효과를 얻을 수 없고, 5.0중량부를 초과하면 사용감에 있어서 만 족스럽지 못한 결과를 초래하게 된다.
본 발명의 일실시예에 따른 상기 피로포스페이트(pyrophosphate)류는 타르 제거 및 금속물질의 킬레이트 효과를 가지는 것으로서, 치석방지를 위해 사용되며, 바람직하게는 Na4P2O7, K4P2O7, Na2H2P2O7 및 K2H2P2O7로 구성된 군에서 선택되는 것을 예시할 수 있다.
상기 피로포스페이트류의 함량은 전체 젤 조성물 100중량부에 대하여, 1.0 내지 10.0중량부인 것이 바람직하다. 만일 상기 피로포스페이트류의 함량이 1.0중량부 미만이면 만족스러운 치석방지 효과를 얻을 수 없고, 10.0중량부를 초과하면 오히려 치아점막을 자극할 위험이 있다.
본 발명의 일실시예에 따른 이산화규소(SiO2)는 통상적으로 치아에 부착된 치구, 치석 등을 제거시키고, 치아 표면에 본래의 광택을 주기위한 연마제로 사용되는 물질로서, 나노입자를 사용하는 것이 바람직하다.
상기 이산화규소(SiO2)의 함량은 전체 젤 조성물 100중량부에 대하여, 0.5 내지 0.8중량부인 것이 바람직하다. 만일 상기 이산화규소(SiO2)의 함량이 0.5중량부 미만이면 만족스러운 미백증진 효과를 얻을 수 없고, 0.8중량부를 초과하면 눈에 보이지 않게 치아표면이 너무 많이 깎여 나가 치아가 시리게 되는 문제점이 있다. 또한 본 발명에 따른 치아미백용 젤 조성물은 상기한 성분 이외에 pH조절제, 감미제, 향료, 지각과민 예방제 등의 치아미백용 조성물에 통상적으로 사용되는 첨가제를 더욱 포함할 수 있다.
상기 pH 조절제는 치아미백용 젤 조성물의 장기 보관 안정성을 위한 것으로서, 제 2 인산나트륨, 제 3 인산나트륨, 피로인산나트륨, 피로인산칼륨, 탄산수소나트륨, 수산화칼륨, 수산화암모늄, 트리에탄올아민, 구연산, 구연산나트륨 및 수산화나트륨으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 예시할 수 있고, 상기 치아미백용 젤 조성물의 pH는 4.0 내지 11.0인 것이 바람직하다. 또한 상기 감미제로는 소르비톨, 만니톨, 자일리톨, 스테비오사이드 및 사카린으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 사용할 수 있고, 상기 향료는 멘톨, 페퍼민트오일, L-멘톨 및 계피산으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 사용할 수 있다. 또한 상기 지각과민 예방제는 질산칼륨(Potassium Nitrate), 플루오르화주석(Stannous fluoride, SnF2), 모노플루오르인산나트륨(Sodium monofluoride phosphate, MFP), 염화불소(Sodium fluoride, NaF), 규불화소다(Sodium silicofluoride, Na2SiF6) 및 수산화칼슘(Ca(OH)2)으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 사용할 수 있다. 그리고, 상기한 조성성분을 용해시키기 위한 용매로는 에탄올, 물 및 이들의 혼합액(9:1~6:4 v/v%)을 사용할 수 있다.
본 발명에 따른 치아미백용 액상 조성물은 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 안정화제, 치아 코팅용 점착성 고분자 및 휘발성 용매를 포함한다.
본 발명에 따른 상기 치아미백용 액상 조성물의 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질 및 안정화제는 상기 치아미백용 젤 조성물에서 사용된 것과 동일하며, 상기 치아미백 활성물질의 함량은, 전체 액상 조성물 100중량부에 대하여, 0.1 내지 50중량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 30중량이다. 만일 상기 치아미백 활성물질의 함량이 0.1중량부 미만이면 원하는 미백 효과를 얻을 수 없으며, 50중량부를 초과하면 치아표면 부식, 구강 점막 손상 등의 안전성에 문제가 있게 된다.
