Claims (7)
- 내포체의 형태로 생성된 단백질을 포함하는 숙주세포를 파쇄하고, 파쇄된 숙주세포에 변성제를 첨가하여 응집된 단백질을 푸는(unfolding) 공정;칼럼의 상부 방향으로 액체가 유입될 경우, 세포찌거기는 칼럼 상부로 이동하고, 고체매트릭스는 칼럼 내에서 평형상태로 부유될 수 있도록, 세포찌꺼기 보다 높은 밀도를 가지는 고체 매트릭스가액체 중에 평형상태로 부유되어 있는 칼럼의 상부 방향으로, 상기 변성제 및 풀려진 단백질을 포함하는 세포파쇄액을 유입시켜, 상기 고체매트릭스에 상기 풀려진 단백질을 흡착시키는 공정;변성제를 포함한 완충용액을 상기 칼럼의 상부 방향으로 유입시켜, 흡착된 단백질을 제외한 세포파쇄액을 상기 칼럼으로부터 제거하는 공정;변성제를 포함하지 않은 완충 용액을 상기 칼럼의 상부 방향으로 공급하여 변성제의 농도를 점차적으로 줄임으로서 풀려진 단백질을 재접힘하는 공정; 및상기 칼럼과 흡착성이 강한 용출 완충 용액을 상기 칼럼의 상부로부터 하부로 유입시킴으로서, 고체매트릭스를 하부 방향으로 침전시킴과 동시에, 상기 고체매트릭스에 흡착된 재접힘된 단백질을 이탈(desorption)시키고, 이를 칼럼 하부로부터수득하는 공정을 포함하는 단백질의 제조방법.
- 삭제
- 제1항에 있어서, 상기 고체매트릭스는 디에틸아미노에틸(DEAE) 세파로우스레진, DEAE-셀룰로오즈 레진, 카르복시메틸 셀룰로오스 레진, 아가로스 레진으로 이루어진 군으로부터 선택되는 것인 단백질의 제조방법.
- 제1항에 있어서, 상기 단백질은 인간성장호르몬(rhGH)과 글루타티온 트랜퍼라제(GST)의 융합단백질(hGH-GST), 인터페론, 프로키모신, 오브알부민, 및 말싸이트크롬C로 이루어진 군으로부터 선택되는 것인 단백질의 제조방법.
- 제1항에 있어서, 상기 변성제는 6 내지 10M 농도의 우레아 수용액인 것인 단백질의 제조방법.
- 제1항에 있어서, 상기 변성제를 함유한 완충용액의 사용량은 칼럼 내 고체매트릭스의 1 내지 5부피이고, 상기 변성제를 함유하지 않은 완충용액의 사용량은 칼럼 내 고체매트릭스의의 6 내지 15부피인 것인 단백질의 제조방법.
- 제1항에 있어서, 상기 숙주세포를 파쇄한 후, 세포파쇄액을 세정하는 공정을 더욱 포함하는 단백질의 제조방법.
