Abstract
본 발명은 피부 외용제 조성물에 함유되어 있는 활성성분의 피부흡수를 증진시킬 수 있는 조성물에 관한 것으로, β-1,3-글루칸 주당쇄에 β-1,6-잔기를 갖는 글루칸으로 안정화한 단백질 분해 효소를 피부흡수증진제로 사용하는 것에 의해 각질층의 장벽기능을 일시적으로 약화시켜 피부투과가 어려운 활성성분의 피부각질층 투과를 증진시킨 외용제 조성물에 관한 것이다.
Description
본 발명은 피부 외용제 조성물에 함유되어 있는 활성성분의 피부 흡수를 증진시킬 수 있는 조성물에 관한 것으로, 더욱 상세하게는 β-1,3-글루칸 주당쇄에 β-1,6-잔기를 갖는 글루칸으로 안정화된 단백질 분해 효소를 피부흡수증진제로 사용하는 것에 의해 활성성분의 피부흡수도를 개선시킨 외용제 조성물에 관한 것이다.
피부는 크게 각질층, 표피층, 진피층의 세 부분으로 구성되어 있는데, 화장품과 같은 피부 외용제 조성물에 함유되어 있는 활성성분의 피부흡수에 있어서 제 1의 장벽은 피부의 최외각에 존재하는 각질층임이 이미 공지되어 있다.
특히, 각질층이 케라틴 단백질을 주성분으로 하는 각질세포와 각질 세포간을 구성하는 지질층으로 이루어져 있으므로, 지용성 활성성분은 원활하게 피부에 흡수되지만, 수용성 및 분자량이 큰 활성성분은 피부를 투과하기 어렵다. 따라서, 대부분이 수용성인 화장품 등의 피부 외용제에 함유되어 있는 활성성분은 피부에의 흡수가 용이하지 않다.
따라서, 종래 이러한 문제를 해결하기 위하여 피부흡수촉진제로 비극성 용매, 계면활성제, 지방산 등을 사용하는 방법이 공지되어 왔으나[Chen LH, Chien YW, "Enhancement of skin penetration" In "Novel cosmetic delivery systems" 1999, 60; Rhein LD, Robbins CR, Fernee K, Cantore R. "Surfactant structure effects on swelling of isolated human stratum corneum, J. Soc Cosmet Chem 1986; 37: 125; Cooper ER, Merritt EW, Smith RL. "Effect of fatty acids and alcohols on the penetration of acyclovir across human skin in vitro J. Pharm Sci 1985; 74:688], 이러한 경피흡수촉진제들은 유효성분의 피부 투과를 어느정도 촉진하기는 하지만, 분자량이 작아서 유효성분과 함께 피부에 흡수되어 심한 자극을 야기시키는 문제점이 있었다. 이밖에도 수용성 물질의 피부 흡수를 위해 초음파 (sonophoresis) 혹은 미세한 전기자극(iontophoresis)을 피부에 부여하는 방법이 공지되어 있으나[Sloan JB, Slotani K. "Iontophoresis in dermatology: a review" J.Am.Acad.Dermatol. 1986, 4:671; Langer R. "Ultrasound-mediated transdermal protein delivery" Science 1995; 269:850], 이러한 방법은 고가의 장비가 필요하다는 이유 등으로 인해 현실적으로 화장료와 같은 피부 외용제제에 적용하기 어렵다.
한편, 상기한 방법 이외에, 효소를 이용하여 피부 장벽기능을 변화시키거나 약화시킴으로써 활성성분의 피부흡수를 증진시키는 방법에 대한 특허들이 공지되어 있다. 예를 들면, 미국특허 제 5,534,260호 및 제 5,296,222호에 단백질 분해 효소를 첨가하여 약물의 피부흡수를 증진시키는 방법이 제시되어 있다. 그러나, 이 방법은 단순히 단백질 분해 효소를 첨가하는 방법에 불과하여 제형에 포함된 성분 물질들의 영향으로 인해 시간이 경과하면서 단백질 분해 효소가 변성되거나 활성을 잃어버리게 된다. 그 결과, 효소에 의한 경피흡수 증진효과를 보이지 않으므로 활성성분 이외에 여러 가지 성분이 함유되는 피부외용제에 적용하기에는 적합하지 않다.
이에, 본 발명자들은 화장품 등과 같은 피부 외용제에 함유된 활성성분의 피부흡수를 증진시킬 수 있는 방법에 대하여 연구하게 되었고, 그 결과, 베타-1,3-글루칸 주당쇄에 규칙적인 베타-1,6-잔기를 갖는 글루칸에 의해 안정화된 단백질 분해 효소를 피부 외용제 조성물에서 피부흡수증진제로 사용한다면, 활성성분의 피부흡수정도가 증진된다는 것을 발견하고 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 조성물 내에 함유된 활성성분의 피부흡수를 증진시킬 수 있는 피부 외용제 조성물을 제공하는 것이다.
