Claims (16)
- E.coli lac억제인자, λ억제인자,E.coli trp억제인자,E.coli galR억제인자,E.coli araC억제인자,E.coli tet억제인자 및E.coli ArgGNV억제인자로 이루어진 군으로부터 선택되는 억제인자에 의하여 결합될 수 있는 작동유전자를 포함하는 플라스미드,상기 억제인자를 암호화하는 제1의 염색체 유전자, 및상기 작동유전자와 기능적으로 결합되고 세포성장에 필수적인 제2의 염색체 유전자를 함유하고,여기에서 상기 플라스미드는 상기 필수적인 유전자가 발현될 수 있도록 상기 억제인자를 적정하기에 충분한 수로 상기 세포내에 존재하므로써, 세포성장이 가능하게 되는 형질전환된 박테리아 세포.
- 제 1 항에 있어서, 상기 박테리아 세포는 그램 음성 박테리아 세포인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 2 항에 있어서, 상기 박테리아 세포는E. coli세포인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 2 항에 있어서, 상기 박테리아 세포는 살모넬라 세포인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 1 항에 있어서, 상기 박테리아 세포는 그램 양성 박테리아 세포인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 5 항에 있어서, 상기 박테리아 세포는 바실러스 세포인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 1 항에 있어서, 상기 플라스미드는 박테리아 세포 당 약 10∼50개의 상기 플라스미드 사본을 복제할 수 있는 복제 기점을 포함하는 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 1 항에 있어서, 상기 플라스미드는 박테리아 세포 당 약 100∼200개의 상기 플라스미드 사본을 복제할 수 있는 복제 기점을 포함하는 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 7 항에 있어서, 상기 플라스미드는 pBR322인 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 8 항에 있어서, 상기 플라스미드는 pUC인 것을 특징으로 하는 형질전환된박테리아 세포.
- 제 1 항에 있어서, 상기 플라스미드는 목적하는 유전자의 삽입을 위한 클로닝 부위를 포함하는 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 1 항에 있어서, 상기 플라스미드는 억제인자에 의해 결합할 수 있는 작동유전자, 복제 기점 및 목적하는 유전자의 삽입을 위한 클로닝 부위로 필수적으로 이루어진 것을 특징으로 하는 형질전환된 박테리아 세포.
- 제 12 항에 있어서, 상기 플라스미드는 약 1000bp의 길이를 갖는 것을 특징으로 하는 형질전환된 박테리아 세포.
- 세포가 성장하기에 충분한 조건 및 시간 동안에 제1항, 제2항 내지 제6항, 제7항 내지 제11항, 제12항 내지 제13항 종 어느 한 항에 따른 형질전환된 박테리아 세포를 배양하는 단계를 포함하는 것을 특징으로 하는 형질전환된 박테리아 세포내에서의 플라스미드의 보존방법.
- 세포가 성장하기에 충분한 조건 및 시간동안에 제1항, 제2항 내지 제6항, 제7항 내지 제11항, 제12항 내지 제13항 중 어느 한 항에 따른 형질전환된 박테리아 세포를 배양하는 단계 및 상기 배양된 박테리아 세포로부터 플라스미드 DNA를분리해내는 단계를 포함하는 것을 특징으로 하는 플라스미드 DNA의 생산방법.
- 세포가 성장하기에 충분한 조건 및 시간 동안에 제12항 또는 제13항중 어느한 항에 따른 형질전환된 박테리아 세포를 배양하는 단계 및 상기 배양된 박테리아 세포로부터 재조합 단백질을 분리해내는 단계를 포함하는 것을 특징으로 하는 재조합 단백질의 생산 방법.
