Claims (8)
- 단백질을 포함하는 생물학적 시료에 하기 화학식 I의 화합물:[화학식 I][식 중, R1, R2, 및 R3는 각각 독립적으로 수소원자 또는 치환 또는 비치환된 탄소수 1 내지 6의 알킬기이며, a는 1 내지 6의 정수이다 (단, 아민은 2개 이상의 알킬기를 포함한다)];및 단백질 핵화 물질(nucleating agent)을 첨가하는 단계;상기 혼합물을 소수성 표면 물질로 처리하는 단계; 및상기 단백질이 결합된 소수성 표면 물질로부터 시료를 분리하는 단계를 포함하는 생물학적 시료 중의 핵산을 유지하면서 단백질을 제거하는 방법.
- 제 1 항에 있어서, 상기 화학식 I의 화합물은 베타인, 카르니틴, 및 N,N-디메틸글리신으로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 화학식 I의 화합물의 농도는 0.15M ∼ 2M인 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 단백질 핵화 물질은 트리클로로아세트산(TCA), 황산암모늄, 아세톤, 및 에탄올로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 4 항에 있어서, 상기 트리클로로아세트산의 농도는 0.5 ∼ 5%인 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 소수성 표면 물질은 표면이 소수성을 띠는 고체 지지체(solid support)로서, 상기 고체 지지체는 비드, 슬라이드 글라스, 실리콘 웨이퍼, 막, 및 금속판으로 구성된 군으로부터 선택되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 단백질이 결합된 소수성 표면 물질로부터 시료의 분리는 원심분리, 여과, 및 자성으로 구성된 군으로부터 선택된 방법에 의해 수행되는 것을 특징으로 하는 방법.
- 제 1 항에 있어서, 상기 단백질이 결합된 소수성 표면 물질로부터 시료의 분리는 마이크로채널에 소수성 물질이 코팅된 미세유동장치를 통과하는 시료의 유동 에 의해 수행되는 것을 특징으로 하는 방법.