상기한 목적을 달성하기 위하여, 본 발명에 따른 조성물은 베타-1,3-글루칸 주당쇄에 규칙적인 베타-1,6-잔기를 갖는 글루칸으로 안정화된 단백질 분해 효소를 피부흡수증진제로 함유함을 특징으로 한다.
이하, 본 발명을 보다 상세히 설명한다.
본 발명은 단백질 분해 효소를 베타-1,3-글루칸 주당쇄에 규칙적인 베타-1,6-잔기를 갖는 글루칸을 이용하여 안정화시킨 후, 이를 활성성분을 함유하는 피부 외용제 조성물에서 피부흡수증진제로 사용하여 활성성분의 피부 흡수를 증진시킨 것으로, 피부자극을 유발시키지 않고, 각질층의 구조를 변형시키거나 각질층을 제거하는 것에 의해 활성성분의 피부 흡수를 증진시킬 수 있다.
본 발명의 조성물에서 피부흡수증진제로 사용된 안정화 단백질 분해 효소는 본 출원인이 출원한 "안정화된 효소 또는 단백질을 제조하는 방법 및 이에 의해 제공되는 안정화 효소 또는 단백질을 함유하는 외용제 조성물"(대한민국 특허출원 제 99-9380호(1999. 3.19))에 기재된 방법을 따른다. 즉, (1) 글루칸을 과요오드산염류와 반응시켜 글루칸중의 베타-1,6-잔기를 알데히드로 만드는 단계; (2) 상기 (1)단계 반응액의 잔류 과요오드산염류를 제거하는 단계; (3) 상기 (2)단계의 용액에 단백질 분해 효소를 0.00001∼10.0중량%의 양으로 첨가하는 단계; (4) 상기 (3)단계의 용액에 0.0001∼1.0중량%의 환원제를 첨가하는 단계; 및 (5) 상기 (4)단계의 용액을 세척하는 단계로 단백질 분해 효소를 안정화시킨다.
상기한 방법에 사용되는 글루칸은 β-1,3-글루칸 주당쇄에 β-1,6-잔기를 갖는 글루칸으로, 화학반응 방법에 따라 β-1,6-잔기에만 반응기를 만들 수 있는 글루칸이다. 이 글루칸의 종류는 특별히 한정되지 않지만, 예를 들면, 치마버섯(Schizopyllum commune)에 의해 생산된 스키조필란(schizophyllan),스클레로티니아속(Sclerotinia sp.)균주에 의해 생산된 스클레로글루칸(scleroglucan), 표고버섯(Lentinus edodes)에 의해 생산된 렌티난(lentinan) 등이다.
한편, 상기한 방법에 의해 안정화되는 단백질 분해 효소는 그 종류가 특별히 한정되지 않지만, 예를 들면, 파파인, 키모트립신, 트립신, 카르복시펩티다제, 펩신 등을 사용할 수 있다.
안정화된 단백질 분해효소는 피부 외용제 조성물에 조성물 총 중량에 대하여 0.0001∼99.9999중량%의 양으로 함유되어 활성성분의 피부흡수를 증진시킨다. 본 발명에서 외용제 조성물은 그 제형에 있어서 특별히 한정되는 바가 없으며, 예를 들면, 유연화장수, 영양화장수, 마사지크림, 영양크림, 메이크업 베이스, 립스틱, 팩, 젤, 삼푸, 린스, 헤어토닉, 비누 등의 제형을 갖는 화장료일 수 있으며, 또한, 로션, 연고, 겔, 크림, 패취, 분무제 등의 경피투여형 제형을 갖는 외용제일 수 있다.
한편, 본 발명의 피부흡수증진제에 의해 피부 흡수도가 증가될 수 있는 활성 성분은 피부 외용제 조성물에서 통상적으로 사용되는 활성성분으로써, 그 종류가 특별히 한정되지 않지만, L-아스코르빈산, L-아스코빌 인산염, L-아스코빌 글루코시드 등과 같은 L-아스코르빈산의 수용성 유도체, 비타민 B군, 코직산, 코질카페익산, 코질아미노프로필인산염, 하이드로퀴논, 알부틴, 물 또는 에탄올과 같은 극성용매를 사용하여 녹차 및 포도종자 등과 같은 천연물로부터 추출한 수용성 천연추출물 등이 있다.
이하, 실시예를 통해 본 발명을 상세히 설명하지만, 본 발명이 이들 예로만 한정되는 것은 아니다.
<참조예 1> 안정화된 파파인의 제조
1) 분자량이 200만인 0.5중량%의 스키조필란 수용액 50㎖에 과요오드산나트륨(NaIO4) 2g을 첨가하고, 4℃의 암소에서 1시간 동안 교반하였다.
2) 반응된 스키조필란 용액을 분자량한계 10,000의 투석막에 넣고, 4℃의 암소에서 48시간 동안 투석하였다. 투석은 2,500㎖의 수중에 12시간마다 물을 교환하면서 실시하였다.
3) 스키조필란 용액의 부피가 약 100㎖ 정도로 증가되었을 때, 0.1중량%의 파파인을 넣고, 이 용액을 2시간 동안 4℃의 암소에서 교반하였다.
4) 스키조필란-파파인 용액에 붕수소화나트륨(NaBH4) 0.05g을 넣고 4시간 동안 교반한 후, 리신 0.1g을 넣고 4℃의 암소에서 2시간동안 교반하였다.
5) 스키조필란-파파인 용액을 상기 2)에서와 같은 방법으로 투석하여 스키조필란-파파인 결합체 용액인 GE-1을 얻었다.
상기 방법에 의해 제조된 GE-1의 수율은 효소활성도기준으로 95%이다.

