Claims (85)
- (a) 프로모터에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 구조물을 함유하는 세포를 제공하는 단계, 여기서 리포터 유전자는 전사인자가 프로모터의 소집 부위 및 표적 부위를 모두 인식하는 경우에는 소정 수준을 초과하여 발현되거나 소정 수준 미만으로 발현되지만 전사인자가 프로모터의 소집 부위만을 인식하는 경우에는 그러하지 아니함;(b) (i) 전사 활성화 또는 억제 도메인, (ii) 소집 부위를 인식하는 DNA 결합 도메인, 및 (iii) 시험 징크 핑거 도메인을 포함하는 비천연적 단백질을 코딩하는 다수의 하이브리드 핵산을 제공하는 단계, 여기서 시험 징크 핑거 도메인의 아미노산 서열은 다수의 하이브리드 핵산 서로간에 서로 상이함;(c) 다수의 핵산 중 하나 이상이 하나 이상의 세포에 들어갈 수 있게 하는 조건하에서 다수의 하이브리드 핵산을 상기 세포와 접촉시키는 단계;(d) 세포 내에서 하이브리드 핵산이 발현될 수 있게 하는 조건에서 상기 세포를 유지하는 단계; 및(e) 세포가 표적 부위를 인식하는 시험 징크 핑거 도메인을 코딩하는 하이브리드 핵산을 포함함을 나타내는 지표로서, 리포터 유전자를 소정 수준을 초과하여 발현하거나 소정 수준 미만으로 발현하는 세포를 동정하는 단계를 포함하는, DNA 상의 표적 서열을 인식하는 징크핑거 도메인을 동정하는 방법.

- 제5항에 있어서, 상기 선택성 마커가 대사산물의 합성에 필요한 영양요구성유전자(auxotrophy gene)이고, 상기 세포의 유전체는 이 영양요구성 유전자에 대한 기능적 카피를 갖고 있지 않으며, 상기 (d) 단계에서 상기 세포는 상기 대사산물이 결핍된 배지에서 유지되는 방법.

- (a) 프로모터에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 구조물을 함유하는 세포를 제공하는 단계, 여기서 리포터 유전자는 전사인자가 프로모터의 소집 부위 및 표적 부위를 모두 인식하는 경우에는 소정 수준을 초과하여 발현되거나 소정 수준 미만으로 발현되지만 전사인자가 프로모터의 소집 부위만을 인식하는 경우에는 그러하지 아니함;(b) 도메인 보존된 경계부분을 코딩하는 핵산에 결찰되는 올리고뉴클레오티드 프라이머를 이용하여, 각각 시험 징크핑거 도메인을 코딩하는 다수의 핵산 서열을 증폭시키는 단계;(c) (b)의 각 핵산 서열을 (i) 전사 활성화 또는 억제 도메인을 코딩하는 핵산서열 및 (ii) 소집 부위를 인식하는 DNA 결합 도메인을 코딩하는 핵산 서열에 연결하여 다수의 하이브리드 핵산을 형성하는 단계;(d) 다수의 핵산 중 하나 이상이 하나 이상의 세포에 들어갈 수 있게 하는 조건하에서 다수의 하이브리드 핵산 (c)를 상기 세포 (a)와 접촉시키는 단계;(e) 세포 내에서 하이브리드 핵산이 발현될 수 있게 하는 조건에서 상기 세포를 유지하는 단계; 및(f) 세포가 하이브리드 핵산 (c)을 포함하고 또한 이 하이브리드 핵산이 DNA 상의 표적 부위를 인식하는 징크 핑거 도메인을 코딩함을 나타내는 지표로서, 리포터 유전자를 소정 수준을 초과하여 발현하거나 소정 수준 미만으로 발현하는 세포를 동정하는 단계를 포함하는, DNA 상의 표적 서열을 인식하는 징크핑거 도메인을 동정하는 방법.