상기 치아미백 활성물질의 투과를 촉진하는 물질의 함량은 전체 액상 조성물 100중량부에 대하여, 0.1 내지 50중량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 20중량부이다. 만일 상기 치아미백 활성물질의 투과를 촉진하는 물질의 함량이 0.1중량부 미만이면 원하는 투과효과를 얻을 수 없으며, 50중량부를 초과하면 오히려 투과를 방해해 점막을 자극할 위험이 있는 등의 안정성에 문제가 있게 된다. 상기 안정화제의 함량은 전체 액상 조성물 100중량부에 대하여, 0.01 내지 5.0중량부인 것이 바람직하다. 만일 상기 안정화제의 함량이 0.01중량부 미만이면 만족스러운 안정화 효과를 얻을 수 없고, 5.0중량부를 초과하면 사용감에 있어서 만족스럽지 못한 결과를 초래한다.
본 발명의 일실시예에 따른 상기 치아 코팅용 점착성 고분자는 점착성을 가지며, 치아표면에 코팅되어 필름을 형성하는 고분자로써, 12 내지 120의 K값을 갖는 폴리비닐피롤리돈(Polyvinylpyrrolidone)을 사용할 수 있으며, K값이 90인 폴리 비닐피롤리돈을 사용하는 것이 바람직하다. 상기 K값은 분자량의 길이(중합도)에 비례해서 일반적으로 나타내는 수치를 의미하며, 분자량이 높아질수록 점도 또한 커지고, K값도 비례해서 커진다. K 값에 대하여는 미국약전에 기술되어 있고, 물리화학적 성질에 관해서는 미국약전 및 Handbook of Pharmaceutical Exipients를 참고할 수 있다.
만일 K값이 12 미만인 폴리비닐피롤리돈을 사용할 경우 점착성이 낮아, 사용상 불편함을 초래할 우려가 있고, K값이 120을 초과하는 폴리비닐피롤리돈을 사용할 경우 이 역시 사용상 불편함을 초래할 우려가 있을 뿐만 아니라, 미백활성물질의 방출이 쉽게 이루어지지 않아, 만족스러운 미백효과를 얻을 수 없다.
상기 치아 코팅용 점착성 고분자의 함량은, 전체 액상 조성물 100중량부에 대하여, 0.1 내지 50중량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 20중량부이다. 만일 상기 치아 코팅용 점착성 고분자의 함량이 0.1중량부 미만이면 원하는 점착성의 효과를 낼 수 없으며 50중량부를 초과하면 점도가 너무 높아져 사용성에 있어서 만족할 만한 결과를 초래하고 또한 치아미백 활성 물질의 방출이 잘 이루어지지 않는 문제점을 초래한다.
본 발명의 일실시예에 따른 상기 휘발성 용매는 상기 치아 코팅용 점착성 고분자를 용해시키기 위한 것으로서, 에탄올(Ethanol), 메탄올(Methanol) 및 부탄올(Butanol)로 구성된 군에서 선택되는 하나 또는 2 이상인 것을 사용할 수 있다. 상기 휘발성 용매의 함량은 전체 액상 조성물 100중량부에 대하여, 0.1 내지 70중 량부인 것이 바람직하며, 더욱 바람직하게는 0.5 내지 60중량부이다.
