Abstract

본 발명은 염산 용액과 접촉시 겔화되지 않고, 즉시 붕해됨으로써 생체이용률이 높은 경구 투여용 염산 서트랄린 조성물에 관한 것으로, 활성 성분으로서 염산 서트랄린 및 상기 염산 서트랄린 100 중량부에 대해 100 중량부 이상의 붕해제를 포함하는 본 발명의 경구투여용 염산 서트랄린 조성물은 염산 용액과 접촉시 겔화되지 않고 1분 이내에 즉시 붕해되어 높은 용출을 나타냄으로써 생체 이용률을 높일 수 있다.

Description

염산 서트랄린 경구투여용 조성물{COMPOSITION FOR ORAL ADMINISTRATION OF SERTRALINE HYDROCHLORIDE}
본 발명은 염산 용액과 접촉시 겔화되지 않고 즉시 붕해됨으로써 생체 이용률을 높일 수 있는 서트랄린(sertraline)의 경구 투여용 조성물에 관한 것이다.
서트랄린은 선택적인 세로토닌 재흡수 억제제(selective serotonin reuptake inhibitor, SSRI)로서 항우울제로 광범위하게 사용되고 있는 약물이며, 특히 용해도 증가를 위해 주로 염산염의 형태로 사용되고 있다. 이는 보통 1일 50 내지 200 ㎎의 용량으로 투여되는 약물로서 반감기가 약 23 시간으로 길어 보통 1일 1회 복용되고 있다. 서트랄린은 우수한 약효에도 불구하고 원료 약물 자체의 특성으로 인하여 염소이온을 함유하는 수용성 매질과 접촉할 경우 겔을 형성하여 용해속도가 저하되고 이로 인해 위장관에서의 흡수가 저하되는 문제점이 있다. 실제로 물에서 서트랄린의 용해도는 3.8 ㎎/㎖인데 반해 0.1 N-염산용액에서는 겔화에 의해 0.5 ㎎/㎖로 저하되는 현상이 나타난다(Bruce M. Johnson and Pei-Tei L. Chang,Analytical profiles of drug substance and excipients, Academic Press. Inc.Vol 24, 443-486(1996)).
화이자(Pfizer)사의 국제공개 제 WO99-01120 호에는, 염소 이온에 의한 겔화로 인한 서트랄린의 용해도 저하를 개선하기 위해 유기산이나 계면활성제와 같은 용해보조제를 사용하여 용해도를 증가시킨 방법이 개시되어 있다. 그러나 이 방법은 많은 양의 용해보조제인 유기산이 사용되어야 하는 문제점과 고형제제 제조시 용해보조제와 약물의 불균질한 혼합 등으로 용출의 저하가 초래되는 문제점이 있다.
이에 본 발명자들은 용해보조제를 사용하지 않으면서 염소이온에 의한 서트랄린의 겔화를 방지할 수 있는 조성물을 제조하기 위해 계속 연구를 진행한 결과, 일정량의 붕해제를 조성물에 사용함으로써 염소이온을 함유하는 염산 용액 중에서 약 1 분내에 서트랄린을 붕해시켜 겔화를 방지할 수 있음을 발견하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 염산용액과 접촉시 겔화되지 않고 즉시 붕해되어 생체 이용률이 높은 경구투여용 염산 서트랄린 조성물을 제공하는 것이다.
상기 목적에 따라, 본 발명에서는 활성 성분으로서 염산 서트랄린 및 상기 염산 서트랄린 100 중량부에 대해 100 중량부 이상의 붕해제를 포함하는, 경구투여용 염산 서트랄린 조성물을 제공한다.
본 발명에 따른 조성물의 구성 성분을 상세히 설명하면 다음과 같다.
(1) 활성성분
본 발명의 조성물에는 활성성분으로서 염산 서트랄린이 사용된다.
(2) 붕해제
본 발명의 조성물에서 사용되는 붕해제 성분은 염소이온에 의한 겔화를 방지하는데 필수적인 성분이다. 이러한 붕해제를 포함하는 조성물은 0.1 N 염산 용액 중에서 1분 미만, 바람직하게는 약 30 초 미만의 붕해시간을 나타낼 때 염소이온에 의한 겔화가 방지될 수 있다. 이러한 효과를 나타내기 위해서, 붕해제는 염산 서트랄린 100 중량부에 대해 약 100 중량부 이상, 바람직하게는 약 130 중량부 내지 1000 중량부의 양으로 사용한다. 본 발명에 가장 적합한 붕해제의 대표적인 예는 다음과 같다:
1) 미세결정 셀룰로오스
2) 가교화된 소디움 카르복시메틸 셀룰로오스
3) 가교화된 폴리비닐 피롤리돈
4) 이온교환수지, 특히 엠버라이트(amberlite) IRP-88
또한, 기타 약제학적으로 허용가능한 붕해제, 예를 들어 소디움 스타치 글리콜레이트, 칼슘 카르복시메틸 셀룰로오스 등을 추가로 첨가할 수 있다.
또한 본 발명의 조성물은 본 발명의 효과를 해치지 않는 범위내에서 활택제,부형제 등 제제화를 돕기 위한 약제학적 첨가제를 추가로 포함할 수 있다. 본 발명의 조성물을 이용하여 통상의 방법에 따라 다양한 경구용 제제를 제조할 수 있는데, 예를 들면, 염산 서트랄린, 붕해제 및 기타 약제학적 첨가제를 혼합하여 직타로 타정하여 정제로 제조하거나, 통상의 방법에 따라 분말제, 과립제 등으로 제형화하거나, 분말 또는 과립의 형태로 경질캅셀에 충진하여 캅셀제를 제조할 수도 있다.
본 발명의 염산 서트랄린 조성물은 일반적으로 알려진 1일 투여량인 50 내지 200 ㎎의 용량으로 투여되도록 1회 내지 수회에 나누어 적절히 투여할 수 있다.
이하 본 발명을 하기 실시예에 의하여 더욱 상세하게 설명하고자 한다. 단, 하기 실시예는 본 발명을 예시하기 위한 것일 뿐, 본 발명의 범위가 이들만으로 한정되는 것은 아니다.
실시예 1
하기와 같은 조성으로 이루어진 성분들을 혼합한 후 타정하여 염산 서트랄린 경구투여용 정제를 제조하였다:

