Claims (12)
- 소정의 염기 배열을 갖는 제2 핵산쇄를 제2 기판상에 고정한 핵산쇄 고정화담체를 제조하는 방법으로서:제1 기판 상에, 상기 제2 핵산쇄의 염기 배열에 대하여 상보적인 염기 배열을 갖는 제1 핵산쇄를 고정화한 제1 핵산쇄 고정화 담체를 준비하는 공정과,핵산쇄 합성 용액중에 있어서, 상기 제1 핵산쇄 고정화 담체의 제1 핵산쇄에 따라서 이것에 상보적인 배열을 갖는 제2 핵산쇄를 합성하는 공정과,상기 제1 기판의 제1 핵산쇄 측에 대면시켜 상기 제2 기판을 배치하는 공정과, 상기 제2 기판으로부터 상기 제1 기판으로 향하는 전계를 인가함으로써, 상기 1 핵산쇄에 따라서 합성된 제2핵산쇄를 상기 제2 기판 표면에 영동시켜, 상기 제2 기판 표면에 결합시키는 공정을 구비한 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 합성된 상기 제2 핵산쇄를 상기 제2 기판 표면에 영동시켜 상기 제2 기판 표면에 결합시키는 공정은, 전해질 용액중에 있어서 이중쇄 핵산이 단일쇄로 변성하는 조건으로 행하여지는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제2항에 있어서, 상기 전해질 용액 중에서 상기 이중쇄 핵산의 단일쇄로의 변성은 상기 전해질 용액을 가열함으로써 행하는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 상기 제2 기판으로부터 상기 제1 기판으로 향하는 전계를 인가하기 위해서 상기 제2 기판 및 상기 제1 기판의 외측에 한쌍의 전극을 설치하고 이 전극쌍에 전위를 가하는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제4항에 있어서, 상기 제2 기판 및 상기 제1 기판의 적어도 한편이 합성 수지 제품 또는 유리 제품의 기판 또는 막인 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 상기 제2 기판으로부터 상기 제1 기판으로 향하는 전계를 인가하기 위해서, 상기 제2 기판 및 상기 제1 기판의 적어도 한쪽에 전극 재료 표면을 갖는 기판 전극을 사용하고 상기 기판 전극에 전위를 가하는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제6항에 있어서, 상기 기판 전극은, 절연 기판 상에 전극막을 피복하고 상기 전극막의 표면을 절연층 영역에서 복수의 전극 영역으로 분리한 것이고, 분리된 각각의 전극 영역에, 각각 다른 배열의 제2 핵산쇄 또는 제1 핵산쇄를 고정하는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제6항에 있어서, 상기 기판 전극의 전극 재료 표면이 금으로 이루어지고 상기 제2 핵산쇄와 상기 제2 기판과의 결합은, 상기 제2 핵산쇄에 결합된 유황과 상기 금 표면의 친화성 결합을 통하여 행하여 지는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 상기 제2 핵산쇄 또는 상기 제1 핵산쇄가 RNA, DNA, PNA 및 이들의 유사 화합물로 이루어지는 군에서 선택되는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 상기 제2 핵산쇄와 상기 제2 기판의 결합은 공유 결합, 친화성 결합 및 정전적 결합으로 이루어지는 군에서 선택되는 결합을 통해 행해지는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 상기 핵산쇄 합성 용액은, 프라이머, 핵산 합성 효소, 뉴클레오티드 모노머 및 전해질을 함유하는 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
- 제1항에 있어서, 상기 제2 핵산쇄를 제2 기판에 고정한 핵산쇄 고정화 담체는 유전자 검사에 사용되고, 상기 제2 핵산쇄는 프로브인 것을 특징으로 하는 핵산쇄 고정화 담체의 제조 방법.