또한 상기 치아미백용 액상 조성물은 상기한 성분 이외에 pH조절제, 감미제, 향료, 지각과민 예방제, 안정화제 등의 치아미백용 조성물에 통상적으로 사용되는 첨가제를 더욱 포함할 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로서, 하기 실시예들은 여러가지 다른 형태로 변형될 수 있으므로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지는 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
[실시예 1] 치아미백용 젤 조성물 제조
1-1: 치아미백용 젤 조성물 제조
점착성 고분자인 카보머 고분자(carbomer 934, carbomer 504, carbomer 505, carbomer 501, carbomer 940 및 carbomer 941)(SIMITOMO SEIKA, Japan) (noveon, USA) 0.4g을 1N NaOH 4㎖에 각각 용해시키고, 24시간동안 방치한 후, 글리세린(glycerin) 3㎖ 및 스팬(span)80(sigma chemical, USA) 0.1㎖를 첨가하였다. 다음으로, 치아미백활성물질인 카르바마이드퍼옥사이드(carbamide peroxide)(sigma-aldrich, USA)를 전체 중량의 8.05% 첨가하여 치아미백용 젤을 만들었다. 상기 젤에 지각과민 방지 효과가 있는 Potassium Nitrate(DAEJUNG, Korea) 0.1g 및 pH 조절 목적과 미백 효과를 위한 sodium citrate(JUNSEI, Japan) 0.1g을 첨가하였다. 그리고 불안정한 과산화수소의 분해를 막기 위하여 안정화제로써 소듐 스테네이트(sodium stannate)(Williamblythe, England) 0.1g 및 소듐에시드 피로포스페이트(sodium acid pyrophosphate) 0.1g를 첨가하였다. 그리고 산화방지제로써 에틸렌 디아민 테트라 아세트산(EDTA)(Fluka, USA)를 0.1g첨가하고 산화방지효과와 미백효과를 내는 소듐 퍼보레이트(sodium perborate)(sigma-aldrich, USA)를 0.1g 첨가하였다. 여기에 향료인 L-Mentol (TIEN YUAN CHEMICAL, Singapore)를 전체 중량의 0.1%를 첨가한 다음, 골고루 섞어 치아미백용 젤 조성물을 제조하였다.
1-2: 치아미백 활성물질의 투과를 촉진하는 물질의 첨가
상기 1-1에서 제조된 젤 조성물에 치아미백 활성물질의 치아표면투과율을 현저히 증가시킬 수 있는 치아미백 활성물질의 투과를 촉진하는 물질로써, LMWP(서열번호1)를 전체 치아미백용 젤 중량 조성물의 0.5%, 1%, 2%, 및 5%의 양으로 첨가하였다.
1-3: 이산화규소(SiO
2
)의 첨가
상기 1-2에서 제조된 젤 조성물에, 치아에 부착된 치구, 치석 등을 제거하며, 치아 면에 본래의 광택을 주는 작용을 하여 연마제로 사용되는 10nm 크기의 SiO2(Sigma, USA)를 첨가하여, 본 발명의 치아미백용 젤 조성물을 제조하였다.
[실시예 2] 치아미백용 액상 조성물 제조
2-1: 치아미백용 액상 조성물 제조
치아 코팅용 점착성 고분자인 폴리비닐 피롤리돈(Polyvinyl pyrrolidone, K=90) (D-BASF, Germany) 7.5g을 70% 에탄올(Hayman, England) 50ml에 용해시킨 후, 글리세린(Glycerin) 2ml을 첨가하였다. 다음으로, 치아미백활성 물질인 카르바마이드퍼옥사이드(carbamide peroxide) (Sigma-Aldrich, USA)를 전체 중량의 8.05%, 향료인 L-Mentol (TIEN YUAN CHEMICAL, Singapore)를 전체 중량의 0.1% 만큼 첨가한 후, 혼합하여 코팅형 치아미백용 액상 조성물을 제조하였다.
2-2: 치아미백 활성물질의 투과를 촉진하는 물질의 첨가
상기 2-1에서 제조된 코팅형 치아미백용 액상 조성물에 치아미백활성 물질의 치아표면투과율을 현저히 증가시킬 수 있는 치아미백 활성물질의 투과를 촉진하는 물질로써, LMWP(서열번호1)를 전체 액상 조성물 중량의 0.5% 양으로 첨가하였다.
