Claims (4)
- a. 성숙 천연 몰로니종 쥐의 백혈병 바이러스(MMLV) 역전사 효소와 실질적으로 크기가 같은 RNA 의존성 및 DNA 의존성 DNA 폴리머라제 활성을 갖는 재조합 폴리펩티드를 암호화하는 MMLV로부터 유래된 핵산 서열을 포함하는 재조합 벡터를 함유하고 RNase가 결핍된 다수의 대장균 숙주 세포를 제공하는 단계; b. 상기 재조합 폴리펩티드를 함유하는 다수의 숙주 세포를 용균시켜 세포 용균물을 형성하는 단계; c. 상기 세포 용균물을 약 0.05M 이하의 NaCl에 해당하는 전도도를 갖는 용액의 존재하에 양이온 교환 배지에 첨가하여, 상기 재조합 폴리펩티드를 양이온 교환 배지에 결합시키는 단계; d. 상기 재조합 폴리펩티드를 약 0.2M NaCl에 해당하는 전도도로 시작하여 약 0.7 M NaCl에 해당하는 전도도로 종결되는 염 구배와 접촉시킴으로써 상기 양이온 교환 배지로부터 결합된 폴리펩티드를 용출시킨 다음, 하나 이상의 분획중에서 상기 재조합 폴리펩티드를 회수하는 단계; e. 상기와 같이 회수된 재조합 폴리펩티드를 함유하는 상기 분획을 겔 여과 컬럼에 가하여 상기 재조합 폴리펩티드를 하나 이상의 분획중에서 회수하는 단계를 포함하는, RNA 의존성 및 DNA 의존성 DNA 폴리머라제 활성을 갖는 재조합 폴리펩티드의 정제방법.
- (정정) 제1항에 있어서, 상기 RNAse 결핍 숙주 세포가 대장균 균주 1200으로 이루어진 것인 방법.