<제조방법>
1) 정제수에 1~7의 성분을 첨가한 후, 교반하여 완전히 용해시켰다.
2) 에탄올에 10~12의 성분을 첨가한 후, 교반하여 완전히 용해시켰다.
3) 상기 1)의 용액을 교반하면서 상기 2)의 용액을 천천히 첨가하였다.
4) 상기 3)에 8의 성분을 첨가하여 젤을 형성시켰다.
5) 진공 탈기시켜 기포를 제거하여, 수상 제형 화장료를 얻었다.

<제조방법>
1) A(성분 1~7)와 B(성분 8~10)를 각각 75℃까지 가열하였다.
2) B에 A를 조금씩 첨가하며 7000rpm 속도로 유화교반하고, A를 전량 첨가한 후 바로 성분 14와 16을 첨가한 후 2분간 유화 교반하였다.
3) 성분 11과 15를 첨가한 후 2분간 유화 교반하였다.
4) 상기 3)을 1분간 2500rpm으로 교반하여 공기를 탈기하였다.
5) 상기 4)를 얼음물에 담그어 28~30℃까지 냉각하였다.
6) 냉각된 조성물에 성분 12 또는 13을 첨가한 후, 2000rpm으로 교반하여 혼합하였다.
7) 조성물을 상온에서 24시간 동안 방치하여 안정화하여 유중수형 유화 화장료를 얻었다.
[시험예 1] 코직산의 피부흡수 측정
코직산의 피부 흡수는 기네아피그 피부를 대상으로 프란츠 투과 셀을 이용하여 측정하였다. 실험직전 기네아피그의 복부부분 피부를 채취하여 평방 1㎠의 면적으로 절단한 후, 이를 투과경의 직경이 0.9cm인 투과셀에 정치한 후 클램프로 고정하였다. 피부의 한쪽면에 측정하고자 하는 실시예 1 및 비교예 1의 화장료를 0.5㎖ 도포하고, 반대쪽은 정제수와 글리세린이 1:1 중량비로 혼합된 용매와 접촉하도록 하였다. 실험시 온도는 실제 피부온도인 32℃를 유지하였다. 실험 시작후 일정시간 간격으로 용매의 일부를 채취한 후, HPLC를 이용하여 피부에 흡수된 코직산의 양을 측정하여, 그 결과를 표 1에 나타내었다.

일반적으로 화장의 지속시간이 4시간에서 8시간 사이임을 감안할 때, 표 1로부터 코직산의 피부흡수는, 안정화된 단백질 분해 효소가 조성물에 첨가되는 경우가 첨가되지 않는 경우보다 최소 4배 이상 증가함을 알 수 있다.
[시험예 2] 아스코르빈산의 피부흡수 측정
실시예 2 및 비교예 3의 유중수형 유화 화장료를 사용한다는 것을 제외하고, 시험예 1과 동일한 방법으로 피부에 흡수된 아스코르빈산의 양을 측정하여, 그 결과를 표 2에 나타내었다.

1) A(성분 1~9 및 성분 17)와 B(성분 10~12)를 각각 75℃까지 가열하였다.
2) A에 B를 첨가하고 7500rpm의 속도로 5분 동안 유화 교반하였다.
3) 상기 2)에 성분 13을 첨가하고, 5분간 7500rpm의 속도로 유화 교반하였다.
4) 상기 3)을 냉각수를 이용하여 25℃까지 냉각시켰다.
5) 상기 4)에 성분 14~16을 첨가하고 2500rpm으로 교반, 혼합하였다.
6) 24시간동안 상온에 방치하여 안정화시켜 연고를 얻었다.
[시험예 3] 하이드로퀴논의 피부흡수 측정
실시예 3 및 비교예 5의 연고를 사용한다는 것을 제외하고, 시험예 1과 동일한 방법으로 피부에 흡수된 하이드로퀴논의 양을 측정하여, 그 결과를 표 3에 나타내었다.