[실험예 1] 치아미백용 젤 조성물에 적합한 계면활성제의 선택
수계와의 접촉을 피하고, 입안에서 젤이 침에 의해 침식되는 것을 보호하여 미백 효과를 장기적으로 유지할 수 있도록 하기 위하여 소수성 계면활성제를 치아미백용 젤 조성물에 첨가하였다.
소수성 계면활성제를 사용하지 않고, 카보머 고분자 carbomer 940 0.4g을 사용한 것을 제외하고는 상기 실시예 1과 동일한 방법으로 치아미백용 젤 조성물을 제조하였다. 제조된 치아미백용 젤 조성물에 Polysorbate 20, Polysorbate 60, Polysorbate 80, Propylene Glycol 및 Span 80을 각각 첨가하여 치아미백용 젤을 만들었고, 제조된 젤들을 이용하여 과산화수소 방출량 테스트를 시행하였다.
즉, 상기 각각의 샘플들을 유리 바이알(vial)에 2㎖씩 담고, 여기에 1㎖의 증류수(distilled water)를 넣은 다음, 1시간 후에, 상기 유리 바이알에 0.5 ㎖의 증류수를 넣어 피펫팅(pipetting)한 후, 0.5㎖의 용액을 취해 tube에 각각 담았다. 과산화수소 방출량 테스트를 위한 표준곡선(standard curve)를 얻기 위하여, 900μM H202 을 단계별로 희석하여 0μM, 150μM, 300μM, 450μM, 600μM 및 900μM의 농도로 standard 용액을 만들었다.
그리고, 2.5M 질산칼륨(Potassium Nitrate) 100uL 및 10mM 페러스 암모늄 설페이트(Ferrous Ammonium Sulfate) 200uL를 첨가하였다. 이때 용액의 색깔이 무색에서 붉은색으로 변하게 되는데, 이때 표준(standard) 용액과 각 샘플에서 200uL를 취하여 96well 에 담은 후, 흡광도를 467nm에서 측정하였다. 상기와 같은 방법으로 3hr, 5hr, 7hr 및 1day에서도 측정하였다(도 1). 도 1은 본 발명에 따른 치아미백용 젤 조성물의 소수성 계면활성제의 종류에 따른 과산화수소 방출량의 정도를 분석한 결과를 나타낸 그래프로서, 도 1에 나타난 바와 같이, span80을 넣은 치아미백용 젤의 과산화수소 방출량이 가장 큰 것으로 나타났다.
[실험예 2] 치아미백 활성물질의 투과를 촉진하는 물질의 실활치 테스트
실시예 1의 1-1에서 제조된 치아미백용 젤 조성물에 전체 치아미백용 젤 조성물 중량의 0.5%, 1%, 2% 및 5%의 양의 치아미백 활성물질의 투과를 촉진하는 물질(LMWP)을 첨가하였다. 실험에 사용된 실활치는 서울대학교 치과병원에서 치료에 의해 발치되어 더 이상 사용하지 못하는 치아를 공급받아서 사용하였다. 실활치는 비커에 담아 증류수(distilled water)를 채운 다음, 초음파 세척기(sonicator)에서 5회 세척하였다. 세척된 치아는 실험을 위해서 깨끗이 건조된 상태로 이용하였다. 치아미백 활성물질의 투과를 촉진하는 물질(LMWP)을 전체 치아미백용 젤 중량의 0.5%, 1%, 2% 및 5%의 양을 첨가하여 만든 젤과 치아미백 활성물질의 투과를 촉진하는 물질의 효과를 보기 위하여 나머지 기본조성은 모두 동일하지만 치아미백 활성물질의 투과를 촉진하는 물질을 넣지 않은 젤도 제조하였다.
