Claims (21)
- 세포 표면발현 모체(cell surface anchoring motif)로서 탄저균(Bacillus anthracis) 포자외막 단백질(BclA)의 단편을 코딩하는 서열번호 1, 서열번호 2 및 서열번호 3으로 구성된 군에서 선택되는 bclA 유전자 단편을 함유하고, 프로모터와 목적단백질을 코딩하는 유전자를 함유하며, 상기 목적단백질을 코딩하는 유전자가 숙주세포 내에서 발현될 경우, 상기 목적단백질이 상기 BclA의 단편과 융합된 형태로 세포 표면에 발현되도록 구성된 발현벡터.

- 제1항에 있어서, 상기 프로모터는 tac 프로모터인 것을 특징으로 하는 발현벡터.

- 제1항 또는 제5항의 발현벡터를 그람음성균, 그람양성균, 방선균, 효모 및 곰팡이로 구성된 군에서 선택되는 세포에 도입하여 수득되는 형질전환된 미생물.
- 제7항에 있어서, 형질전환에 사용된 미생물은 목적단백질의 세포 표면발현에 유리하도록, 발현된 목적단백질을 분해하는데 관련된 세포 내 또는 세포 외의 단백질 분해효소를 생산하지 못하도록 변형된 것임을 특징으로 하는 형질전환된 미생물.
- 제8항에 있어서, 상기 미생물은 대장균(Escherichia coli)인 것을 특징으로 하는 형질전환된 미생물.
- 제7항의 형질전환된 미생물을 배양하여 목적단백질을 세포 표면에 발현하는 단계 및 목적단백질이 표면에 발현된 세포를 회수하는 단계를 포함하는 목적단백질의 세포 표면발현방법.
- 제10항에 있어서, 상기 목적단백질은 호르몬, 호르몬 유사체, 효소, 효소저해제, 신호전달 단백질, 항체, 단일사슬항체, 결합단백질, 결합도메인, 펩타이드, 항원, 부착단백질, 구조단백질, 조절단백질, 독소단백질, 사이토카인, 전사조절 인자, 혈액응고 인자 및 식물 생체방어 유도 단백질로 구성된 그룹으로부터 선택되는 어느 하나인 것을 특징으로 하는 방법.
- 제11항에 있어서, 상기 목적단백질은 스트렙타비딘인 것을 특징으로 하는 방법.
- 제11항에 있어서, 상기 효소는 리파아제인 것을 특징으로 하는 방법.
- 제11항의 방법에 의하여 제조되고, 효소활성을 갖는 목적단백질이 표면에 발현된 세포를 이용하는 것을 특징으로 하는 생물전환 방법.
- 제11항의 방법에 의하여 제조되고, 목적단백질이 표면에 발현된 세포를 기질 표면에 고정화시키는 것을 특징으로 하는 단백질 어레이의 제조방법.
- 제11항의 방법에 의하여 제조되고, 항원이 표면에 발현된 세포를 인간을 제외한 척추동물에 투여하여 면역반응을 유도하고, 상기 면역반응에 의해 생성된 항체를 회수하는 것을 특징으로 하는 척추동물에서 항체를 제조하는 방법.
- 리파제를 이용하여 라세믹 에스터 화합물로부터 키랄성 에스터, 키랄성 유기산 또는 키랄성 알코올로 광학분할하는 키랄 화합물의 제조방법에 있어서, 제13항의 방법에 의하여 제조된 세포 표면에 발현된 리파아제를 이용하는 것을 특징으로 하는 방법.
- 탄저균(Bacillus anthracis) 유래 포자외막 단백질(BclA)의 단편을 코딩하는 서열번호 1, 서열번호 2 및 서열번호 3으로 구성된 군에서 선택되는 bclA 유전자 단편 및 스트렙타비딘을 코딩하는 유전자를 포함하는 발현벡터에 의하여 형질전환된 미생물을 유효성분으로 포함하는 전세포 바이오센서 매개체.
- 제18항에 있어서, 미생물은 대장균(Escherichia coli)인 것을 특징으로 하는 전세포 바이오센서 매개체.
- 다음의 단계를 포함하는 목적단백질의 개량방법:(a) 목적단백질을 코딩하는 유전자의 변이체 라이브러리를 구축하는 단계;(b) 상기 목적단백질의 변이체와 탄저균 포자외막 단백질(BclA)의 단편을 코딩하는 서열번호 1, 서열번호 2 및 서열번호 3으로 구성된 군에서 선택되는 bclA 유전자 단편을 함유하는 유전자 재조합체를 제작하는 단계;(c) 상기 유전자 재조합체 또는 상기 유전자 재조합체를 포함하는 벡터로 그람음성균, 그람양성균, 방선균, 효모 및 곰팡이로 구성된 그룹으로부터 선택되는 숙주세포를 형질전환하는 단계;(d) 상기 형질전환된 숙주세포를 배양하여 상기 유전자 변이체 라이브러리를 세포 표면에 발현하는 단계; 및(e) 개량된 형질을 갖는 목적단백질이 발현된 세포를 스크리닝하는 단계.
- 제20항에 있어서, 상기 스크리닝 단계는 목적단백질의 활성, 목적단백질에 라벨링된 물질을 인식하는 단백질, 목적단백질에 결합하는 라벨링된 리간드 또는 목적단백질에 특이적으로 결합하는 항체를 이용하는 것을 특징으로 하는 목적단백질의 개량방법.