- 제21항에 있어서,도메인의 보존된 경계부위를 코딩하는 서열에 결찰하는 올리고뉴클레오티드 프라이머를 이용하여, 유전체 핵산, 전령 RNA(mRNA) 혼합물, 또는 상보적 DNA(cDNA) 혼합물로부터 시험 징크핑거 도메인을 코딩하는 핵산을 증폭하는 단계를 더 포함하는 방법.
- 제21항에 있어서,(i) 서열 데이터베이스에서 후보 징크핑거 도메인의 아미노산 서열을 동정하는 단계;(ii) 상기 후보 징크핑거 도메인의 아미노산을 코딩하는 후보 핵산을 제공하는 단계; 및(iii) 상기 후보 핵산을 이용하여 단계(b)의 다수의 하이브리드 핵산 내에 포함될 하이브리드 핵산을 구축하는 단계를 더 포함하는 방법.
- (a) 프로모터에 작동가능하게 연결된 리포터 유전자를 포함하는 리포터 구조물을 함유하는 제1 세포를 제공하는 단계, 여기서 리포터 유전자는 전사인자가 프로모터의 소집 부위 및 표적 부위를 모두 인식하는 경우에는 소정 수준을 초과하여 발현되거나 소정 수준 미만으로 발현되지만 전사인자가 프로모터의 소집 부위만을 인식하는 경우에는 그러하지 아니함;(b) (i) 전사 활성화 또는 억제 도메인, (ii) 소집 부위를 인식하는 DNA 결합 도메인, 및 (iii) 시험 징크 핑거 도메인을 포함하는 단백질을 코딩하는 하이브리드 핵산을 함유하는 제2 세포를 제공하는 단계;(c) 상기 제1 세포 및 제2 세포를 융합하여 융합된 세포를 형성하는 단계;(d) 세포 내에서 하이브리드 핵산이 발현될 수 있게 하는 조건에서 상기 융합된 세포를 유지하는 단계; 및(e) 융합 세포 내 리포터 유전자의 발현량의 변화를 검출하는 단계(이 때 소정 수준을 초과하는 리포터 유전자의 발현 또는 소정 수준 미만의 발현은 시험 징크 핑거 도메인이 표적 부위를 인식함을 나타냄)를 포함하는, 시험 징크핑거 도메인이 프로모터 상의 표적 부위를 인식하는지 여부를 결정하는 방법.
- 제24항에 있어서, 상기 제1 및 제2 세포가 서로 상반되는 교배형의 효모 세포인 방법.
- (a) 프로모터에 작동가능하게 연결된 리포터 유전자를 포함하는 다수의 리포터 구조물을 제공하는 단계, 여기서 리포터 유전자는 전사인자가 프로모터의 소집 부위 및 표적 부위를 모두 인식하는 경우에는 소정 수준을 초과하여 발현되거나 소정 수준 미만으로 발현되지만 전사인자가 프로모터의 소집 부위만을 인식하는 경우에는 그러하지 아니함;(b) (i) 전사 활성화 또는 억제 도메인, (ii) 소집 부위를 인식하는 DNA 결합 도메인, 및 (iii) 시험 징크 핑거 도메인을 포함하는 비천연적 단백질을 코딩하는 하이브리드 핵산을 함유하는 세포를 제공하는 단계;(c) 다수의 리포터 구조물 중 하나 이상이 세포 내로 들어갈 수 있는 조건하에서 다수의 리포터 구조물을 세포와 접촉시키는 단계;(d) 세포 내에서 하이브리드 핵산이 발현될 수 있게 하는 조건에서 상기 세포를 유지하는 단계; 및(e) 세포가 상기 (a)의 리포터 유전자를 포함하고 세포 내 리포터 구조물이 시험 징크핑거 도메인에 의해 인식되는 표적 부위를 포함함을 나타내는 지표로서 리포터 유전자를 소정 수준을 초과하여 발현하거나 소정 수준 미만으로 발현하는 세포를 동정하는 단계를 포함하는, 시험 징크핑거 도메인이 프로모터 상의 표적 부위를 인식하는지 여부를 결정하는 방법.
- 제26항에 있어서, 상기 표적 결합 부위가 2 내지 6개의 뉴클레오티드 길이인 방법.
- 제27항에 있어서, 다수의 리포터 구조물이 그 표적 결합 부위의 둘 이상의 위치에 A, T, G, C의 모든 가능한 조합을 포함하는 방법.