제조 후 30일이 경과한 실시예 1과 비교예 2의 수상 젤형 화장료를 사용하여 시험예 1과 동일한 방법으로 피부에 흡수된 코직산의 양을 측정하여, 그 결과를 표 4에 나타내었다.

[시험예 5] 제조후 30일이 경과된 유중수형 유화 화장료의 아스코르빈산 피부흡수 측정
제조 후 30일이 경과한 실시예 2과 비교예 4의 유중수형 유화 화장료를 사용하여 시험예 2와 동일한 방법으로 피부에 흡수된 아스코르빈산의 양을 측정하여, 그 결과를 표 5에 나타내었다.

제조 후 30일이 경과한 실시예 3과 비교예 6의 연고를 사용하여 시험예 3과 동일한 방법으로 피부에 흡수된 하이드로퀴논의 양을 측정하여, 그 결과를 표 6에 나타내었다.

표 6으로부터, 파파인을 모두 함유하는 연고일지라도, 안정화되지 않은 파파인을 함유하는 비교예 6의 경우, 제조한지 30일이 경과한 후에는 파파인이 연고화내의 다른 성분들의 영향으로 인해 효소의 활성이 거의 없어져 파파인에 의한 경피흡수 증진효과를 거의 보이지 않음을 알 수 있다.
[시험예 7] 피부손상관찰
상기 시험예 1에서와 동일한 방법으로 실시예 1~3의 외용제를 도포한 후, 도포 24시간이 지난 기네아 피그의 피부를 프란쯔 투과셀에서 꺼내 육안으로 관찰하였다.
그 결과, 실시예 1~3의 외용제를 도포한 경우 모두 피부 손상은 관찰되지 않았다. 따라서, 안정화된 단백질 분해 효소는 피부에 손상을 주지 않으면서 활성성분을 효과적으로 투과시킴을 알 수 있다.
[시험예 8] 피부 안정성
단백질 분해 효소의 피부 안전성을 조사하기 위해 살아있는 무모생쥐를 대상으로 등부위에 실시예 1~3의 외용제를 하루에 한번씩 30일동안 300㎖씩 동일부위에 도포하면서, 매일 육안으로 피부의 부풀어오름, 홍반 등의 피부자극을 관찰하였다. 그 결과, 어떠한 증상도 관찰되지 않았다.
본 발명의 피부흡수증진제는 피부자극, 피부 손상 등의 문제가 없으며, 각질층과의 친화도가 낮거나 분자량이 커서 피부흡수가 어려운 피부 활성성분의 피부흡수 증진에 적용이 가능하다.
Claims (5)
- 활성성분을 함유하는 피부 외용제 조성물에 있어서, 피부흡수증진제로 β-1,3-글루칸 주당쇄에 β-1,6-잔기를 갖는 글루칸에 의해 안정화된 단백질 분해효소를 함유하는 것에 의해 활성성분의 피부흡수를 증진시킬 수 있는 피부 외용제 조성물.
- 제 1항에 있어서, 상기 활성성분이 L-아스코르빈산 및 그 유도체, 비타민 B군, 코직산, 코질카페익산, 코질아미노프로필인산염, 하이드로퀴논, 알부틴 및 수용성 천연 추출물로 이루어진 군에서 선택된 것임을 특징으로 하는 조성물.
- 제 1항에 있어서, 상기 베타-1,3-글루칸 주당쇄에 규칙적인 베타-1,6-잔기를 갖는 글루칸이 스키조필란(schizophyllan), 스클레로글루칸(scleroglucan) 또는 렌티난(lentinan)임으로 특징으로 하는 조성물.
- 제 1항에 있어서, 단백질 분해효소는 파파인, 트립신, 키모트립신(chymotrypsin), 카복시펩티다제 및 펩신으로 이루어진 군에서 선택된 것임을 특징으로 하는 조성물.
- 제 1항에 있어서, 안정화된 단백질 분해효소는(1) β-1,3-글루칸 주당쇄에 β-1,6-잔기를 갖는 글루칸과 과요오드산염류를 반응시켜 글루칸중의 베타-1,6-잔기를 알데히드로 만드는 단계;(2) 상기 (1)단계 반응액의 잔류 과요오드산염류를 제거하는 단계;(3) 상기 (2)단계의 용액에 단백질 분해 효소를 0.00001∼10.0중량%의 양으로 첨가하는 단계;(4) 상기 (3)단계의 용액에 0.0001∼1.0중량%의 환원제를 첨가하는 단계; 및(5) 상기 (4)단계의 용액을 세척하는 단계;에 의해 안정화된 것임을 특징으로 하는 조성물.