각각의 젤 2㎖를 각각의 well에 넣은 후, 여기에 치아의 에나멜(enamel) 부분을 박은 후, 각각 NT, 30min, 1hr 및 2hr 동안 실험을 하였다. 치아의 shade 변화 정도는 VITAPAN 3D-MASTER shade guide(VITA Zahnfabrik H.Rauter GmbH & Co. KG, Germany)를 이용하여 실험하였다. 도 2는 치아미백 활성물질의 투과를 촉진하는 물질을 넣지 않은 젤을 처리한 군의 사진이고, 도 3은 치아미백 활성물질의 투과를 촉진하는 물질을 0.5%를 넣은 젤을 처리한 군의 사진, 도 4는 치아미백 활성물질의 투과를 촉진하는 물질을 1%를 넣은 젤을 처리한 군의 사진, 도 5는 치아미백 활성물질의 투과를 촉진하는 물질을 2%를 넣은 젤을 처리한 군의 사진, 도 6은 치아미백 활성물질의 투과를 촉진하는 물질을 5%를 넣은 젤로 처리한 군의 사진이다. 그 결과, 치아미백 활성물질의 투과를 촉진하는 물질을 0.5% 넣은 젤이 치아미 백 활성물질의 투과를 촉진하는 물질을 넣지 않은 젤과 비교하였을 때, 가장 효과가 좋은 것을 알 수 있었다. 이는 치아미백 활성물질의 투과를 촉진하는 물질은 치아미백 물질의 투과를 촉진시켜 주지만, 많은 양을 넣었다고 해서 그 효과가 양에 비례하여 커진 것은 아니라는 것을 나타내는 것이다. 도 7은 치아미백용 젤 조성물에 들어간 치아미백 활성물질의 투과를 촉진하는 물질의 함량 차이에 따른 치아의 색상 변화의 정도를 수치화한 그래프이다.
[실험예 3] 이산화규소(SiO 2 ) 첨가량에 따른 실활치 테스트
이산화규소(SiO2)의 미백 증진효과를 측정하기 위하여 치아미백 활성물질의 투과를 촉진하는 물질(LMWP)을 실시예 1의 1-1에서 제조된 치아미백용 젤 조성물 중량의 0.5%, 0.8%의 양을 첨가하고, 이와 동량으로 10nm 크기의 이산화규소(SiO2) 나노 입자를 첨가하여 치아미백용 젤을 만들었다. 그리고 치아미백 활성물질의 투과를 촉진하는 물질 및 이산화규소(SiO2) 모두를 넣지 않은 젤과 이산화규소(SiO2) 0.5% 및 0.8%를 넣어 만든 치아미백용 젤을 만든 후, 이 젤들을 실활치에 테스트하였다. 실활치는 실험예 2와 마찬가지로 깨끗이 세척하여 건조한 후 실험에 사용하였다.
도 8은 치아미백 활성물질의 투과를 촉진하는 물질과 이산화규소(SiO2)를 넣지 않은 젤을 처리한 군이고, 도 9는 치아미백 활성물질의 투과를 촉진하는 물질 0.5%와 이산화규소(SiO2) 0.5%를 넣은 젤을 처리한 군, 도 10은 치아미백 활성물질의 투과를 촉진하는 물질을 0.8%와 이산화규소(SiO2) 0.8%를 넣은 젤을 처리한 군이고, 도 11은 이산화규소(SiO2) 0.5%를 넣은 젤을 처리한 군, 도 12는 이산화규소(SiO2) 0.8%를 넣은 젤을 처리한 군이다. 그 결과, 치아미백 활성물질의 투과를 촉진하는 물질 0.5%와 이산화규소(SiO2) 0.5%를 넣은 젤을 처리한 군의 효과가 가장 뛰어남을 알 수 있었다. 또한, 이산화규소(SiO2)는 치아미백 활성물질의 투과를 촉진하는 물질과 마찬가지로 많이 넣었다고 해서 그 효과가 양에 비례하여 커진 것은 아님을 알 수 있었다. 도 13은 치아미백 활성물질의 투과를 촉진하는 물질과 이산화규소(SiO2)의 함량에 따른 치아색상 변화의 정도를 그래프로 나타낸 것이다.