실험예 : 용출시험
실시예 1 내지 3 및 비교예 1에서 얻은 정제에 대해 대한약전 시험법에 따라 용출시험(패들법) 및 붕해시험을 실시하여 그 결과를 하기 표 1에 나타내었다. 이 때 용출시험조건 및 액체 크로마토그래피 조건은 다음과 같다.
용출시험장치 : 에르웨카(Erweka) DT 80
용출액 : 0.1 N 염산 900 ㎖
용출액의 온도 : 37±0.5℃
회전속도 : 100 rpm
분석법 : 액체 크로마토그래피법
칼럼-코스모실(Cosmosil) C18(150 mm x 4.6 mm)
이동상-0.25 M 인산이수소칼륨: 아세토니트릴(65:35)
주입량-20 ㎕
유속-1.0 ㎖/분

상기 표 1에서 보듯이, 본 발명에 따른 제제는 붕해시간이 1분 미만으로 매우 짧으므로 염소이온에 의한 서트랄린의 겔화를 방지할 수 있으며, 이로 인해 용출이 증가하고 생체 이용율이 개선됨을 확인할 수 있었다. 이에 반해 비교예 1의 제제는 붕해시간이 1분 이상 소요되며 붕해 후에도 작은 덩어리가 겔화되어 있음을 확인하였다.
본 발명의 속붕해제를 함유하는 경구투여용 조성물은 염산 용액과 접촉시 겔화되지 않고 1분 이내에 즉시 붕해되어 높은 용출을 나타냄으로써 생체 이용률을 높일 수 있다.

Claims (3)

  1. 염산 서트랄린을 유효성분으로 하는 약학 조성물에 있어서, 상기 염산 서트랄린 100 중량부에 대해 100 내지 1000 중량부의 붕해제를 포함하여 염소이온에 의한 서트랄린의 겔화를 방지한 경구투여용 염산 서트랄린 조성물.
  2. 제 1 항에 있어서,
    상기 붕해제가 미세결정 셀룰로오스, 가교화된 소디움 카르복시메틸 셀룰로오스, 가교화된 폴리비닐 피롤리돈, 이온교환수지 또는 이들의 혼합물인 것을 특징으로 하는 조성물.
