Claims (5)
- 염색 폐수 색도 분해 미생물의 배양 방법에 있어서;상기 염색 폐수 색도 분해 미생물은,폐수 방출지역의 토양을 채취한 후 증류수와 1:10의 비율로 현탁한 후 30분 동안 정치시키고, 정치된 현탁액중 2%를 취출하고,상기 취출된 현탁액을 취출된 현탄액 대비 99배의 부피를 가지는 멸군 복합배양 배지에 접종하며,상기 접종된 배지에는, 배지 부피 대비 1/99부피를 가지는 광유를 중층하여 30℃에서 정치 배양하며,상기 배양액을 98well microtiter plate(BECTON DIKINSON)에 순차적으로 희석하여 1.5%agar가 첨가된 영향 한천 평판배지에 도말하여 투명환을 생성하는 간균를 분리하여 얻는 것을 특징으로 하는 염색 폐수 방출 지역의 토양을 기조로 한 색도 분해 미생물의 배양 방법.
- 제 1 항에 있어서;상기 분리된 간균는,그람 염색 작업과,LB평판 배지에 24시간 배양 후 포자 염색 시약(Malachite green, sapranin)을 이용하여 7-10여분 가열 염색하는 포자 염색 및,분리 간균의 동정을 위해 API(Analytical Profile Index)20A를 이용하여 기질 이용능 및 생리 생화학적 검사를 수행하여 확인되는 절대혐기성 간균인 CLOSTRIDIUM spp.인 것을 특징으로 하는 염색 폐수 방출 지역의 토양을 기조로 한 색도 분해 미생물의 배양 방법.
- 제 1 항에 있어서;상기 복합 배양 배지는 2%의 LB배지(DIFCO, Lennox)이고,첨가 염료는 멸균 증류수 1ℓ에 염료 100㎎의 비율로 혼합되어 사용하는 것을 특징으로 하는 염색 폐수 방출 지역의 토양을 기조로 한 색도 분해 미생물의 배양 방법.
- 제 1 항 또는 제 2 항에 있어서;상기 분리된 간균인 CLOSTRIDIUM spp.의 염색 폐수에 잔류하는 색도 분해 작용에 있어서,상기 간균인 CLOSTRIDIUM spp.는 염색 폐수에 투입된 36시간 후에 산성도 pH9-pH12사이의 강산성 조건하에서 3색의 기본 구조인 적색, 황색, 청색의 탈색률이 모두 90% 이상을 발현하는 것을 특징으로 하는 염색 폐수 방출 지역의 토양을 기조로 한 색도 분해 미생물의 배양 방법.
- 간균인 CLOSTRIDIUM spp.를 이용한 염색 폐수 처리 공정은,분리 배양된 간균인 CLOSTRIDIUM spp.를 LB 고체 또는 액체 배지에 접종한 후, 혐기 조건하에서 37℃를 유지하여 24시간 배양하고,염색 폐수에 2XLB 액체 배지를 동일 분량으로 첨가하여 교반하고,교반된 염색 폐수에 6NHCl(염산)을 투입하여 수소이온농도를 10이 되도록 형성하여,상기 전배양된 간균을 전체 염색 폐수 부피의 2%에 해당하는 양을 투입하여 혐기 상태로 37℃의 기온하에서 24시간 배양하는 처리공정을 포함하는 것을 특징으로 하는 염색 폐수 방출 지역의 토양을 기조로 한 색도 분해 미생물을 이용한 염색 폐수 처리 방법.