[실험예 4] 치아 코팅용 점착성 고분자를 첨가한 치아미백용 액상 조성물의 실활치 테스트
치아미백 활성물질의 전달을 촉진 시켜주는 물질을 포함하지 않은 실시예 2-1에서 제조된 치아미백용 액상 조성물 및 치아미백 활성물질의 전달을 촉진 시켜주는 물질을 포함하는 실시예 2-2에서 제조된 치아미백용 액상 조성물에 대한 실활치 테스트를 실험예 2와 동일한 방법으로 수행하였다. 도 14는 치아미백 활성물질의 투과를 촉진하는 물질을 포함하지 않는 치아미백용 액상 조성물을 처리한 군의 사진이고, 도 15는 치아미백 활성물질의 투과를 촉진하는 물질을 포함하는 치아미백 용 액상 조성물을 처리한 군의 사진이다. 그 결과, 치아미백 활성물질의 투과를 촉진하는 물질을 0.5% 넣은 액상 조성물(실시예 2-1)이 치아미백 활성물질의 투과를 촉진하는 물질을 넣지 않은 액상 조성물(실시예 2-2)보다 치아미백 효과가 더 좋은 것을 알 수 있었다.
이상에서 상세히 설명하고 입증하였듯이, 본 발명에 따르면, 점착력을 가지는 고분자 내에 치아미백 활성물질, 치아미백 활성물질의 투과를 촉진하는 물질, 소수성 계면활성제 및 치아에 부착된 치구, 치석 등을 제거하며 치아 면에 본래의 광택을 주는 작용을 하여 연마제로 사용되는 이산화규소(SiO2)의 나노입자가 첨가되어 종래의 치아미백제에 비해 미백성분의 투과율이 증가되고, 미백 효과가 뛰어난 치아미백용 조성물을 제공하는 효과가 있다. 즉, 본 발명의 치아미백용 조성물은, 종래의 치아미백제에 비해 투과율이 현저히 증가되었고, 미백 효과를 장기적으로 유지할 수 있어, 기존 미백제의 사용감은 그대로 유지하면서 미백 효과는 신속하고, 장기적으로 유지할 수 있다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

- (a) 점착성 고분자, (b) 과산화물, 아염소산 금속, 과붕산염, 과탄산염 및 과산화산으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백 활성물질, (c) 상기 치아미백 활성물질의 투과를 촉진하는 양전하(+)를 띠는 펩타이드, (d) 소수성 계면 활성제, (e) 안정화제, (f) 피로포스페이트(pyrophosphate)류 및 (g) 이산화규소(SiO2)를 포함하는 치아미백용 젤 조성물.

- 제1항에 있어서, 상기 안정화제는 에틸렌디아민 테트라아세트산(ethylenediamine tetra acetic acid)과 그의 염, 소듐 스테네이트(sodium stannate) 및 포타슘 스테네이트(potassium stannate)로 구성된 군에서 선택되는 어느 하나 또는 2 이상의 킬레이트화제 또는 부틸레이티드 하이드록시 톨루엔(butylated hydroxy toluene), 노디하이드로구아이아레틱에시드(nordihydroguaiaretic acid), 프로필 갈레이트(propyl gallate), 트리하이드록시부티로페논(trihydroxybutyrophenone) 및 부틸레이티드 하이드록시아니솔(butylated hydroxyanisol)로 구성된 군에서 선택되는 어느 하나 또는 2 이상의 산화방지제인 것을 특징으로 하는 치아미백용 젤 조성물.
- 제1항에 있어서, 상기 피로포스페이트(pyrophosphate)류는 Na4P2O7, K4P2O7, Na2H2P2O7 및 K2H2P2O7로 구성된 군에서 선택되는 것을 특징으로 하는 치아미백용 젤 조성물.
