Claims (22)
- 탄소원으로서 단당류 및 이당류로 이루어진 군에서 선택된 2종 이상의 당류가 혼합된 혼합당이 첨가된 식물세포배양용 배지를 사용하여 식물세포를 배양하여, 상기 식물세포에서의 이차대사산물의 생산성을 향상키는 것을 특징으로 하는, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 혼합당은 포도당, 과당, 만노오스, 리보오스, 아라비노오스, 자일로오스, 갈락토오스, 자당, 멜리비오스, 트레할로오스, 셀로비오스, 락토오스, 및 말토오스로 이루어진 군에서 2종 이상의 당류가 혼합된 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 혼합당의 함량이 전체배지에 대해서 2 내지 12 %(v/w)인 것을 특징으로 하는, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 혼합당이 포도당과 과당이 1:5 내지 5:1의 중량비로 혼합되어 있는 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 혼합당이 자당과 과당이 1:4 내지 4:1의 중량비로 혼 합되어 있는 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 배지는 하기의 배지들로 이루어진 군 중에서 선택된 배지로부터 탄소원이 배제되고 포도당, 과당, 만노오스, 리보오스, 아라비노오스, 자일로오스, 갈락토오스, 자당, 멜리비오스, 트레할로오스, 셀로비오스, 락토오스, 및 말토오스로 이루어진 군에서 선택된 2종 이상의 당류가 혼합된 혼합당이 전체 배지에 대하여 2 내지 12 %(v/w)의 양으로 첨가된 것인, 식물세포배양에 의한 이차대사산물의 생산 방법:앤더슨 로도덴드론 배지(Anderson rhododendron medium), 씨에이치유 배지(CHU(N6) medium), 씨엘씨 이포모에 배지(CLC/Ipomoea medium), 체-풀 비티스 배지(Chee & Pool (C2D) vitis medium), 데 그리프-야곱 배지(De greef & jacobs medium), 디케이더블유-정글란 배지(DKW/JUNGLANS medium), 에릭슨 배지(Eriksson(er) medium), 갬보그 배지(Gamborg B5 medium), 그리숍-도이 배지(Gresshof & doy (DBM2) medium), 헬러 배지(Hellers medium), 카오 미카이룩(kao michayluk medium), 운드슨 코키드 배지(knudson corchid medium), 린더만 오키드 배지(Lindemann Orchid medium), 리트베이 배지(Litvay medium), 린스마이어-스쿡 배지(Linsmaier & Skoog medium), 맥카운 우디플랜트 배지(McCowns woody plant medium), 무라시케-스쿡 배지(Murashige & Skoog medium), 무라시케- 밀러 배지(murashige & Miller medium), 니취 배지(nitsch medium), 엔엘엔 배지(NLN medium), 오키맥스 배지(orchimax medium), quoirin & Lepoivre medium, rugini olive medium, schenk & hildebrandt medium, 에스 배지(S-Medium), 베이신-웬트 배지(vacin and went medium), 화이트 배지(white medium), 웨스트바코 배지(westvaco WV3 medium) 및 이들의 변형 배지.
- 제1항에 있어서, 상기 식물세포는 택서스 바카타 (Taxus bacata), 택서스 브레비폴리아 (Taxus brevifolia), 택서스 카나덴시스 (Taxus canadensis), 택서스 취넨시스 (Taxus chinensis), 택서스 쿠스피다타 (Taxus cuspidata), 택서스 플로리다나 (Taxus floridana), 택서스 글로보사 (Taxus globosa), 택서스 메디아(Taxus media), 택서스 월리치아나 (Taxus wallichiana), 및 택서스 유나넨시스 (Taxus yunnanensis)로 이루어진 군으로부터 선택된 식물로부터 유래된 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제 1 항에 있어서, 상기 이차대사산물은 파클리탁셀 및 택센으로 이루어진 군 중에서 선택된 한 가지 이상의 화합물인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 식물세포의 배양은 회분식 배양 (batch culture), 연속식 배양 (continuous culture), 유가식 배양 (fed-batch culture), 반연속식 회분 배양 (semi-continuous batch process), 고정화 배양 (immobilized culture), 이원상 배양 (two-phase culture), 및 이단계 배양(two stage culture)으로 이루어 진 군 중에서 선택된 식물세포배양 방법에 의하여 수행되는 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항에 있어서, 상기 식물세포의 배양이(i) 식물세포를 상기 혼합당을 첨가한 배지에서 20℃ 내지 25℃의 온도조건 하에서 배양하고,(ii) 배양된 식물세포가 대수기에 이르렀을 때, 배양온도를 26℃ 내지 32℃로 변화시켜 배양을 계속 진행하여 수행되는 것을 특징으로 하는,식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 상기 탄소원으로서 혼합당이 첨가된 식물세포배양용 배지를 배양 개시시부터 사용하여 식물세포 배양을 수행하는 것을 특징으로 하는, 첨가된 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 상기 탄소원으로 혼합당이 첨가된 식물세포배양용 배양배지를 배양 개시시부터 사용하고, 상기 배지 내 당의 함량이 2% 이하일 때 포도당, 과당, 만노오스, 리보오스, 아라비노오스, 자일로오스, 갈락토오스, 자당, 멜리비오스, 트레할로오스, 셀로비오스, 락토오스, 및 말토오스로 이루어진 군에서 선택된 1종 이상의 당류를 배지내 당농도가 6% 이하가 되도록 추가로 첨가하여 배양을 계속 진행시키는 것을 특징으로 하는, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 상기 식물세포의 배양개시시에는 탄소원으로서 상기의 혼합당 이외의 탄소원을 포함하는 식물세포배양용 배지를 사용하여 배양을 수행하고, 상기 배지 내 당 함량이 2% 이하로 된 때에 상기 혼합당을 추가로 첨가하여 배양을 계속 진행시키는 것을 특징으로 하는, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 상기 식물 세포의 배양시에, 식물세포 생장기, 또는 이차대사산물 생산기, 또는 식물세포 생장기와 이차대사산물 생산기 모두에 상기 혼합당을 첨가하여 식물세포 배양을 수행하는 것을 특징으로 하는, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 제14항에 있어서, 상기 혼합당의 첨가 시기가 식물세포 생장기와 이차대사산물 생산기 모두인 경우, 상기 두 시기에 첨가되는 혼합당이 서로 상이한 것인, 식물세포배양에 의한 이차대사산물의 생산 방법.
- 탄소원으로서 자당과 과당이 1:4 내지 4:1 중량비로 혼합된 혼합당 또는 포도당과 과당이 1:5 내지 5:1 중량비로 혼합된 혼합당을 전체 배지에 대하여 2 내지 12 %(v/w)의 양으로 포함하는 것을 특징으로 하는, 식물세포에 의한 이차대사산물 생산용 배지.