- 제1항에 있어서, 전체 젤 조성물 100중량부에 대하여, 점착성 고분자 5 내지 80중량부, 치아미백 활성물질 0.1 내지 50중량부, 치아미백 활성물질의 투과를 촉진하는 양전하(+)를 띠는 펩타이드 0.1 내지 50중량부, 소수성 계면 활성제 5.0 내지 50중량부, 안정화제 0.01 내지 5.0중량부, 피로포스페이트(pyrophosphate)류 1.0 내지 10.0중량부 및 이산화규소(SiO2) 0.5 내지 0.8중량부를 함유하는 치아미백용 젤 조성물.
- 제1항, 제2항, 제3항, 제5항, 제6항, 제8항, 제9항, 제10항, 제11항 또는 제12항중 어느 한 항에 있어서, pH 조절제, 감미제, 향료 및 지각과민 예방제로 구성된 군에서 선택되는 어느 하나 또는 2 이상을 추가로 포함하는 것을 특징으로 치아미백용 젤 조성물.
- 제13항에 있어서, 상기 pH 조절제는 제 2 인산나트륨, 제 3 인산나트륨, 피로인산나트륨, 피로인산칼륨, 탄산수소나트륨, 수산화칼륨, 수산화암모늄, 트리에탄올아민, 구연산, 구연산나트륨 및 수산화나트륨으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백용 젤 조성물.
- 제13항에 있어서, 상기 치아미백용 젤 조성물의 pH는 4.0 내지 11.0인 것을 특징으로 하는 치아미백용 젤 조성물.
- 제13항에 있어서, 상기 감미제는 소르비톨, 만니톨, 자일리톨, 스테비오사이드 및 사카린으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백용 젤 조성물.
- 제13항에 있어서, 상기 향료는 멘톨, 페퍼민트오일, L-멘톨 및 계피산으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백용 젤 조성물.
- 제13항에 있어서, 상기 지각과민 예방제는 질산칼륨(Potassium Nitrate), 플루오르화주석(Stannous fluoride, SnF2), 모노플루오르인산나트륨(Sodium monofluoride phosphate, MFP), 염화불소(Sodium fluoride, NaF), 규불화소다(Sodium silicofluoride, Na2SiF6) 및 수산화칼슘(Ca(OH)2)으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백용 젤 조성물.
- (a) 과산화물, 아염소산 금속, 과붕산염, 과탄산염 및 과산화산으로 구성된 군에서 선택되는 어느 하나 또는 2 이상인 것을 특징으로 하는 치아미백 활성물질, (b) 상기 치아미백 활성물질의 투과를 촉진하는 양전하(+)를 띠는 펩타이드, (c) 안정화제, (d) 치아 코팅용 점착성 고분자 및 (e) 휘발성 용매를 포함하는 치아미백용 액상 조성물.
- 제19항에 있어서, 상기 치아 코팅용 점착성 고분자는 12 내지 120의 K값을 갖는 폴리비닐피롤리돈(Polyvinylpyrrolidone)인 것을 특징으로 하는 치아미백용 액상 조성물.
- 제19항에 있어서, 상기 휘발성 용매는 에탄올, 메탄올 및 부탄올로 구성된 군에서 선택되는 하나 또는 2 이상인 것을 특징으로 하는 치아미백용 액상 조성물.
- 제19항에 있어서, 상기 치아미백용 액상 조성물은 pH조절제, 감미제, 향료, 지각과민 예방제, 안정화제로 구성된 군에서 선택되는 어느 하나 또는 2 이상을 추가로 포함하는 것을 특징으로 치아미백용 액상 조성물.
- 제19항에 있어서, 전체 액상 조성물 100 중량부에 대하여, 치아미백 활성물질 0.1 내지 50중량부, 치아미백 활성물질의 투과를 촉진하는 양전하(+)를 띠는 펩타이드 0.1 내지 50중량부, 안정화제 0.01 내지 5.0중량부, 치아 코팅용 점착성 고분자 0.1 내지 50중량부, 휘발성 용매 0.1 내지 70중량부를 함유하는 치아미백용 액상 조성물.
